      • MiR-886-5p Inhibition Inhibits Growth and Induces Apoptosis of MCF7 Cells

        Background and Aims: To explore the molecular mechanisms of miR-886-5p in breast cancer., we examined roles in inhibiting growth and migration of MCF-7 cells. Methods: MiR-886-5p mimics and inhibitors were used to express or inhibit MiR-886-5p, respectively, and MTT and clone formation assays were used to determine the survival and proliferation. Hoechst 33342/ PI double staining was applied to detect apoptosis. The expression of caspase-3, caspase-8, caspase-9, MT1-MMP, VEGF-C and VEGF-D was detected by Western blotting, and the levels of MMP2 and MMP9 secreted from MCF-7 cells were assessed by ELISA. MCF-7 cell migration was determined by wound healing and Transwell assays. Results: We found that the growth of MCF-7 cells was inhibited upon decreasing miR-886-5p levels. Inhibiting miR-866-5p also significantly induced apoptosis and decreased the migratory capacity of these cells. The expression of VEGF-C, VEGF-D, MT1-MMP, MMP2, and MMP9 was also found to be decreased as compared to controls. Conclusions: Our data show that downregulation of miR-886-5p expression in MCF-7 cells could significantly inhibit cell growth and migration. This might imply that inhibiting miR-886-5p could be a therapeutic strategy in breast cancer.

        Experimental and Numerical Investigation of Axial Tensile Strength of CHS X-Joints Reinforced with External Stiffening Rings

        To study the reinforcement eff ect of the external stiff ening ring reinforced and unreinforced X-joints having a circular hollow section subjected to brace tensile load, an experimental and computer-simulated research has been done. Six full-scale X-joints are examined in this study: three reinforced specimens and three corresponding unreinforced ones. The test specimens cover diff erent brace to chord diameters ratios ( β ) of 0.25, 0.51, and 0.73. The experimental facilities and detailed parameters are presented. The experimental results including failure modes, load–displacement curves, and ultimate capacity are analyzed and compared. Two failure modes, weld failure and chord plastifi cation, are observed in the tests. The test results reveal an ultimate load enhancement in the reinforced joints by approximately 50% in comparison with the unreinforced joints and an initial stiff ness increase of 248.1% for the β of 0.25 and approximately 49–76% for the β of 0.51 and 0.73. A fi nite element model using shell elements is developed, which accurately predicts the static performance of the X-joints subjected to brace tensile load with and without external stiff ening rings.

      • Parthenolide-Induced Apoptosis, Autophagy and Suppression of Proliferation in HepG2 Cells

        Purpose: To investigate the anticancer effects and underlying mechanisms of parthenolide on HepG2 human hepatocellular carcinoma cells. Materials and Methods: Cell viability was assessed by MTT assay and cell apoptosis through DAPI, TUNEL staining and Western blotting. Monodansylcadaverin(MDC) and AO staining were used to detect cell autophagy. Cell proliferation was assessed by Ki67 immunofluorescence staining. Results: Parthenolide induced growth inhibition in HepG2 cells. DAPI and TUNEL staining showed that parthenolide could increase the number of apoptotic nuclei, while reducing the expression of the anti-apoptotic protein Bcl-2 and elevating the expression of related proteins, like p53, Bax, cleaved caspase9 and cleaved caspase3. Parthenolide could induce autophagy in HepG2 cells and inhibited the expression of proliferation-related gene, Ki-67. Conclusions: Parthenolide can exert anti-cancer effects by inducing cell apoptosis, activating autophagy and inhibiting cell proliferation.

      • Robust Object Tracking with Mutualism between Particle Filter and Geometric Active Contour

        Robust object tracking is important in many real applications. This study proposes a novel robust object tracking algorithm that combines particle filter and geometric active contour. In contrast to conventional combinations, the proposed algorithm establishes mutualism, wherein the particle filter provides curve initialization for the active contour by using the alpha shape algorithm, and the active contour provides evolved boundary for particle filter classification. The refined particles are used to estimate the states which reduce the distance error. Discriminable object feature is proposed to enhance the tracking robustness, and prior information-based curve evolution is used to improve the accuracy of boundary segmentation. Comparable experiments are performed using a stand particle filter and continuously adaptive mean-shift methods. The proposed algorithm is found to be highly robust for long video sequences with non-linear motion, scale changes and clutter background.

        The morphologies and structures of Ni(OH)2–graphene hybrid materials were tailored by using different mineralizers in this work. It was revealed that the synergic effects of the highly oxidized graphene sheets and the mineralizers played a crucial role in controlling the morphology and structure of the nanocomposites, and Na2CO3 is a very effective mineralizer for growing chiseled 2D nanoplates of Ni(OH)2 on graphene sheets. When produced with NaOH, fragmental Ni(OH)2 crystals with irregular shapes erratically decorated on graphene sheets. In contrast, chiseled Ni(OH)2 hexagonal nanoplates grown on graphene sheets were obtained when Na2CO3 was used as the mineralizer. These unique 2D–2D nanoarchitectures with higher contact area between the nanocrystals and graphene substrate can increase the interfacial interaction and then efficiently improve the structural stability of the composite material, thus exhibiting an enhanced Li storage capacity and excellent cycling performance of 562 mAh g-1 after the 36th cycle.

