        Telomerase Activity in Gastric Adenocarcinomas: Frozen Tissues Versus Methacarn-fixed Paraffin-embedded Tissues

        Purpose: Telomerase is an RNA-dependent DNA polymerasethat synthesizes TTAGGG telomeric DNA ontochromosome ends to compensate for sequence loss duringDNA replication. It has been detected in 85က90% ofall primary human cancers, implicating that its apparentreactivation in tumors may play a role in the tumorigenicprocess. The purpose of this study was to evaluatetelomerase activity in stomach cancer, and to determinewhether methacarn-fixed paraffin-embedded tissues canreplace frozen tissue sections for the telomerase (TRAP)assay.Materials and Methods: Frozen and corresponding methacarn-fixed paraffin-embedded tissue samples wereobtained from 51 patients with gastric adenocarcinomaand analyzed for telomerase activity by using a TRAPezeELISA kit.Results: Telomerase activity was detected in 37 (73%)frozen samples, and in 13 (25%) methacarn-fixed paraffinblocks. Telomerase activity was well correlated with depthof invasion (p=.037) and tumor differentiation (p=.022).Conclusion: These data suggest that reactivated telomerasemay play a significant role in the tumorigenesisof gastric cancer and may reflect the malignant potentialof the tumor. It is noteworthy that methacarn- fixed tissuecannot as yet substitute for the frozen tissue in the TRAPassay. (Cancer Res Treat. 2003;35:478-482)

        Expression of Matrix Metalloproteinase-9 Correlates with Poor Prognosis in Human Malignant Fibrous Histiocytoma

        Purpose: 92 kDa matrix metalloproteinase-9 (MMP-9) is believed to play an important role in degrading the matrix and basement membrane, contributing to the invasion and metastasis of malignant solid tumors. However, little is known about its involvement in a malignant fibrous histiocytoma. The aim of this study was to investigate the expression of MMP-9 and to correlate its expression with clinicopathologic parameters in human malignant fibrous histiocytomas. Materials and Methods: Archival tumor tissues from 20 patients with a malignant fibrous histiocytoma were analyzed by immunohistochemistry for the expression of MMP-9. Clinical information was obtained through the computerized retrospective database from the tumor registry. Results: Seventeen of 20 (85%) tumors showed a positive reaction for MMP-9. MMP-9 activity was inversely correlated with patients' survival time (p=.011). There was no significant correlation between the activated MMP-9 expression and all the other clinicopathologic variables. Conclusion: Our data demonstrate that MMP-9 activation is likely to occur in human malignant fibrous histiocytomas. It is also noteworthy that the expression of MMP-9 may aid in predicting patients' poor prognosis.(Cancer Res Treat. 2004;36:384-388)

        Mullerian-Type Gland Inclusions in Pelvic Lymph Nodes Mimicking Metastasis: A Case Report and Review of the Literature

        Benign lymph node inclusions are rare, and can bemistaken for metastasis. We report, herein, a case of a50-year-old woman who underwent a hysterectomy, witha lymphadenectomy, for an endometrial carcinoma. Therewas no lymph node metastasis; however, the left externaland common iliac lymph nodes demonstrated a fewglands, consistent with Müllerian-type inclusions (endosalpingiosis).Awareness of these lesions is important toavoid either unnecessary therapy or any delay in treatment.Furthermore, pelvic and aortic lymphadenectomiesmay be warranted, as neoplastic transformation of preexistingmetaplastic tubal-type epithelium is strongly suggested.This paper presents a case of intranodal endosalpingiosismimicking metastasis.         The Expression of c-myc, bcl-2 and p53 Proteins in Adenocarcinomas of Lung

        Purpose: c-myc, bcl-2 and p53 are known to regulate apoptosis. There has been growing interest in analyzing their contribution to the pathogenesis and prognosis in a variety of human cancers. This study was undertaken to investigate the expression of these proteins in pulmonary adenocarcinomas and to determine their relationship with clinicopathologic parameters and survival. Materials and Methods: Archival tumor tissues from 61 patients with adenocarcinoma of lung were analyzed by immunohistochemistry for the expression of c-myc, bcl-2 and p53 proteins. Clinical information was obtained through the computerized retrospect database from the tumor registry. Results: Of 61 patients, 32 were men and 29 women with the median age 63 years. 4 had stage I disease, 2 had stage II disease and 55 had stage III disease. The expression of c-myc protein was identified in 13% (8/61) tumors, bcl-2 protein was detected in 1.6% tumors (1/61) and p53 was detected in 77% (47/71) tumors. The association of the expression of c-myc, bcl-2 and p53 was not detected. The survival time was longer in patients expressing c-myc protein than in patients without the c-myc protein expression (p=.045). Neither bcl-2 nor p53 showed the correlation to clinicopathologic variables. Conclusion: Our data suggest the involvement of p53 alteration in the pathogenesis of lung adenocarcinoma. The c-myc expression in some tumors indicates that c-myc alone may not contribute critically to the development and/or the progression of these tumors. It, however, correlated to the survival time, suggesting the c-myc expression as a favorable prognostic factor possibly through the apoptosis pathway.(Cancer Res Treat. 2004;36:146-150)

