Drug-induced hyperglycaemia and diabetes: pharmacogenomics perspectives

        Mou-Ze Liu,Hai-Yan He,Jian-Quan Luo,Fa-Zhong He,Zhang-Ren Chen,Yi-Ping Liu,Da-Xiong Xiang,Hong-Hao Zhou,Wei Zhang 대한약학회 2018 Archives of Pharmacal Research Vol.41 No.7

Drug-induced diabetes is widely reported inclinical conditions, and it is becoming a global issuebecause of its potential to increase the risk of severe cardiovascularcomplications. However, which drug mechanismsexert their diabetogenic effects and why the effectspresent significant inter-individual differences remain largelyunknown. Pharmacogenomics, which is the study ofhow genomic variation influences drug responses, providesan explanation for individual differences in drug-induceddiabetes. We highlight that pharmacogenomics can beinvolved in regulating the expression of genes in signalingpathways related to the pharmacokinetics or pharmacodynamicsof drugs or the pathogenesis of diabetes, contributingto the differences in drug-induced glucoseimpairment. The pharmacogenomics studies of the majordiabetogenic drugs are reviewed, including calcineurininhibitors, antipsychotics, hormones, and antihypertensivedrugs. We intend to elucidate the genetic basis of druginduceddiabetes and pave the way for the precise use ofthese drugs in the clinic.

      • The DNA Repair Gene ERCC6 rs1917799 Polymorphism is Associated with Gastric Cancer Risk in Chinese

Liu, Jing-Wei,He, Cai-Yun,Sun, Li-Ping,Xu, Qian,Xing, Cheng-Zhong,Yuan, Yuan Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.10

Objective: Excision repair cross-complementing group 6 (ERCC6) is a major component of the nucleotide excision repair pathway that plays an important role in maintaining genomic stability and integrity. Several recent studies suggested a link of ERCC6 polymorphisms with susceptibility to various cancers. However, the relation of ERCC6 polymorphism with gastric cancer (GC) risk remains elusive. In this sex- and age-matched case-control study including 402 GC cases and 804 cancer-free controls, we aimed to investigate the association between a potentially functional polymorphism (rs1917799 T>G) in the ERCC6 regulatory region and GC risk. Methods: The genotypes of rs1917799 were determined by Sequenom MassARRAY platform and the status of Helicobacter pylori infection was detected by enzyme-linked immunosorbent assay. Odd ratios (ORs) and 95% confidential interval (CI) were calculated by logistic regression analysis. Results: Compared with the common TT genotype, the ERCC6 rs1917799 GG genotype was associated with increased GC risk (adjusted OR=1.46, 95%CI: 1.03-2.08, P=0.035). When compared with (GT+TT) genotypes, the GG genotype also demonstrated a statistical association with increased GC risk (adjusted OR=1.38, 95%CI: 1.01-1.89, P=0.044). This was also observed for the male subpopulation (GG vs. TT: adjusted OR=1.71, 95%CI: 1.12-2.62, P=0.013; G allele vs. T allele: adjusted OR=1.32, 95%CI: 1.07-1.62, P=0.009). Genetic effects on increased GC risk tended to be enhanced by H. pylori infection, smoking and drinking, but their interaction effects on GC risk did not reach statistical significance. Conclusions: ERCC6 rs1917799 GG genotype might be associated with increased GC risk in Chinese, especially in males.

Numerical Research on Suppression of Thermally Induced Wavefront Distortion of Solid-state Laser Based on Neural Network

        Liu, Hang,He, Ping,Wang, Juntao,Wang, Dan,Shang, Jianli Optical Society of Korea 2022 Current Optics and Photonics Vol.6 No.5

To account for the internal thermal effects of solid-state lasers, a method using a back propagation (BP) neural network integrated with a particle swarm optimization (PSO) algorithm is developed, which is a new wavefront distortion correction technique. In particular, by using a slab laser model, a series of fiber pumped sources are employed to form a controlled array to pump the gain medium, allowing the internal temperature field of the gain medium to be designed by altering the power of each pump source. Furthermore, the BP artificial neural network is employed to construct a nonlinear mapping relationship between the power matrix of the pump array and the thermally induced wavefront aberration. Lastly, the suppression of thermally induced wavefront distortion can be achieved by changing the power matrix of the pump array and obtaining the optimal pump light intensity distribution combined using the PSO algorithm. The minimal beam quality β can be obtained by optimally distributing the pumping light. Compared with the method of designing uniform pumping light into the gain medium, the theoretically computed single pass beam quality β value is optimized from 5.34 to 1.28. In this numerical analysis, experiments are conducted to validate the relationship between the thermally generated wavefront and certain pumping light distributions.

      • HMGB1 Promotes the Synthesis of Pro-IL-1β and Pro-IL-18 by Activation of p38 MAPK and NF-κB Through Receptors for Advanced Glycation End-products in Macrophages

        He, Qiang,You, Hong,Li, Xin-Min,Liu, Tian-Hui,Wang, Ping,Wang, Bao-En Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.4

The high mobility group box-1 (HMGB1) protein and NALP3 inflammasome have been identified to play important roles in inflammation and cancer pathogenesis, but the relationships between the two and cancer remain unclear. The current study investigated the relationship between HMGB1 and the NALP3 inflammasome in THP-1 macrophages. HMGB1 was found unable to activate the NALP3 inflammasome and failed to induce the release of the IL-$1{\beta}$ and IL-18 in THP-1 macrophages. HMGB1 was also found significantly enhanced the activity of ATP to induce IL-$1{\beta}$ and IL-18 by the induction of increased expression of pro-IL-$1{\beta}$ and pro-IL-18. This process was dependent on activation of RAGE, MAPK p38 and NF-${\kappa}B$ signaling pathway. These results demonstrate that HMGB1 promotes the synthesis of pro-IL-$1{\beta}$ and pro-IL-18 in THP-1 macrophages by the activation of p38 MAPK and NF-${\kappa}B$ through RAGE. HMGB1 likely plays an important role in the first step of the release of the IL-$1{\beta}$ and IL-18, preparing for other cytokines to induce excessive release of IL-$1{\beta}$ and IL-18 which promote inflammation and cancer progression.

