      • ANXA2 Regulates the Behavior of SGC-7901 Cells

        ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

        A design of in-transit logistics remote video monitoring system is proposed based on cotex-A8 processor hardware and Android software systems platform, by the terminal USB infrared camera driver transplant, video streaming H.264 codec, and 3G wireless transmission network, the in-transit logistics remote video monitoring real-time display is achieved, which can prevent the loss of goods and cargo in logistics process and be served as the evidence for the responsibility distinguishing in the event of goods damage. Managers and customers can track the transport status of goods by computer or mobile phone in real-time, and provide better service to customer, therefore enhance the competitiveness of logistics enterprises.

        An Orthogonal Study of Industrial Scale Colour Fading Process of Cotton Fabric

        Colour fading is now a popular process used for imparting a vintage look to textile and fashion products, whichenhances market value because of the current fashion trends. This study examined a non-aqueous colour fading process with the use of oxygen plasma-induced ozone treatment. An industrial scale machine and commercially available red sulpur-dyed cotton fabric (with 0.5 %, 1.5 % and 2.5 % colour depths) were used in this study. Since the colour fading process factors are inter-related to each other, a specific experiment approach, i.e. orthogonal method, was used for obtaining the optimum conditions in an industrial scale colour fading process. Three process factors used in the industrial scale colour fading process, i.e. (i) oxygen gas concentration (%); (ii) amount of water in fabric (%); and (iii) treatment time (minutes), would be studied in this paper. Through the orthogonal method, the optimum conditions for colour fading of the three colour depths of cotton fabric dyed by red sulphur dye were determined and their optimum conditions were same. The optimum conditions of the colour fading of the three colour depths were: (i) 70 % oxygen gas concentration; (ii) 35 % amount of water in fabric; and (iii) 30 minutes treatment time. Although colour fading conditions are the same, the order of importance of these process factors was different. Unlike the conventional colour fading process, oxygen plasma-induced ozone colour fading treatment can achieve uniform and even colour fading effect in the cotton fabric effectively.

        Radiological gastrostomy: A comparative analysis of different image-guided methods

        Background: Radiographic guided percutaneous gastrostomy has become a safe and effective enteral nutrition method for patients who can not eat by mouth. Fluoroscopy, computed tomography (CT) and cone-beam CT have been routinely used clinically. The aim of this study was to compare the advantages and disadvantages of percutaneous gastrostomy using different radiographic guided methods. Methods: We retrospectively analyzed the clinical data of 538 patients undergoing percutaneous gastrostomy in our department. According to the image guidance method used in gastrostomy, the patients were divided into groups A by fluoroscopy guidance, group B by fluoroscopy combined with C-arm CT guidance, and group C with the whole process CT guidance. The gastrostomy success rate, complication rate, procedure time, and patient radiation dose were analyzed in the three groups. Results: Among 538 patients, 534 were successful and the success rates are 94.3%, 99.3%, and 100% in group A, B, and C, respectively (P > 0.05). There were 3 cases occurred postoperative bleeding as serious adverse events and transferred to surgical gastrostomy. The minor complications include local infection, hyperplasia of granulation tissue, tube obstruction or prolapse, and local pain of the ostomy. The minor complication rates were 10.5%, 10.4%, and 7.7% in group A, B, and C, respectively (P > 0.05). The average procedure time was 25.57 ± 5.99 minutes, 29.01 ± 6.63 minutes, and 45.47 ± 8.98 minutes, respectively (χ2 = 87.98, P < 0.001). The average radiation dosage was 27.30 ± 19.27 mGy, 145.07 ± 106.08 mGy, and 2,590.26 ± 1,088.22 mGy, respectively (χ2 = 204.44, P < 0.001). Conclusion: There were no significant differences in the success rates and complication rates of gastrostomy under the three guiding methods. For difficult cases, CT-guided gastrostomy may be a very useful supplemental method.

      • Component-based model for posttensioned steel connections against progressive collapse

        A component-based method for the estimation of the posttensioned (PT) steel frame against progressive collapse is proposed and presented in the current paper. A mechanical model of PT steel connections is developed and benchmarked with experimental data of a PT beam-column substructure from literature. The developd mechanical models of four PT connections are able to capture the initial elastic stiffness, decompression load, and residual stiffness under lateral loading. Then, analysis of a reduced-scale three-storey two-bay PT steel frame is carried out with sufficient accuracy by incorporating the proposed joint model into the frame analysis. The proposed method is then applied to assessing progressive collapse of a one-storey two-bay PT frame under middle column removal scenario, and is verified against existing experimental and ANSYS finite element results. Three resistance mechanism for progressive collapse of the PT frame are evaluated, which consists of angle flexural mechanism, beam compression arching action and strand tensile catenary action. Finally, parameter analyses of the PT frames are conducted to investigate the effects of the connection details on the behavior and resistance of progressive collapse. The proposed model can be used to predict the quasi-static behavior of PT frames under monotonic vertical loading conditions with satisfactory accuracy.

