        Relative strength of 5’ splice-site strength defines functions of SRSF2 and SRSF6 in alternative splicing of Bcl-x pre-mRNA

        Bcl-x, a member of the Bcl-2 family, plays a key role in apoptosis. Alternative splicing of Bcl-x pre-mRNA through alternative 5’ splice-site selection produces an anti-apoptotic mRNA isoform that includes exon 2b and a pro-apoptotic Bcl-x mRNA isoform that excludes exon 2b. Here we used Bcl-x minigene and identified SRSF2 and SRSF6 as two regulatory factors of 5’ splice-site selection of Bcl-x pre-mRNA. We selected binding clusters closer to 5’ splice-sites from multiple potential binding sites of SRSF2 and SRSF6 to perform loss of functions analysis through site-directed mutagenesis. Our results demonstrated that these mutations did not abolish regulatory functions of SRSF2 or SRSF6, indicating that a single binding motif or a cluster was not a functional target of these proteins in Bcl-x pre-mRNA splicing. Random deletion mutagenesis did not disrupt the role of SRSF2 and SRSF6. Importantly, mutagenesis of 5’ splice-site to a conserved or a weaker score demonstrated that the weaker strength of the target 5’ splice-site or higher strength of the other 5’ splice-site strength limited the role of SRSF2 and SRSF6 in 5’ splice-site activation.         수학활동자료 보유현황 및 활용정도에 관한 교사 인식

        본 연구는 수학활동자료 보유현황 및 활용정도에 대한 교사 인식을 알아보고자 하는데 그 목적이 있다. 본 연구는 부산시 소재 공ㆍ사립 유치원교사 386명을 대상으로 질문지를 사용하였으며, 수집된 자료는 x2(chi-square)검증으로 분석하였다. 본 연구의 결과는 다음과 같다. 첫째, 교사의 경력에 따라 분류하기와 순서짓기, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기, 컴퓨터에서 보유현황 및 활용정도에 유의미한 차이가 있었다. 둘째, 교사의 학력에 따라 분류하기와 순서짓기는 보유현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기는 보유현황 및 활용정도에, 기초적인 통계와 관련된 경험하기는 활용정도에, 컴퓨터는 보유현황 및 활용정도에서 유의미한 차이가 있었다. 셋째, 교사의 담당유아연령에 따라 분류하기와 순서짓지는 보뉴현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기느 보유현황 및 활용정도에, 시간에 대한 기초개념 알기는 활용정도에, 공간과 도형의 기초개념 알기, 기초적인 통계와 관련된 경험하기, 컴퓨터는 보뉴현황 및 활용정도에서 유의미한 차이가 있었다. 넷째, 기관의 설립유형에 따라 분류하기와 순서짓기는 보유현황에, 수의 기초개념 이해하기, 기초적인 측정과 관련된 경험하기, 공간과 도형의 기초개념 알기, 컴퓨터는 보유현황 및 활용정도에서 유의미한 차이가 있었다. The purpose of this study is to examine teachers" perceptions of possession and use of mathematical activity materials. For this purpose, the researchers utilized a questionnaire survey of 386 public and private kindergarten teachers located in Busan. Data obtained from the survey were statistically processed and analyzed through x2(chi-square) test. Results of the study are summarized below. First, there were differences in the possession and use of mathematical activity materials such as "classification and ordering", "understanding the basic concept of number", "experience related to basic measurement", "knowing basic concepts of space and figure" and "computers" based on the amount of teaching experience of those surveyed. Second, there were differences in the possession of such mathematical activity material as "classification and ordering, the possession and use of "understanding the basic concept of number", "experience related to basic measurement" and "knowing basic concepts of space and figure", use of "experience related to basic statistics" and in the possession and use of "computers" based on the educational background of the teachers surveyed. Third, there were differences in the possession of such mathematical materials as "classification and ordering", the possession and use of "understanding the basic concept of number" and "experience related to basic measurement", use of "knowing the basic concept of time", and in the possession and use of "knowing basic concepts of space and figure", "experience related to basic statistics" and "computers" based on the age of the students the surveyed teachers were in charge of educating. Last, there were differences in the possession of such materials as "classification and ordering" and the possession and use of "understanding the basic concept of number", "experience related to basic measurement", "knowing basic concepts of space and figure" and "computers" based on the type of pre-school educational institution.

