<P><B>Objectives.</B> The present study was conducted in order to assess whether extracts or isolated compounds from <I>Vigna angularis</I> were able to suppress IL-6 signalling and to show the therapeutic effect on collagen-induced arthritis (CIA) in mice.</P><P><B>Methods.</B> The effect of <I>V. angularis</I> on IL-6 signalling was studied by measuring Stat3-dependent luciferase activity, expression of inflammation-related genes, and phosphorylation of Janus kinase 2 (JAK2), signal transducer and activator of transcription 3 (STAT3) and extracellular signal-regulated kinase (ERK) induced by IL-6. CIA was induced by immunizing with bovine type II collagen. <I>V. angularis</I> extract (VAE) was administrated orally at 50 and 100 mg/kg from day 1 to day 28. Induction of arthritis was evaluated with a visual scoring system and histological analysis.</P><P><B>Results.</B> Extracts or two triterpenoid compounds from <I>V. angularis</I> showed potent inhibitory effects on pSTAT3-inducible luciferase activity, STAT3 tyrosine phosphorylation and the expression of inflammation-related genes induced by IL-6. Administration of VAE significantly suppressed the progression of CIA, accompanied by a reduced antibody response to type II collagen and protection from tissue damage in knee joints.</P><P><B>Conclusion.</B> Administration of VAE has a therapeutic effect on CIA and this effect is associated with the inhibitory activity on IL-6/STAT3 signalling. These results suggest that extracts or compounds from <I>V. angularis</I> could be a useful treatment for diseases related to IL-6, including RA.</P>

Cholangiocarcinoma (CC) is a chemoresistant intrahepatic bile duct carcinoma with a poor prognosis. The aims of this study were to identify molecular pathways that enhance sesquiterpene lactone parthenolide (PTL)-induced anticancer effects on CC cells. The effects of PTL on apoptosis and hemoxygenase-1 (HO-1)induction were examined in CC cell lines. The enhancement of PTL-mediated apoptosis by modulation of HO-1 expression and the mechanisms involved were also examined in an in vitro cell system. Low PTL concentrations (5 to 10 μM) led to Nrf2-dependent HO-1 induction,which attenuated the apoptogenic effect of PTL in Choi-CK and SCK cells. PTL-mediated apoptosis was enhanced by the protein kinase C-alpha inhibitor Ro317549 (Ro) through inhibition of expression and nuclear translocation of Nrf2, resulting in blockage of HO-1 expression. Finally, HO-1 silencing resulted in enhancement of apoptotic cell death in CC cells. The combination of PTL and Ro efficiently improved tumor growth inhibition compared to treatment with either agent alone in an in vivo subcutaneous tumor model. In conclusion, the modulation of HO-1 expression substantially improved the anticancer effect of PTL. The combination of PTL and Ro could prove to be a valuable chemotherapeutic strategy for CC.

There have been numerous human casualties caused by various incidents around the world; overcrowding and convenience from urban development are becoming dangerous factors in people"s lives. which leads to the purpose of the study; to plan the National Fire History Museum to enhance people’s safety awareness from education and furthermore, prevent accidents. The study analyzes exhibition programs and spacial compositions of Fire History museums and proposes an exhibition plan for an efficient exhibition program and spatial composition. Fire museum cases overseas were analyzed with an exhibition system of narratives to identify problems from existing exhibition programs and spatial composition and propose a solution. The research methods were analyzing preceding studies, the constructive system of the exhibition, and composing elements of narratives. According to the analysis, fire museum overseas had simple narrative structure and text-oriented education programs—visitors can easily lose interest from the exhibition—lack of narrative characteristics was analyzed. Under the assumption that the museum program and space provides interesting experience to visitors from analysis of visitor’s various sensual experiences and information can lead to a better educational effect than before. This study analyzes the program and spatial composition of fire museums overseas and structure of exhibition narratives to provide guidelines—structural elements of programs, spatial composition, and narratives—designing a Fire History museum. Furthermore, the study focuses on the problems that existing exhibition systems have and provide academic help to the National Fire History museum; Effective education of knowledge and thinking from the exhibition.

Objective: Insufficiency fracture (IF) occurs when normal or physiological muscular activity stresses a bone that is deficient in mineral or elastic resistance. We studied clinical characteristics of IF in patients with chronic inflammatory joint diseases in Korea. Methods: Between Aug. 1997 and Feb. 2003, thirty five patients with 77 fractures were studied at the authors` institution when they were being treated for their rheumatic diseases. The clinical and laboratory data were collected by review of medical record retrospectively. Results: All patients except four were postmenopausal women (mean age 63.0±10.0 years) with long disease duration (mean 14.2±11.6 years). Thirty three patients had rheumatoid arthritis, 1 ankylosing spondylitis and 1 systemic lupus erythematosus. Twenty nine patients (85.7%) were receiving regular steroid treatment (mean dose 4.0±2.3 mg/day, mean duration 6.1±4.2 years). Twenty four patients were treated with methotrexate. The significant reduction in their bone mineral density was found 27 patients based on BMD or QCT. Eight patients without osteoporosis were treated with steroid or MTX. Twenty three patients were ever used for osteoporosis treatment. Most patients except four presented with pain in the low back, groin, hip, pelvic, leg and knee. Initial simple radiography was positive in only 7 patients, with vertebral compression fracture in 11 patients and no effect on mobility except ten. Diagnosis was delayed (mean duration of symptom until diagnosis was 45.6±64.5 days). IF was confirmed using the bone scan. Sacrum and pelvic bone was most frequently affected site. The other sites were SI joint, iliac wing, symphysis pubis, acetabulum and femur neck. Twenty nine patients required in-patient stay (mean 17.4 days). All but one patient showed an uneventful recovery with conservative treatment. Conclusion: The low grade nature of symptoms, minimal effect on mobility, absence of significant trauma and missed on initial plain radiography make diagnosis difficult and delayed. IF should be suspected in cases of unexplained pain with local tenderness in patients of chronic inflammatory joint diseases. The technetium-99m diphosphonate bone scintigraphy was valuable diagnostic tool in the early recognition of IF.