        Increasing Demeclocycline Production in Streptomyces aureofaciens by Manipulating the Expression of a Novel SARP Family Regulator and Its Genes

        Demeclocycline (DMCTC), a tetracycline derivative antibiotic produced by Streptomyces aureofaciens, has attracted attention owing to its high bioavailability, prolonged maintenance of a therapeutic concentration, and greater efficacy against many infectious microorganisms. However, the productivity of the DMCTC-producing strains has remained low. Thus, it is necessary to identify gene-knockout or amplification targets to increase DMCTC production. Here, we demonstrated that ctcB, which encodes a Streptomyces antibiotic regulatory protein (SARP), and ctcC, which encodes a resistance gene, positively regulate the biosynthesis of DMCTC in S. aureofaciens strain DT1. In particular, overexpression of the ctcB gene in S. aureofaciens DT1 significantly enhanced DMCTC production, resulting in increased expression of ctcG, ctcN, ctcQ, ctcH, ctcV, and ctcC. The deletion of ctcB dramatically reduced the DMCTC level, implying that CtcB is an activator of DMCTC biosynthesis. Although overexpression of the ctcC, which encodes a ribosomal protection protein, enhancing DMCTC biosynthesis in S. aureofaciens DT1, the improvement was limited compared with that achieved by ctcB overexpression. This is the first study to identify the role of ctcB and ctcC in DMCTC accumulation; these genes may also be ideal candidate targets for facilitating DMCTC production by other Streptomyces strains.

        Dynamic increase factor for progressive collapse analysis of semi-rigid steel frames

        An empirical and efficient method is presented for calculating the dynamic increase factor to amplify the applied loads on the affected bays of a steel frame structure with semi-rigid connections. The nonlinear static alternate path analysis is used to evaluate the dynamic responses. First, the polynomial models of the extended end plate and the top and seat connection are modified, and the proposed polynomial model of the flush end plate connection shows good agreement as compared with experimental results. Next, a beam model with nonlinear spring elements and plastic hinges is utilized to incorporate the combined effect of connection flexibility and material nonlinearity. A new step-by-step analysis procedure is established to obtain quickly the dynamic increase factor based on a combination of the pushdown analysis and nonlinear dynamic analysis. Finally, the modified dynamic increase factor equation, defined as a function of the maximum ratio value of energy demand to energy capacity of an affected beam, is derived by curve fitting data points generated by the different analysis cases with different column removal scenarios and five types of semi-rigid connections.

        SP0454, A Putative Threonine Dehydratase, Is Required For Pneumococcal Virulence In Mice

        Increasing pressure in antibiotic resistance and the requirement for the design of new vaccines are the objectives of clarifying the putative virulence factors in pneumococcal infection. In this study, the putative threonine dehydratase sp0454 was inactivated by erythromycin-resistance cassette replacement in Streptococcus pneumoniae CMCC 31203 strain. The sp0454 mutant was tested for cell growth, adherence, colonization, and virulence in a murine model. The Δsp0454 mutant showed decreased ability for colonization and impaired ability to adhere to A549 cells. However, the SP0454 polypeptide or its antiserum did not affect pneumococcal CMCC 31203 adhesion to A549 cells. The sp0454 deletion mutant was less virulent in a murine intranasal infection model. Real-time RT-PCR analysis revealed significant decrease of the pneumococcal surface antigen A expression in the sp0454 mutant. These results suggest that SP0454 contributes to virulence and colonization, which could be explained in part by modulating the expression of other virulence factors, such as psaA in pneumococcal infection.

        Progressive collapse analysis of steel frame structure based on the energy principle

        The progressive collapse potential of steel moment framed structures due to abrupt removal of a column is investigated based on the energy principle. Based on the changes of component's internal energy, this paper analyzes structural member's sensitivity to abrupt removal of a column to determine a sub-structure resisting progressive collapse. An energy-based structural damage index is defined to judge whether progressive collapse occurs in a structure. Then, a simplified beam damage model is proposed to analyze the energies absorbed and dissipated by structural beams at large deflections, and a simplified modified plastic hinges model is developed to consider catenary action in beams. In addition, the correlation between bending moment and axial force in a beam during the whole deformation development process is analyzed and modified, which shows good agreement with the experimental results.
