<P>Deguelin exhibits potent apoptotic and antiangiogenic activities in a variety of transformed cells and cancer cells. Deguelin also exhibits potent tumor suppressive effects in xenograft tumor models for many human cancers. Our initial studies confirmed that deguelin disrupts ATP binding to HSP90 and consequently induces destabilization of its client proteins such as HIF-1α. Interestingly, a fluorescence probe assay revealed that deguelin and its analogues do not compete with ATP binding to the N-terminus of HSP90, unlike most HSP90 inhibitors. To determine the key parts of deguelin that contribute to its potent HSP90 inhibition, as well as its antiproliferative and antiangiogenic activities, we have established a structure–activity relationship (SAR) of deguelin. In the course of these studies, we identified a series of novel and potent HSP90 inhibitors. In particular, analogues <B>54</B> and <B>69</B>, the B- and C-ring-truncated compounds, exhibited excellent antiproliferative activities with IC<SUB>50</SUB> of 140 and 490 nM in the H1299 cell line, respectively, and antiangiogenic activities in zebrafish embryos in a dose dependent manner (0.25–1.25 μM).</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jmcmar/2012/jmcmar.2012.55.issue-24/jm301488q/production/images/medium/jm-2012-01488q_0019.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/jm301488q'>ACS Electronic Supporting Info</A></P>

<P>Enantioselective synthesis of (-)-deguelin was accomplished via an iterative pyran-ring formation approach. The key features involve the anionic addition of a chromene unit to aryloxy alkyl aldehyde for the double cyclization precursor and iterative pyran ring formation by Pd-catalyzed O-arylation and C-arylation, respectively.</P>

Purpose: Laparoscopic Roux-en-Y gastric bypass (LRYGB) and laparoscopic adjustable gastric banding (LAGB) are the most effective intervention and cure in achieving sustained weight loss in the morbidly obese with co-morbities, especially in type 2 diabetes mellitus (DM). Currently, there are few data in the literature presenting early diabetic outcomes between the 2 major bariatric surgeries in Korea. The aim of this study is to observe weight loss, improvement of type 2 DM after LRYGB and LAGB. Methods: 33 retrospective data were obtained from diabetic subjects undergoing LRYGB (n=53) and LAGB (n=77). These measurements included age, sex, body mass index (BMI), pre-operative diabetic duration, post-operative diabetic at follow-up visit after surgery. Paired t-test, and SPSS12.0 were used for statistical analysis. Results: 33 morbidly obese patients with type 2 DM who had both procedures complain of general weakness. Most patients were taking oral medications or insulin and tired of them. Pre and post-op 12 months of percent of excess weight loss (%EWL), fetal bovine serum (FBS), hemoglobin A1c (HbA1C) presented at 0%, 185.6 ㎎/㎗, 8.7% and 70.6%, 123.4 ㎎/㎗, 6.6% in patients who had LRYGB (P<0.05). Pre and post-op 12 months of %EWL, FBS, HbA1C presented at 0%, 180.6 ㎎/㎗, 8.4% and 32.8%, 136.5 ㎎/㎗, 6.8% in patients who had LAGB (P<0.05). Rate of discontinuance or decreasing for treatment of type 2 DM after surgery showed 94% for LRYGB versus 80% for LAGB, and unchanged rates were 6% versus 20%, respectively. Conclusion: This study showed LRYGB and LAGB are quite satisfactory and promising bariatric procedures with significant weight loss and improvement of type 2 DM in Koreans. LRYGB is significantly associated with more weight loss, more diabetic outcome than LAGB. Clinical experiences in Korea with both procedures are limited and a larger number of cases are need, but the recent data shows promising results that are comparable to the international experience.

최근 서비스 지향 아키텍처를 적용한 많은 시스템들이 개발되어 왔고 그에 따라 서비스 지향 아키텍처에 대한 관심도 증가하고 있다. 하지만 여태껏 연구되어왔던 주제들은 전체 아키텍처의 효율적인 구성 및 개발 프로세스 구축에 초점을 맞춘 반면, 데이터 프레임워크를 보다 효율적으로 비즈니스 프레임워크와 분리하는 데 초점을 맞춘 연구는 부족하다. 이에 우리는 실 서비스되고 있는 대용량 서버 관리 서비스를 서비스 지향 아키텍처로 전환하고 그 중 데이터 프레임워크를 보다 추상화시키고 비즈니스 프레임워크와 느슨한 결합을 이룰 수 있도록 하는 연구를 진행하였다. 본 논문에서 제안하는 데이터 프레임워크는 실시간 데이터 전송에 유리한 트리 구조의 데이터를 사용자가 정의한 규칙을 기반으로 비즈니스 프레임워크의 다양한 비즈니스 로직에서 효율적으로 활용할 수 있는 다양한 형태의 데이터로 바꿀 수 있는 계층을 제공한다. 또한 이러한 계층에 대해 실 서비스에서 활용할 수 있는 정의된 제약조건 하에서 실험을 진행하였고 그 결과 만족스러운 성능을 보이는 것으로 나타났다.
An Empirical Performance Analysis on Hadoop via Optimizing the Network Heartbeat Period

To support a large-scale Hadoop cluster, Hadoop heartbeat messages are designed to deliver the significant messages, including task scheduling and completion messages, via piggybacking to reduce the number of messages received by the NameNode. Although Hadoop is designed and optimized for high-throughput computing via batch processing, the real-time processing of large amounts of data in Hadoop is increasingly important. This paper evaluates Hadoop’s performance and costs when the heartbeat period is controlled to support latency sensitive applications. Through an empirical study based on Hadoop 2.0 (YARN) [1] architecture, we improve Hadoop’s I/O performance as well as application performance by up to 13 percent compared to the default configuration. We offer a guideline that predicts the performance, costs and limitations of the total system by controlling the heartbeat period using simple equations. We show that Hive performance can be improved by tuning Hadoop’s heartbeat periods through extensive experiments.
External Mergesort for Flash-Based Solid State Drives

<P>Mergesort is the most widely-known external sorting algorithm, which is used when the data being sorted do not fit into the available main memory. There have been several attempts to improve mergesort by reducing I/O time, since mergesort is I/O intensive. However, these methods assumed that mergesort runs on hard disk drives (HDDs). Flash-based solid state drives (SSDs) are emerging as next generation storage devices and becoming alternatives to HDDs. SSDs outperform HDDs in access latency, because they have no physical arms to move. In addition, SSDs benefit from their inner structure by exploiting internal parallelism, resulting in high I/O bandwidth. Previous methods for improving mergesort focused on reducing random access cost, which is insignificant on SSDs. In this paper we propose an external mergesort algorithm for SSDs called FMsort. FMsort calculates a block read order which is the order of blocks needed in the merge phase. With a block read order, a number of blocks required during the merge phase are read into main memory via multiple asynchronous I/Os. Our experiments show that FMsort outperforms other mergesort algorithms, at an invisible cost of calculating a block read order.</P>
