      • Hair growth promoting effect of dermal papilla like tissues from canine adipose-derived mesenchymal stem cells through vascular endothelial growth factor

LEE, Aeri,BAE, Sohee,LEE, Seung Hoon,KWEON, Oh-Kyeong,KIM, Wan Hee The Japanese Society of Veterinary Science 2016 The Journal of veterinary medical science Vol.78 No.12

<P>The purpose of this study was to investigate the protein expression pattern and the <I>in vivo</I> trichogenicity of dermal papilla-like tissues (DPLTs) made from canine adipose-derived mesenchymal stem cells (ASCs) in athymic nude mice. Canine ASCs were isolated and cultured from adipose tissue, and differentiation was induced by culturing ASCs in dermal papilla forming media. DPLTs were embedded in collagen gel, and their structural characteristics and protein expression were evaluated by hematoxylin and eosin stain and immunohistochemistry. Athymic nude mice were divided into two groups (control and DPLTs groups), and DPLTs were injected in skin wounds of mice in the DPLTs group. The trichogenicity of DPLTs was assessed by gross and histological evaluations for 30 days. The fate and the growth factor-secretion effect of DPLTs were evaluated by immunohistochemistry and Western blotting. DPLTs have a compact aggregated structure, form extracellular matrix and highly express the protein specific for dermal papillae, including ALP and versican. New hair follicle formation was remarkable in nude mice of the DPLTs group in gross findings and H&E stain. Vascularization was increased in the DPLTs group, which was the effect of vascular endothelial growth factor secreted by DPLTs <I>in vitro</I> and <I>in vivo</I>. These data suggest that engineered canine DPLTs have characteristics of dermal papillae and have a positive effect on hair regeneration by secreting growth factors.</P>

The Effect of Learners’ Characteristics on Learning Performance in Non-Face-To-Face Liberal Arts Coding Class

        Aeri Lee(Aeri Lee) J-INSTITUTE 2022 Robotics & AI Ethics Vol.7 No.2

Purpose: In this study, we investigate the characteristics of learners in the liberal arts coding class and study the relationship between learners’ characteristics and learning outcomes to find methods to enhance learning performance. Through this study, we intend to provide the basic data to be used by the schools and educators operating coding education in the real world to establish the teaching strategy for coding education according to the characteristics of the learners. Method: The collected data was analyzed using SPSS 23, as follows. The reliability of the measurement tools on self-efficacy, learning interest, learning motivation, digital literacy, and learning satisfaction were calculated and verified. Through a survey, data on self-efficacy, learning motivation, digital device efficacy, learning satis-faction, and academic achievement were collected and descriptive statistical analysis was performed to identify trends. We performed multiple regression analyses with self-efficacy, learning motivation, and digital device ef-ficacy as independent variables, whereas learning satisfaction and academic achievement as dependent varia-bles. Before the regression analysis, a Pearson correlation analysis was performed between self-efficacy, learning motivation, digital device efficacy, learning satisfaction, and academic achievement to confirm the significance of the relationship between the measurement variables. Results: The significance test results of the regression model showed that self-efficacy and digital device effi-cacy had a positive effect on learning satisfaction and learning motivation and digital device efficacy also had a positive effect on academic achievement. In other words, we evaluated that higher self-efficacy and digital device efficacy lead to higher learning satisfaction whereas higher learning motivation and digital device efficacy lead to higher academic achievement. Conclusion: Through this study, we confirmed that learners’ characteristics have a high influence on both learning satisfaction and academic achievement. In the future study, we plan to add multiple factors of learners’ characteristics that affect learning performance and studies on educators’ characteristics to obtain various and meaningful results.

Apoptosis-induced Cell Growth Inhibitory Effects of a Novel Compound, As4O6 in a Cervical Cancer Cell Line, SiHa in Vitro

        Woong-Shick Ahn,Su-Mi Bae,Sun-Young Kwak,Ae Ju Lee,Aery Lee,Yong Seok Lee,Il-Ju Bae,Jin-Young Yoo,Young-Joo Lee,김종국,Hong-Seok Chang 대한암예방학회 2006 Journal of cancer prevention Vol.11 No.1

As2O3 has been reported to be effective for treating acute leukemia and induce apoptosis in many tumor cells. In this study, we evaluated the ability of a novel arsenical compound, As4O6, along with As2O3 to induce cell growth inhibition as well as apoptosis in human cervical cancer cells, SiHa cells in vitro. To examine the levels of apoptosis, SiHa cells were given two sensitive doses, 0.5 and 1μM of arsenical compounds, and then, DNA fragmentation assay and FACS analysis were conducted. In addition, Western blotting assay was done for the identification of target molecules for apoptosis. Both As2O3 and As4O6 caused dose-dependent inhibition of SiHa cell proliferation. In particular, As4O6 was more effective for suppressing SiHa cell growth, as compared to As2O3. In parallel with inhibition of cell proliferation, As4O6 caused an increase of the sub-G1 cell population significantly more than As2O3, as determined by propidium iodide DNA staining. This was confirmed by DNA fragmentation assay and annexin V staining. Western blotting analysis also showed that the proliferating cell nuclear antigen (PCNA) expression was suppressed by As4O6 significantly more than As2O3, and that Bcl-XL with sequence homology to Bcl-2 was significantly suppressed by As4O6. However, apoptosis-related proteins, p21 and Bax, were expressed by As4O6 significantly more than As2O3. Taken together, these findings suggest that a novel arsenic compound, As4O6 possesses more potent anti-proliferative effects on human cervical cancer cells with induction of apoptosis at least through activation in p21 and Bax proteins in vitro. Landing with Visual Control Reveals Limb Control for Intrinsic Stability