      • Million-Fold Electrical Conductivity Enhancement in Fe <sub>2</sub> (DEBDC) versus Mn <sub>2</sub> (DEBDC) (E = S, O)

        <P/><P>Reaction of FeCl<SUB>2</SUB> and H<SUB>4</SUB>DSBDC (2,5-disulfhydrylbenzene-1,4-dicarboxylic acid) leads to the formation of Fe<SUB>2</SUB>(D<B>S</B>BDC), an analogue of M<SUB>2</SUB>(D<B>O</B>BDC) (MOF-74, DOBDC<SUP>4–</SUP> = 2,5-dihydroxybenzene-1,4-dicarboxylate). The bulk electrical conductivity values of both Fe<SUB>2</SUB>(D<B>S</B>BDC) and Fe<SUB>2</SUB>(D<B>O</B>BDC) are ∼6 orders of magnitude higher than those of the Mn<SUP>2+</SUP> analogues, Mn<SUB>2</SUB>(DEBDC) (E = O, S). Because the metals are of the same formal oxidation state, the increase in conductivity is attributed to the loosely bound Fe<SUP>2+</SUP> β-spin electron. These results provide important insight for the rational design of conductive metal–organic frameworks, highlighting in particular the advantages of iron for synthesizing such materials.</P>

        이 논문은 전통지식과 유전자원의 국내외적인 보호문제를 연구함을 목적으로 한다. 전통지식의 지재권적 보호에 관하여는 전통지식 근본적 이유의 분석을 바탕으로 그 보호를 위한 방어적 방법론과 적극적 방법론을 살펴보고 현재 국제적인 논의에서의 관련 내용을 검토한다. 결론적으로 국내적 차원에서의 기존의 지재권적 보호방법이 아닌 전통지식에 관한 독자적인 보호 방법이 필요하다는 결론을 도출한다. 또한 유전자원의 보호와 관련하여 현재 시행되고 있는 ‘이익분배정책’을 평가하고 이와 관련된 대표적인 사례와 국제기구에서의 논의 등을 검토한다. 결론적으로 유전자원에 관한 보호 방법론으로는 보다 현실적이고 공평한 이익분배 정책을 제안한다.

        Overexpression of a Phosphatidylinositol-Specific Phospholipase C Gene from Populus simonii ×  P. nigra Improves Salt Tolerance in Transgenic Tobacco

        Phosphatidylinositol-specific phospholipase C (PI-PLC) catalyzes the hydrolysis of phosphatidylinositol-4,5-bisphosphate into diacylglycerol and inositol 1,4,5-trisphosphate. It can play an essential role in plant stress response and signaling. However, the functions of PLCs remain unclear in wooden plants. This study carried out the bioinformatics analysis of the PLC gene family in poplar. The expression pattern analysis suggested the transient up-regulation of PsnPLC in salt stress conditions. The transgenic tobacco plants overexpressing PsnPLC were generated by the Agrobacterium-mediated leaf disc method. The transgenic lines showed a significant increase in plant height, SOD and POD activity, and proline content under salt stress. By contrast, transgenic lines showed a decreased malondialdehyde content under salt stress. Comparative transcriptome analysis indicated that the overexpression of PsnPLC mainly affects membrane-associated GO terms and signaling pathways, and the differential expression of salt-responsive genes may contribute to the enhanced salt tolerance. Overall, the results indicated that PsnPLC plays an essential role in modulating salt tolerance in transgenic tobacco.

        Numerical Analysis on Mechanical Property of Concrete Filled Steel Tube Reinforced Concrete (CFSTRC) Columns Subjected to ISO-834 Standard Fire

        The present study established a numerical investigation on the behavior of concrete filled steel tube reinforced concrete (CFSTRC) columns subjected to fire. A finite element analysis (FEA) model was built to simulate the performance of CFSTRC columns under combined loading and fire. The FEA model was verified by the experimental results of CFSTRC columns subjected to fire. The comparison demonstrated an acceptable accuracy for the proposed FEA model. Afterwards, the FEA model was used to analyze the failure modes, the redistribution of internal force, the changes and development of the stress and strain, and the contact stress between the tube and concrete of CFSTRC columns subjected to fire. Influencing factors that may affect the bearing capacity of the CFSTRC columns subjected to fire were analyzed. The bearing capacity and stiffness of the CFSTRC columns gradually decreased in the fire; the parameters such as the fire duration time, sectional dimension, slenderness ratio, and sectional core area ratio significantly influenced the bearing capacity of the CFSTRC columns. Finally, a simplified calculating formula was proposed to calculate the influence factors of the bearing capacity of the CFSTRC columns subjected to ISO-834 standard fire. The formula-calculated results were well in agreement with the finite element analysis results, which provide a simple and feasible method for evaluating the fire-resistance design of these types of components in practical engineering.