        Telomerase Activity and Expression of hTR and TERT in Human Soft Tissue Sarcomas

        Purpose: Sarcomas have rarely been analyzed fortelomerase, which is an RNA-dependent DNA polymeraseto maintain telomeres and prevent telomere shortening.This study was undertaken to determine telomeraseactivity and the expression of the telomerasesubunits human telomerase RNA (hTR) and telomerasereverse transcriptase (TERT) in soft tissue sarcomas.Materials and Methods: Twenty three sarcomas wereanalyzed for the telomerase activity by a radioactive PCRbasedTRAP assay. All of the samples were furtherinvestigated for the expression of hTR by in situ hybridizationand for TERT and p53 by immunohistochemistry.Results: Telomerase activity was detected in four (17%)samples. Expression of hTR was demonstrated in 11(48%) cases, whereas TERT was expressed in 20 (87%).Of the four telomerase-positive tumors, three were positivefor both hTR and TERT, and one was positive only forTERT. p53 overexpression was observed in nine (39%)tumors. The frequency of p53 expression increased asthe tumor grade advanced (p= .064).Conclusion: These data indicate that the reactivation oftelomerase is an uncommon event in human soft tissuesarcomas. The high frequency of the expression of hTRand TERT in these tumors suggests that telomeraseactivity may be regulated at the transcriptional level andan additional event leading to telomerase activation exist.(Cancer Res Treat. 2001;34:46-51)

        Luxury Images on Instagram : Computational Analysis of Images Posted by Luxury Fashion Brands

        본 연구는 어떠한 요소들이 인스타그램 상에서 '좋은' 럭셔리 패션 이미지를 만드는지, 그리고 그러한 이미지들에 대해 인스타그램 사용자들이 어떻게 반응하는지를 탐구하는 것에 목적을 두고 있다. 이를 위해 본 연구에서는 세계적인 유명 럭셔리 패션 브랜드들이 인스타그램에 게시한 대규모 패션 이미지 를 수집하고, 브랜드 이름 및 브랜드 로고의 사용, 텍스트의 사용, 해시태그의 사용 등 주요 시각 콘텐 츠 요소를 바탕으로 정량적 분석을 실시하여 이미지들에 대한 구체적인 패턴을 파악하고자 하였다. 또 한 본 연구에서는 각 시각 콘텐츠 변인에 따라 이미지에 대한 사용자의 반응 수준이 달라지는지 살펴보 았다. 그 결과 (1) 브랜드 이름의 사용은 “좋아요” 수와 댓글의 수 측면에서 사용자 반응과 긍정적인 관 련이 있는 반면, 사용된 브랜드 이름의 크기는 “좋아요” 수와 부정적인 관련이 있는 것으로 나타났다. (2) 브랜드 로고의 사용은 사용자 반응과 긍정적인 관련이 있는 것으로 나타났다. (3) 텍스트와 해시태 그의 사용은 사용자 반응과 긍정적 관계가 있는 것으로 나타난 반면, 판매촉진을 위한 상업적 메시지는 사용자 반응과 부정적 관련이 있는 것으로 나타났다. The main purpose of this research is to explore what makes “good” luxury fashion images on Instagram and how Instagrammers respond to those images. A quantitative analysis of a large collection of fashion photos posted by notable luxury fashion brands was conducted to identify specific patterns in these images based on four important visual content variables: use of a brand name, brand logo, text, and hashtag. This study also examined how customer engagement levels vary depending on each visual content variable. Several interesting findings emerged from this study: (1) featuring a brand name was positively related to viewers’ engagement level in terms of the number of likes and comments, while the size of the brand name was negatively related to the number of likes; (2) featuring a brand logo was also positively related to viewers’ engagement level; and (3) the use of text and a hashtag was positively related to viewers’ engagement level, while sharing promotional information was likely to prompt a lower level of interaction.

