Chitosan Oligomer as a Hopeful Adjuvant for H5n1 Influenza Vaccine

( Le Van Hiep ),( Mai Thi Thanh ),( Dang Thi Hong Van ),( Vo Thi Phuong Khanh ),( Nguyen Anh Dzung ) 한국키틴키토산학회 2008 한국키틴키토산학회지 Vol.13 No.1

Effect of Chitosan oligomer (dp = 8-16) and other adjuvant such as: Aluminum phosphate (AlPO4) and Freund on immune response of H5N1 influenza vaccine in Swiss mice has been investigated. The Swiss mice were twice vaccinated alternative 20 days with dose 0.1 ml/mouse. After 10, 20 and 30 days, the immune response of Swiss mice with H5N1 vaccine was detected. The result showed that after 20 days of the first vaccination, immune response of chitosan oligomer test and Freund was 100%, but Freund test reduced 80% after 30days of vaccination. Meanwhile, immune response of the control and AlPO4 test was very low, only 40%. The Effect of chitosan oligomer and the others on antibody titer has been also investigated. The result showed that the antibody titer of chitosan oligomer test was 60 HIU (Hemagglutination units), 70 HIUs after 10 and 20 days of the first vaccination, and increased up to 112 HIUs after 10 days of the second vaccination and the antibody titer of the chitosan oligomer test was higher than the others. Therefore, chitosan oligomer is a hopeful adjuvant for H5N1 influenza vaccine.

Correlations between abnormalities of morphological details and DNA fragmentation in human sperm

Nguyen, Hiep Tuyet Thi,Dang, Hong Nhan Thi,Nguyen, Thai Thanh Thi,Nguyen, Trung Van,Dang, Thuan Cong,Nguyen, Quoc Huy Vu,Le, Minh Tam The Korean Society for Reproductive Medicine 2022 Clinical and Experimental Reproductive Medicine Vol.49 No.1

Objective: As the associations of sperm DNA fragmentation with morphology have not been examined in detail, this study aimed to investigate the relationship between abnormalities of morphological details and DNA integrity in human sperm. Methods: In this cross-sectional study, men from infertile couples were enrolled at Hue Center for Reproductive Endocrinology and Infertility, Vietnam. Conventional semen parameters, including morphological details, were analyzed following the World Health Organization 2010 criteria. Sperm DNA fragmentation was evaluated using a sperm chromatin dispersion assay. The relationships and correlations between semen parameters, sperm morphology, and the type of halosperm and the DNA fragmentation index (DFI) were analyzed. Results: Among 130 men in infertile couples, statistically significant differences were not found in the sperm halo type between the normal and abnormal sperm morphology groups. The percentage of round-head spermatozoa was higher in the DFI >15% group (16.98%±12.50%) than in the DFI ≤15% group (13.13% ±8.82%), higher values for amorphous heads were found in the DFI >15% group, and lower values for tapered heads were observed in the DFI ≤15% group; however, these differences were not statistically significant. Small-halo sperm and the DFI were positively correlated with round-head sperm (r=0.243, p=0.005 and r=0.197, p=0.025, respectively). Conclusion: The rate of general sperm morphological abnormalities in semen analysis was not related to sperm DNA integrity. However, round sperm heads were closely associated with sperm DNA fragmentation.

Physiological intracytoplasmic sperm injection does not improve the quality of embryos: A cross-sectional investigation on sibling oocytes

Minh Tam Le,Hiep Tuyet Thi Nguyen,Trung Van Nguyen,Thai Thanh Thi Nguyen,Hong Nhan Thi Dang,Thuan Cong Dang,Quoc Huy Vu Nguyen The Korean Society for Reproductive Medicine 2023 Clinical and Experimental Reproductive Medicine Vol.50 No.2

