      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

        Effects of Pilger Rolling and Heat Treatment on Microstructure and Corrosion Resistance of Ag-electroplated 304 Stainless Steel Tubes

        We introduce a cost-effective method that combines electroplating with metal cold working processes to manufacture compositetubes with robust corrosion resistance. First, a 3-step electroplating process was developed to form an adhesive anduniform Ag coating on the outer wall of a 304 stainless steel tube. The process consisted of a Ni-strike step for removing thepassivation layer of the initial 304 tube, Cu deposition for smoothing the surface by adding a buffer layer, and Ag deposition. To reinforce the interfacial adhesion between the electroplated layers and the 304 tube and increase the area of the Ag coating,a pilger rolling or pilger rolling + heat-treatment process was performed after the Ag electroplating process. Scanningelectron microscopy of the composite tubes after each process indicated that the as-deposited Ag coating changed fromhaving a rough to smooth surface after only pilger rolling and with additional heat treatment. Electron backscatter diffractionanalysis of the microstructures and textures of the tubes revealed that dynamic recrystallization occurred extensively duringpilger rolling, resulting in the formation of a relatively defect-free grain structure without heat treatment. Furthermore,electrochemical polarization curves determined that the Ag-coated composite tubes are superior to the uncoated 304 tubein terms of corrosion resistance under Cl−atmosphere, owing to the formation of a AgCl passivation layer during testing. Surface analysis of the composite tubes suggests that the properties of the AgCl passivation layer are related to the grainsize of the Ag coating and the density of plastic-deformation-induced defects.

        Environmental Microbiology / Microbial Diversity : Calcium Carbonate Precipitation by Bacillus and Sporosarcina Strains Isolated from Concrete and Analysis of the Bacterial Community of Concrete(s)

        Microbially induced calcium carbonate precipitation (CCP) is a long-standing but re-emerging environmental engineering process for production of self-healing concrete, bioremediation, and long-term storage of CO2. CCP-capable bacteria, two Bacillus strains (JH3 and JH7) and one Sporosarcina strain (HYO08), were isolated from two samples of concrete and characterized phylogenetically. Calcium carbonate crystals precipitated by the three strains were morphologically distinct according to field emission scanning electron microscopy. Energy dispersive X-ray spectrometry mapping confirmed biomineralization via extracellular calcium carbonate production. The three strains differed in their physiological characteristics: growth at alkali pH and high NaCl concentrations, and urease activity. Sporosarcina sp. HYO08 and Bacillus sp. JH7 were more alkali- and halotolerant, respectively. Analysis of the community from the same concrete samples using barcoded pyrosequencing revealed that the relative abundance of Bacillus and Sporosarcina species was low, which indicated low culturability of other dominant bacteria. This study suggests that calcium carbonate crystals with different properties can be produced by various CCP-capable strains, and other novel isolates await discovery.

      • 인체 대장암 및 자궁경부암에서 PCR-SSCP법을 이용한 Ki-ras 암유전자의 점돌연변이에 관한 연구

        인체 대장암 20례와 자궁경부암 10례의 암조직을 대상으로 c-Ki-ras 유전자 codon 12와 13에서의 점돌연변이를 알아보고자 polymerase chain reaction(PCR)과 single-strand conformation polymorphism(SSCP) 검사법을 시행하였다. 대장암의 경우 20례중 9례(45%)에서, 자궁경부암의 경우 10례중 1례(10%)에서 양성으로 나왔다. 대장암에서는 codon 12에서 GGT가 TGT로의 치환이 4례로서 가장 많았으며, AGT, CGT로의 치환이 각 1례였고 codon 13에서는 GGC가 GAC로의 치환이 2례, TGC로의 치환이 1례인 것으로 나타났다. 자궁경부암의 경우 codon 12에서 GGT가 AGT로의 치환이 1례인 것으로 나타났다. In an attempt to clarify the role of genetic alteration in the genesis of human colorectal and cervical cancers, tissue specimens from 20 patients with colorectal cancer and 10 patients with cervical cancer were examined for the presence of point mutation in K-ras2 exon 1 by single strand conformation polymorphism analysis of PCR product. Exon I of c-Ki-ras2 was amplified by polymerase chain reaction(PCR) and comparison was made between the normal and mutated genes by nondenaturing polyacrylamide gel electrophoresis(PAGE) of PCR product and nucleotide sequence analysis using asymmetric PCR with direct sequencing. Genomic DNA from white blood cells were used as normal control and those from A427 cell line were used as mutated control. 1.PCR product from A427 cell line showed a distinct migration shift pattern compared to the normal control in PAGE and the direct sequencing indicated that nucleotide sequence of codon 12 was mutated from GGT to GAT. In addition, there was a loss of normal allele in A427 cell line. 2.PCR product from 9 cases(45%) out of a 20 colon cancer patients showed migration shifts in PAGE, and all of these 9 patients invariably demonstrated mutation of ras gene, either in codon 12(6 cases:30%) or 13(3 cases:15%). The base substitutions in codon 12 were: from GGT to AGT(1), CGT(1), or TGT(4). Base changes in codon 13 were from GGC to TGC(1) or GAC(2). 3.PCR product from 1 cases(10%) out of a 10 cervical cancer patients showed migration shrifts in PAGE, Invariably demonstrated mutation of ras gene in codon 12. The base substitutions was from GGT to AGT(1) TQE incidence of point mutation of c-Ki-ras in colon cancer was high, however, it was low in cervical cancer, which showed the relation of ruts gene mutation with colon cancer. PCR -SSCP analysis is a simple, rapid and efficient method of detection of point mutation, especially when dealing with multiple samples.