        단위 조정에 따른 초등학생의 분수 개념 이해 분석

        본 연구의 목적은 학생의 단위 조정 능력이 분수 개념 이해와 어떻게 관련되는지 탐구하는 데 있다. 이를 위해 초등학교 4학년 학생을 대상으로 4개월(2019.3.~2019.6.)에 걸쳐 교수 실험을 진행하였고 본 논문에서는 학생의 분수개념 이해와 관련된 스킴과 조작이 교수 실험 동안 어떻게 변화하였는지에 대한 상세한 분석을 제시하였다. 학생의 단위 조정 조작은 분수 개념을 이해하는 능력과 밀접한 연관이 있는 것으로 나타났는데, 수업 초반에 부분 분수 스킴의 학생은 분수를 2수준 단위를 가지고 조작함으로써 분수를 또 다른 종류의 자연수로 인식하였다. 학생은 진분수와 전체 1을 단위 분수의 배수로 동시에 인식하면서 분수를 자연수와 명확히 구분하였다. 역 부분 분수 스킴의 학생은 1보다 큰 자연수를 내재화된 3수준 단위로, 자연수 아닌 가분수를 활동 중에 3수준 단위로 구성하였다. 본 연구의 결과를 바탕으로 결론 및 교수학적 시사점을 제시하였다. The purpose of this study is to explore how units-coordination ability is related to understanding fraction concepts. For this purpose, a teaching experiment was conducted with one fourth grade student, Eunseo for four months(2019.3. ~ 2019.6.). We analyzed in details how Eunseo’s units-coordinating operations related to her understanding of fraction changed during the teaching experiment. At an early stage, Eunseo with a partitive fraction scheme recognized fractions as another kind of natural numbers by manipulating fractions within a two-levels-of-units structure. As she simultaneously recognized proper fraction and a referent whole unit as a multiple of the unit fraction, she became to distinguish fractions from natural numbers in manipulating proper fractions. Eunseo with a reversible partitive fraction scheme constructed a natural number greater than 1, as having an interiorized three-levels-of-units structure and established an improper fraction with three levels of units in activity. Based on the results of this study, conclusions and pedagogical implications were presented.

        EU Directive 2006/40/EC prohibits use of refrigerants with global warming potential greater than 150 in Mobile Air Condition(MAC) starting 2011. This paper presents an experimental study of various alternative refrigerant MAC systems to identify a possible replacement for current R-134a. Of many proposed refrigerants, HFO-1234yf is overall more environmental friendly than R-744 to replace R-134a, based on a number of performance evaluation and risk assessments. Because HFO-1234yf is a mildly flammable, a number of different risk assessments were completed in order to evaluate its risk in vehicle usage. Many evaluations concluded that only oil ignites even at unrealistic high temperature & pure HFO-1234yf failed to ignite without oil.

      • 스테인리스 스틸과 고밀도 폴리에틸렌 쿠폰 표면에서의 E. coli O157:H7 바이오필름 형성

        대장균(Escherichia coli)은 그람 음성 간균으로 온혈동물의 창자에서 많이 볼 수 있다. 대부분의 대장균 변종들은 해롭지 않지만, E. coli O157:H7은 사람에게 심각한 식중독을 일으키며, 식품에 오염된 경우 대규모 리콜의 원인이 된다. 바이오필름은 미생물이 세포외중합체(Extracellular Polymeric Substance, EPS)의 매트릭스에 내장되어 있는 응집체이며, EPS는 점액질로 세포의 DNA, 단백질과 다당류의 중합체 결합물이다. 바이오필름에는 세균, 곰팡이, 효모, 원생동물 등과 같은 다양한 미생물이 존재하며 바이오필름을 형성한 미생물은 식품, 기기 및 조리 도구 등에 부착하여 교차 오염을 통해 식재료 및 조리된 식품에 잔존하게 되어 결과적으로 식중독을 유발할 수 있다. 본 연구는 식품 및 농산물 가공시설의 안전성 유해요소인 바이오필름을 신속하게 검출하기 위한 장치개발의 기초연구로서, 스테인리스 스틸과 고밀도 폴리에틸렌(High-Density Polyethylene, HDPE) 쿠폰 표면에서 E. coli O157:H7의 바이오필름 형성 특성을 조사하기 위해 수행되었다. 스테인리스 스틸과 HDPE 쿠폰을 각각 Tryptic Soy Broth (TSB) 배지에 투입하고 E.coli O157:H7을 접종· 배양하여 바이오필름을 형성시켰다. 배양된 쿠폰은 세척· 건조 후 Phosphate buffered saline (PBS)에 바이오필름을 탈리· 회수· 평판배양 해서 균수를 측정하는 것으로 바이오필름 형성 특성을 분석하였다. 바이오필름 회수방법은 glass beads와 함께 고속 교반하여 회수하는 방법과 고속 교반만으로 회수하는 방법을 사용하였다. Glass beads를 사용하여 바이오필름을 회수한 경우 스테인리스 스틸 쿠폰의 부착 균수는 4.39 log cfu/㎠이고, HDPE 쿠폰의 부착 균수는 3.10 log cfu/㎠로 두 재질의 쿠폰에서 바이오필름 형성능의 확연한 차이를 보였다. 향후 더 다양한 재질의 쿠폰에서 대장균의 바이오필름 형성 평가와 바이오필름 회수 방법에 관한 연구를 진행할 계획이다.