Objective: This study aimed to compare the efficacy of physiological intracytoplasmic sperm injection (PICSI) and intracytoplasmic sperm injection (ICSI) in terms of the fertilization rate and embryo quality using sibling oocyte cycles. Methods: This prospective, cross-sectional study collected data from 76 couples who underwent their first cycle at the Hue Center for Reproductive Endocrinology and Infertility, Vietnam, between May 2019 and November 2021. The inclusion criteria were cycles with at least eight oocytes and a sperm concentration of 5×106/mL. Sperm parameters, sperm DNA fragmentation (SDF), fertilization, and the quality of cleavage-stage embryos on day 2 and blastocysts on day 5 were examined. Results: From 76 ICSI cycles, 1,196 metaphase II (MII) oocytes were retrieved, half of which were randomly allocated to either the PICSI (n=592) or ICSI (n=604) treatment group. The results showed no significant difference between the two groups in terms of fertilization (72.80% vs. 75.33%, p=0.32), day 2 cleavage rate (95.13% vs. 96.04%, p=0.51), blastulation rate (52.68% vs. 57.89%), and high-quality blastocyst rate (26.10% vs. 31.13%, p=0.13). However, in cases where SDF was low, 59 cycles consisting of 913 MII oocytes produced a considerably higher blastulation rate with PICSI than with ICSI (50.49% vs. 35.65%, p=0.00). There were no significant differences between the pregnancy outcomes of the PICSI and ICSI embryo groups following embryo transfer. Conclusion: Using variable sperm quality provided no benefit for PICSI versus ICSI in terms of embryo outcomes. When SDF is low, PICSI appears to be able to produce more blastocysts.

        Tran, Tuan Hiep,Nguyen, Hanh Thuy,Le, Nam Van,Tran, Thi Thu Phuong,Lee, Jong Seong,Ku, Sae Kwang,Choi, Han-Gon,Yong, Chul Soon,Kim, Jong Oh Elsevier 2017 International journal of pharmaceutics Vol.528 No.1

Abstract

Heterogeneity of cancer cells and drug resistance require multiple therapeutic approaches for comprehensive treatment. In this study, temperature-sensitive liposomes containing anti-cancer agent tanespimycin (17-AAG) and photosensitizer IR 820 were developed for combination of phototherapy and chemotherapy. The temperature-sensitive liposomes composed of DPPC, cholesterol, DSPE-PEG, 17-AAG, and IR 820 (LP-AI) at weight ratio of 35/15/3/2/2 were formulated as a thin film using extrusion and evaluated for particle size, morphology and drug release profile. Furthermore, the anticancer effect of combined therapy was examined in vitro and in vivo in SCC-7 and MCF-7 cell lines. As a result, LP-AI was prepared at particle size of 166.7±1.3nm, PDI of 0.153±0.012, and ζ-potential of −32.6±0.8mV. After NIR irradiation (660 and 808nm laser), LP-AI could generate heat and ROS and enhance drug release from nanoparticles which were useful to kill the cancer cells. These were confirmed by in vitro cytotoxicity as well as in vivo effective ablation of tumors. In conclusion, fast drug release and enhanced treatment efficacy of LP-AI indicate the potential of integrating photo- and chemotherapy for synergistic anti-cancer effects.

Graphical abstract

Temperature-sensitive liposomes containing anti-cancer agent tanespimycin and photosensitizer IR 820 (LP-AI).

[DISPLAY OMISSION]

Pancreaticoduodenectomy with superior mesenteric artery first-approach combined total meso-pancreas excision for periampullary malignancies: A high-volume single-center experience with short-term outcomes

        Thanh Khiem Nguyen,Ham Hoi Nguyen,Tuan Hiep Luong,Kim Khue Dang,Van Duy Le,Duc Dung Tran,Van Minh Do,Hong Quang Pham,Hoan My Pham,Thi Lan Tran,Cuong Thinh Nguyen,Hong Son Trinh,Yosuke Inoue 한국간담췌외과학회 2024 Annals of hepato-biliary-pancreatic surgery Vol.28 No.1