Lee, Aeri,Hyun, Seunghyun,Ryew, Checheong The Institute of Internet 2020 International Journal of Internet, Broadcasting an Vol.12 No.3

Repetition of landing with visual control in sports and training is common, yet it remains unknown how landing with visual control affects postural stability and lower limb kinetics. The purpose of this study was to test the hypothesis that landing with visual control will influence on lower limb control for intrinsic dynamic postural stability. Kinematics and kinetics variables were recorded automatically when all participants (n=10, mean age: 22.00±1.63 years, mean heights: 177.27±5.45 cm, mean mass: 73.36±2.80 kg) performed drop landings from 30 cm platform. Visual control showed higher medial-lateral force, peak vertical force, loading rate than visual information condition. This was resulted from more stiff leg and less time to peak vertical force in visual control condition. Leg stiffness may decrease due to increase of perturbation of vertical center of gravity, but landing strategy that decreases impulse force was shifted in visual control condition during drop landing. These mechanism explains why rate of injury increase.

There was no mediating effect of learning flow in the relationship between learn-ing motivation, class content, and academic achievement. In online liberal arts coding classes, learning flow was found to have a complete mediating effect in the relationship between learning motivation and learning satisfaction. In addition, learning flow was found to partially mediate between class content and learning satisfaction. Conclusion: Through this study, it can be confirmed that learning motivation and class content factors directly affect academic achievement and significantly affect learning satisfaction of online liberal arts coding classes through learning flow.

      • Collagen gel three-dimensional matrices combined with adhesive proteins stimulate neuronal differentiation of mesenchymal stem cells

        Lee, Jae Ho,Yu, Hye-Sun,Lee, Gil-Su,Ji, Aeri,Hyun, Jung Keun,Kim, Hae-Won The Royal Society 2011 Journal of the Royal Society, Interface Vol.8 No.60

<P>Three-dimensional gel matrices provide specialized microenvironments that mimic native tissues and enable stem cells to grow and differentiate into specific cell types. Here, we show that collagen three-dimensional gel matrices prepared in combination with adhesive proteins, such as fibronectin (FN) and laminin (LN), provide significant cues to the differentiation into neuronal lineage of mesenchymal stem cells (MSCs) derived from rat bone marrow. When cultured within either a three-dimensional collagen gel alone or one containing either FN or LN, and free of nerve growth factor (NGF), the MSCs showed the development of numerous neurite outgrowths. These were, however, not readily observed in two-dimensional culture without the use of NGF. Immunofluorescence staining, western blot and fluorescence-activated cell sorting analyses demonstrated that a large population of cells was positive for NeuN and glial fibrillary acidic protein, which are specific to neuronal cells, when cultured in the three-dimensional collagen gel. The dependence of the neuronal differentiation of MSCs on the adhesive proteins containing three-dimensional gel matrices is considered to be closely related to focal adhesion kinase (FAK) activation through integrin receptor binding, as revealed by an experiment showing no neuronal outgrowth in the FAK-knockdown cells and stimulation of integrin β1 gene. The results provided herein suggest the potential role of three-dimensional collagen-based gel matrices combined with adhesive proteins in the neuronal differentiation of MSCs, even without the use of chemical differentiation factors. Furthermore, these findings suggest that three-dimensional gel matrices might be useful as nerve-regenerative scaffolds.</P>

Hesperetin suppresses LPS/high glucose-induced inflammatory responses via TLR/MyD88/NF-κB signaling pathways in THP-1 cells

        Aeri Lee,HyunJi Gu,Min-Hee Gwon,Jung-Mi Yun 한국영양학회 2021 Nutrition Research and Practice Vol.15 No.5

BACKGROUND/OBJECTIVES: Unregulated inflammatory responses caused by hyperglycemia may induce diabetes complications. Hesperetin, a bioflavonoid, is a glycoside in citrus fruits and is known to have antioxidant and anticarcinogenic properties. However, the effect of inflammation on the diabetic environment has not been reported to date. In this study, we investigated the effect of hesperetin on proinflammatory cytokine secretion and its underlying mechanistic regulation in THP-1 macrophages with co-treatment LPS and hyperglycemic conditions. MATERIALS/METHODS: THP-1 cells differentiated by PMA (1 μM) were cultured for 48 h in the presence or absence of hesperetin under normoglycemic (5.5 mM/L glucose) or hyperglycemic (25 mM/L glucose) conditions and then treated with LPS (100 ng/mL) for 6 h before harvesting. Inflammation-related proteins and mRNA levels were evaluated by enzyme-linked immunosorbent assay, western blot, and quantitative polymerase chain reaction analyses. RESULTS: Hesperetin (0–100 μM, 48 h) treatment did not affect cell viability. The tumor necrosis factor-α and interleukin-6 levels increased in cells co-treated with LPS under hyperglycemic conditions compared to normoglycemic conditions, and these increases were decreased by hesperetin treatment. The TLR2/4 and MyD88 activity levels increased in cells co-treated with LPS under hyperglycemic conditions compared to normoglycemic conditions; however, hesperetin treatment inhibited the TLR2/4 and MyD88 activity increases. In addition, nuclear factor-κB (NF-κB) and Acetyl-NF-κB levels increased in response to treatment with LPS under hyperglycemic conditions compared to normoglycemic conditions, but those levels were decreased when treated with hesperetin. SIRT3 and SIRT6 expressions were increased by hesperetin treatment. CONCLUSIONS: Our results suggest that hesperetin may be a potential agent for suppressing inflammation in diabetes.