        Effects of Yoga on Pain, Function, and Depression in Individuals with Nonspecific-Low Back Pain

        Objective: Nonspecific low back pain (NSLBP) is experienced worldwide by many age groups. Yoga is recommended as an exercise to reduce back pain and stress because it is a breathing exercise, posture, and meditation as key elements. The aim of this study is to compare the effects of yoga and stabilization exercise on pain intensity, function, and depression. Design: An open-label, parallel arm, randomized controlled trialMethods: Twenty-four participants were allocated to the experimental and the control group in a ratio of 1:1. Yoga (experimental group) and stabilization exercise (control group) were received twice a week for 6 weeks Participants were assessed at baseline and post-intervention for pain intensity (numeric pain rating scale), function (Aberdeen low back pain scale, flexibility,and strength), and depression (Beck depression inventory). Results: When the experimental group (Yoga) and control group (stabilization exercise) were performed twice a week for 6 weeks, numeric pain rating scale, Aberdeen low back pain scale, and flexibility in post-intervention showed significant improvement in both groups (P<0.05), However, in all variables, the experimental group showed a positive benefit compared to the control group (P<0.06). Conclusions: The results of this study show that yoga has more positive benefits compared to stabilization exercise in pain intensity, function, and depression in individuals with NSLBP.

        RRM but not the Asp/Glu domain of hnRNP C<sub>1</sub>/C<sub>2</sub> is required for splicing regulation of Ron exon 11 pre-mRNA

        The Ron proto-oncogene is a human receptor for macrophage-stimulating protein (MSP). The exclusion of exon 11 in alternative splicing generates △RON protein that is constitutively activated. Heterogenous ribonucleaoprotein (hnRNP) C<sub>1</sub>/C<sub>2</sub> is one of the most abundant proteins in cells. In this manuscript, we showed that both hnRNP C<sub>1</sub> and C<sub>2</sub> promoted exon 11 inclusion of Ron pre-mRNA and that hnRNP C<sub>1</sub> and hnRNP C<sub>2</sub> functioned independently but not cooperatively. Moreover, hnRNP C<sub>1</sub> stimulated exon 11 splicing through intron 10 activation but not through intron 11 splicing. Furthermore, we showed that, whereas the RRM domain was required for hnRNP C<sub>1</sub> function, the Asp/Glu domain was not. In conclusion, hnRNP C<sub>1</sub>/C<sub>2</sub> promoted exon 11 splicing independently by stimulating intron 10 splicing through RRM but not through the Asp/Glu domain.         Binding of SRSF4 to a novel enhancer modulates splicing of exon 6 of Fas pre-mRNA

        <P><B>Abstract</B></P> <P>Alternative splicing of exon 6 in Fas pre-mRNA generates a membrane bound pro-apoptotic isoform or soluble anti-apoptotic isoform. SRSF4 is a member of Arginine-Serine rich (SR) protein family. Here we demonstrate that increased SRSF4 expression stimulates exon 6 inclusion, and that reduced SRSF4 expression promotes exon 6 exclusion. We also show that weaker but not stronger 5’ splice-site strength of exon 6 abolishes the SRSF4 effects on exon 6 splicing. Furthermore, we identified a novel enhancer on exon 6, on which SRSF4 interacts functionally and physically.         SRSF2 directly inhibits intron splicing to suppresses cassette exon inclusion

        SRSF2, a Serine-Arginine rich (SR) protein, is a splicing activator that mediates exon inclusion and exclusion events equally well. Here we show SRSF2 directly suppresses intron splicing to suppress cassette exon inclusion in SMN pre-mRNA. Through a serial mutagenesis, we demonstrate that a 10 nt RNA sequence surrounding the branch-point (BP), is important for SRSF2-mediated inhibition of cassette exon inclusion through directly interacting with SRSF2. We conclude that SRSF2 inhibits intron splicing to promote exon exclusion. [BMB Reports 2017; 50(8): 423-428]

        SR proteins regulate V6 exon splicing of CD44 pre-mRNA

        CD44 pre-mRNA includes 20 exons, of which exons 1-5 (C<sub>1</sub>-C<sub>5</sub>) and exons 16-20 (C<sub>6</sub>-C<sub>10</sub>) are constant exons, whereas exons 6-15 (V<sub>1</sub>-V<sub>10</sub>) are variant exons. V6-exon-containing isoforms have been known to be implicated in tumor cell invasion and metastasis. In the present study, we performed a SR protein screen for CD44 V<sub>6</sub> splicing using overexpression and lentivirus-mediated shRNA treatment. Using a CD44 V<sub>6</sub> minigene, we demonstrate that increased SRSF3 and SRSF4 expression do not affect V<sub>6</sub> splicing, but increased expression of SRSF1, SRSF6 and SRSF9 significantly inhibit V<sub>6</sub> splicing. In addition, using a constitutive exon-specific primer set, we could not detect alterations of CD44 splicing after SR protein-targeting shRNA treatment. However, using a V<sub>6</sub> specific primer, we identified that reduced SRSF2 expression significantly reduced the V6 isoform, but increased V<sub>6-10</sub> and V<sub>6,8-10</sub> isoforms. Our results indicate that SR proteins are important regulatory proteins for CD44 V<sub>6</sub> splicing. [BMB Reports 2016; 49(11): 612-616]