Backgrounds/Aims: Pancreaticoduodenectomy (PD) is the only radical treatment for periampullary malignancies. Superior mesenteric artery (SMA) first approach combined with total meso-pancreas (MP) excision was conducted to improve the oncological results. There has not been any previous research of a technique that combines the SMA first approach and total MP excision with a detailed description of the MP macroscopical shape. Methods: We prospectively assessed 77 patients with periampullary malignancies between October 2020 and March 2022 (18 months). All patients had undergone PD with SMA first approach combined total MP excision. The perioperative indications, clinical data, intra-operative index, R0 resection rate of postoperative pathological specimens (especially mesopancreatic margin), postoperative complications, and follow-up results were evaluated. Results: The median operative time was 289.6 min (178−540 min), the median intraoperative blood loss was 209 mL (30−1,600 mL). Microscopically, there were 19 (24.7%) cases with metastatic MP, and five cases (6.5%) with R1-resection of the MP. The number of lymph nodes (LNs) harvested and metastatic LNs were 27.2 (maximum was 74) and 1.8 (maximum was 16), respectively. Some (46.8%) patients had pancreatic fistula, but mostly in grade A, with 7 patients (9.1%) who required re-operations. Some 18.2% of cases developed postoperative refractory diarrhea. The rate of in-hospital mortality was 1.3%. Conclusions: The PD with SMA first approach combined TMpE for periampullary malignancies was effective in achieving superior oncological statistics (rate of MP R0-resection and number of total resected LNs) with non-inferior short-term outcomes. It is necessary to evaluate survival outcomes with long-term follow-up.

      • Soft computing-based estimation of ultimate axial load of rectangular concrete-filled steel tubes

        Panagiotis G. Asteris,Minas E. Lemonis,Thuy-Anh Nguyen,Hiep Van Le,Binh Thai Pham 국제구조공학회 2021 Steel and Composite Structures, An International J Vol.39 No.4

In this study, we estimate the ultimate load of rectangular concrete-filled steel tubes (CFST) by developing a novel hybrid predictive model (ANN-BCMO) which is a combination of balancing composite motion optimization (BCMO) - a very new optimization technique and artificial neural network (ANN). For this aim, an experimental database consisting of 422 datasets is used for the development and validation of the ANN-BCMO model. Variables in the database are related with the geometrical characteristics of the structural members, and the mechanical properties of the constituent materials (steel and concrete). Validation of the hybrid ANN-BCMO model is carried out by applying standard statistical criteria such as root mean square error (RMSE), coefficient of determination (R2), and mean absolute error (MAE). In addition, the selection of appropriate values for parameters of the hybrid ANN-BCMO is conducted and its robustness is evaluated and compared with the conventional ANN techniques. The results reveal that the new hybrid ANN-BCMO model is a promising tool for prediction of the ultimate load of rectangular CFST, and prove the effective role of BCMO as a powerful algorithm in optimizing and improving the capability of the ANN predictor.

Parenchymal-sparing anatomical hepatectomy based on portal ramification of the right anterior section: A prospective multicenter experience with short-term outcomes

        Truong Giang Nguyen,Thanh Khiem Nguyen,Ham Hoi Nguyen,Hong Son Trinh,Tuan Hiep Luong,Minh Trong Nguyen,Van Duy Le,Hai Dang Do,Kieu Hung Nguyen,Van Minh Do,Quang Huy Tran,Cuong Thinh Nguyen 한국간담췌외과학회 2024 Annals of hepato-biliary-pancreatic surgery Vol.28 No.1

Backgrounds/Aims: Parenchymal-sparing anatomical hepatectomy (Ps–AH) based on portal ramification of the right anterior section (RAS) is a new technique to avoid unnecessarily transecting too much liver parenchyma, especially in cases of major anatomical hepatectomy. Methods: We prospectively assessed 26 patients with primary hepatic malignancies having undergone major Ps–AH based on portal ramification of the RAS from August 2018 to August 2022 (48 months). The perioperative indications, clinical data, intra-operative index, pathological postoperative specimens, postoperative complications, and follow-up results were retrospectively evaluated. Results: Among the 26 patients analyzed, there was just one case that had intrahepatic cholangiocarcinoma The preoperative level of α– Fetoprotein was 25.2 ng/mL. All cases (100%) had Child–Pugh A liver function preoperatively. The ventral/dorsal RAS was preserved in 19 and 7 patients, respectively. The mean surgical margin was 6.2 mm. The mean surgical time was 228.5 minutes, while the mean blood loss was 255 mL. In pathology, 5 cases (19.2%) had microvascular invasion, and in the group of HCC patients, 92% of all cases had moderate or poor tumor differentiation. Six cases (23.1%) of postoperative complications were graded over III according to the Clavien–Dindo system, including in three patients resistant ascites or intra-abdominal abscess that required intervention. Conclusions: Parenchymal-sparing anatomical hepatectomy based on portal ramification of the RAS to achieve R0-resection was safe and effective, with favorable short-term outcomes. This technique can be used widely in clinical practice.