Dieless bulging and nonlinear buckling of longan-shaped pressure hull

He Rui,Zhang Jian,Kaewunruen Sakdirat,Zhan Ming,Liu Ping 대한조선학회 2023 International Journal of Naval Architecture and Oc Vol.15 No.-

In this study, the dieless bulging and nonlinear buckling behaviours of a stainless longan-shaped pressure hull were investigated. According to the Cassini oval equation, the longan shape has a shape index of 0.1. The lengths of the major and minor axes of the designed longan-shaped pressure hull were approximately 400 and 396 mm, respectively. The wall thickness of this pressure hull was 1 mm. The Huberevon Mises stress and first yielding load of the designed longan-shaped shell and its inscribed polyhedral preform were investigated analytically and experimentally. Moreover, the dieless bulging of the designed longan-shaped pressure hull was investigated numerically and experimentally by using the nonlinear finite element method and a hydrostatic test, respectively. The analytical, experimental, and numerical results exhibited good agreement with each other.

Temporal and spatial impact of Spartina alterniflora invasion on methanogens community in Chongming Island, China

        Xue Ping Chen,Jing Sun,Yi Wang,Heng Yang Zhang,Chi Quan He,Xiao Yan Liu,Nai Shun Bu,Xi-En Long 한국미생물학회 2018 The journal of microbiology Vol.56 No.7

Methane production by methanogens in wetland is recognized as a significant contributor to global warming. Spartina alterniflora (S. alterniflora), which is an invasion plant in China’s wetland, was reported to have enormous effects on methane production. But studies on shifts in the methanogen community in response to S. alterniflora invasion at temporal and spatial scales in the initial invasion years are rare. Sediments derived from the invasive species S. alterniflora and the native species Phragmites australis (P. australis) in pairwise sites and an invasion chronosequence patch (4 years) were analyzed to investigate the abundance and community structure of methanogens using quantitative real-time PCR (qPCR) and Denaturing gradient gel electrophoresis (DGGE) cloning of the methyl-coenzyme M reductase A (mcrA) gene. For the pairwise sites, the abundance of methanogens in S. alterniflora soils was lower than that of P. australis soils. For the chronosequence patch, the abundance and diversity of methanogens was highest in the soil subjected to two years invasion, in which we detected some rare groups including Methanocellales and Methanococcales. These results indicated a priming effect at the initial invasion stages of S. alterniflora for microorganisms in the soil, which was also supported by the diverse root exudates. The shifts of methanogen communities after S. alterniflora invasion were due to changes in pH, salinity and sulfate. The results indicate that root exudates from S. alterniflora have a priming effect on methanogens in the initial years after invasion, and the predominate methylotrophic groups (Methanosarcinales) may adapt to the availability of diverse substrates and reflects the potential for high methane production after invasion by S. alterniflora.

        Qin Ping Yu,Ding Yuan Feng,Juan Xiao,Fan Wu,Xiao Jun He,Min Hao Xia,Tao Dong,Yi Hua Liu,Hui Ze Tan,Shi Geng Zou,Tao Zheng,Xian Hua Ou,Jian Jun Zuo 아세아·태평양축산학회 2017 Animal Bioscience Vol.30 No.12

Objective: This experiment investigated meat color, myoglobin content, enzyme activities, and expression of genes associated with oxidative potential of pigs slaughtered at different growth stages. Methods: Sixty 4-week-old Duroc×Landrace×Yorkshire pigs were assigned to 6 replicate groups, each containing 10 pigs. One pig from each group was sacrificed at day 35, 63, 98, and 161 to isolate longissimus dorsi and triceps muscles. Results: Meat color scores were higher in pigs at 35 d than those at 63 d and 98 d (p<0.05), and those at 98 d were lower than those at 161 d (p<0.05). The total myoglobin was higher on 161 d compared with those at 63 d and 98 d (p<0.05). Increase in the proportions of metmyoglobin and deoxymyoglobin and a decrease in oxymyoglobin were observed between days 35 and 161 (p<0.05). Meat color scores were correlated to the proportion of oxymyoglobin (r = 0.59, p<0.01), and negatively correlated with deoxymyoglobin and metmyoglobin content (r = –0.48 and –0.62, p<0.05). Malate dehydrogenase (MDH) activity at 35 d and 98 d was higher than that at 161 d (p<0.05). The highest lactate dehydrogenase/MDH ratio was achieved at 161 d (p<0.05). Calcineurin mRNA expression decreased at 35 d compared to that at 63 d and 98 d (p<0.05). Myocyte enhancer factor 2 mRNA results indicated a higher expression at 161 d than that at 63 d and 98 d (p<0.05). Conclusion: Porcine meat color, myoglobin content, enzyme activities, and genes associated with oxidative potential varied at different stages.