        It has been understand that a laboratory information system(LIS) plays an important role in a hospital information system(HIS). However, few HIS has been equipped with a fully automated LIS. This is partly because, in order to achieve a fully automated LIS, there are many testing equipment has its own communication protocol. In this paper, a bi-directional communication system between an ABBOTT SPECTRUM EPx auto-chemical analyzer and a PC is constructed. With the communication system, it is possible that a PC controls an EPx's operations and that the PC receives test results form the EPx automatically. RS-232C serial ports are used in order to make bi-directional communication possible between them

      • 조밀한 다이아몬드 막의 성장

        RF-MW 혼합프로세스를 사용하여 다이아몬드 막을 만들 때, 우선 RF 플라즈마CVD법으로 실리콘 기판 위에 다이아몬드 시드를 증착시키고, 그 시드로부터 MW 플라즈마CVD법에 의해 다이아몬드 막을 성장시켰다. RF-MW 혼합프로세스를 사용하여 성장된 다이아몬드 막은 MW 프로세스만으로 성장된 막에 비해 결정입계의 크기는 작고, 조밀해졌으며, 결정성 또한 더 좋아졌다. 성장된 다이아몬드 막은 전자현미경, X-선 회절기, Raman분광기 등으로 분석하였다. In case of making the diamond films by using RF-MW mix-process, at first. diamond seeds were deposited on silicon substrate by RF plasma CVD, and then diamond layer grown by MW plasma CVD from the seeds. The grain-size of diamond films deposited by using RF-MW mix-process was smaller and denser, in addition, crystallity of diamond films was better than MW plasma CVD process. The deposited diamond films were analyzed by SEM, XRD, and Raman spectroscopy.

        전치부 공극의 치료후 재발에 관한 임상적 고찰

        전치부 공극의 치료법으로는 교정적인 폐쇄, 수술에 의한 방법, 보철에 의한 치료등의 방법이 있으며 이중 교정적인 폐쇄에 의한 치료법은 가장 보존적인 방법으로 많이 사용되고 있으나 그 치료이후의 보정에 많은 문제가 있었다. 저자등은 전치부 공극을 가진 3증례의 치료후 재발과정과 보정과정을 관찰한 결과 다음과 같이 요약할 수 있었다. 양 중절치가 서로 직접 연결되지 않는 보정장치의 사용은 영구보철물에 의한 경우에도 재발을 가져왔다. 교정적으로 공극을 폐쇄한후, 순소대절제술, circumferential supracrestal fibrotomy 그리고 양 중절치가 서로 연결된 설측 부착 보정장치의 시행으로 좋은 유지 효과를 얻었다. 이상의 결과로 미루어 볼 때 전치부 공극은 교정적 치료후, 순소대절제술, fibrotomy등을 시행한 후 반드시 양 중절치를 연결하는 고정성 보정장치가 필요하다고 생각된다. There are orthodontic closure, surgincal closure, prosthetic solution in the treatment of diastema. The orthodontic closure has been widely used owing to its conservative nature, but retention after treatment has been difficult and problematic. So, authors observed relapse and retention after the orthodontic closure of 3 diastema cases. The results might be summarized as follows : The space that had been approximated between central incisors, was reopened by the use of retention appliance which did not join two central incisors together though fixed prothesis. By the frenectomy, circumferential supracrestal fibrotomy and lingual bonded retainer which joined two central incisors together after the orthodontic closure of diastema, were good results obtained. The frenectomy, fibrotomy, and the fixed retention appliance which joined two central incisors together must be needed after the orthodontic closure of diastema.

        Effect of Graphene on Growth of Neuroblastoma Cells

        The unique properties of graphene have earned much interest in the fields of materials science and condensedmatter physics in recent years. However, the biological applications of graphene remain largely unexplored. In this study, we investigated the conditions and viability of a cell culture exposed to graphene onto glass and SiO2/Si, using a human nerve cell line, SH-SY5Y. Cell viability was 84% when cultured on glass and SiO2/Si coated with graphene as compared with culturing on polystyrene surface. Fluorescence data showed that the presence of graphene did not influence cell morphology. These findings suggest that graphene may be used for biological applications.

        셀레콕시브 및 그 합성유도체들의 항암활성 스크리닝

        Selective COX (cyclooxygenase)-2 inhibitors including celecoxib have been shown to induce apoptosis and cell cycle changes in various tumor cells. New inhibitors are recently being developed as chemomodulating agents. We evaluated celecoxib and screened 150 synthetic compounds for anti-proliferative activities in vitro. Effects of celecoxib on COX activity, cell growth, cell cycle distribution, and apoptosis induction were determined in A549 COX-2 overexpressing human non-small cell lung cancer (NSCLC) cells. The COX inhibition of celecoxib increased with concentration up to 82% at 1μM after 24 hr exposure. Forty μM and 50μM of celecoxib induce G_1 arrest, and TUNEL-positive apoptotic cells, respectively. Among 150 compounds, several compounds were selected for having greater COX-2 inhibitory activity and higher selectivity than celecoxib with growth inhibitory activity. Celecoxib showed concentration-dependent COX inhibitory activity, and ability to induce cell cycle arrest and apoptosis in human NSCLC cells in vitro. Among synthetic analogues screened, several compounds showed promising in vitro activity as COX-2 inhibitory anticancer agents, which warrant further evaluation in vitro and in vivo.
