        Nanomemulsion of megestrol acetate for improved oral bioavailability and reduced food effect

        Megestrol acetate (MGA) belongs to the BCSclass II drugs with low solubility and high permeability,and its oral absorption in conventional dosage form MGAmicrocrystal suspension (MGA MS) is very limited andgreatly affected by food. In this study, MGA nanoemulsion(MGA NE) was formulated based on solubility, phasediagramand release studies. Then oral bioavailability ofMGA NE and MGA MS was evaluated. A randomizedtwo-way crossover trial was conducted on six male dogsunder fed and fasting conditions. Blood concentrations ofMGA were analyzed using LC–MS/MS. MGA NE yielded5.00-fold higher oral bioavailability in fasting conditionsand displayed more stable absorption profiles after foodintake compared with MGA MS.

        In silico investigation of Panax ginseng lead compounds against COVID-19 associated platelet activation and thromboembolism

        Hypercoagulability is frequently observed in patients with severe coronavirus disease-2019 (COVID-19). Platelets are a favorable target for effectively treating hypercoagulability in COVID-19 patients as platelet hyperactivity has also been observed. It is difficult to develop a treatment for COVID-19 that will be effective against all variants and the use of antivirals may not be fully effective against COVID-19 as activated platelets have been detected in patients with COVID-19. Therefore, patients with less severe side effects often turn toward natural remedies. Numerous phytochemicals are being investigated for their potential to treat a variety of illnesses, including cancer and bacterial and viral infections. Natural products have been used to alleviate COVID-19 symptoms. Panax ginseng has potential for managing cardiovascular diseases and could be a treatment for COVID-19 by targeting the coagulation cascade and platelet activation. Using molecular docking, we analyzed the interactions of bioactive chemicals in P. ginseng with important proteins and receptors involved in platelet activation. Furthermore, the SwissADME online tool was used to calculate the pharmacokinetics and drug-likeness properties of the lead compounds of P. ginseng. Dianthramine, deoxyharrtingtonine, and suchilactone were determined to have favorable pharmacokinetic profiles.

      • Topology Optimization for Large-displacement Compliant Mechanisms Using Element Free Galerkin Method

        This paper presents a topology optimization approach using element-free Galerkin method (EFGM) for the optimal design of compliant mechanisms with geometrically non-linearity. Meshless method has an advantage over the finite element method(FEM) because it is more capable of handling large deformation resulted from geometrical nonlinearity. Therefore, in this paper, EFGM is employed to discretize the governing equations and the bulk density field. The sensitivity analysis of the optimization problem is performed by incorporating the adjoint approach with the meshless method. The Lagrange multipliers method adjusted for imposition of both the concentrated and continuous essential boundary conditions in the EFGM is proposed in details. The optimization mathematical formulation is developed to convert the multi-criteria problem to an equivalent single-objective problem. The popularly applied interpolation scheme, solid isotropic material with penalization (SIMP), is used to indicate the dependence of material property upon on pseudo densities discretized to the integration points. A well studied numerical example has been applied to demonstrate the proposed approach works very well and the non-linear EFGM can obtain the better topologies than the linear EFGM to design large-displacement compliant mechanisms.

        Certificateless multi-signer universal designated multi-verifier signature from elliptic curve group

        Certificateless public key cryptography resolves the certificate management problem in traditional public key cryptography and the key escrow problem in identity-based cryptography. In recent years, some good results have been achieved in speeding up the computation of bilinear pairing. However, the computation cost of the pairing is much higher than that of the scalar multiplication over the elliptic curve group. Therefore, it is still significant to design cryptosystem without pairing operations. A multi-signer universal designated multi-verifier signature scheme allows a set of signers to cooperatively generate a public verifiable signature, the signature holder then can propose a new signature such that only the designated set of verifiers can verify it. Multi-signer universal designated multi-verifier signatures are suitable in many different practical applications such as electronic tenders, electronic voting and electronic auctions. In this paper, we propose a certificateless multi-signer universal designated multi-verifier signature scheme and prove the security in the random oracle model. Our scheme does not use pairing operation. To the best of our knowledge, our scheme is the first certificateless multi-signer universal designated multi-verifier signature scheme.

        Ginsenoside Rg3 reduces the adhesion, invasion, and intracellular survival of Salmonella enterica serovar Typhimurium

        Background: Invasive infections due to foodborne pathogens, including Salmonella enterica serovar Typhimurium, are prevalent and life-threatening. This study aimed to evaluate the effects of ginsenoside Rg3 (Rg3) on the adhesion, invasion, and intracellular survival of S. Typhimurium. Methods: The impacts of Rg3 on bacterial growth and host cell viability were determined using the time kill and the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assays, respectively. Gentamicin assay and confocal microscopic examination were undertaken to determine the effects of Rg3 on the adhesive and invasive abilities of S. Typhimurium to Caco-2 and RAW264.7 cells. Quantitative reverse transcription polymerase chain reaction was performed to assess the expression of genes correlated with the adhesion, invasion, and virulence of S. Typhimurium. Results: Subinhibitory concentrations of Rg3 significantly reduced (p < 0.05) the adhesion, invasion, and intracellular survival of S. Typhimurium. Rg3 considerably reduced (p < 0.05) the bacterial motility as well as the release of nitrite from infected macrophages in a concentration-dependent manner. The expression of genes related to the adhesion, invasion, quorum sensing, and virulence of S. Typhimurium including cheY, hilA, OmpD, PrgK, rsgE, SdiA, and SipB was significantly reduced after Rg3 treatment. Besides, the compound downregulated rac-1 and Cdc-42 that are essential for actin remodeling and membrane ruffling, thereby facilitating Salmonella entry into host cells. This report is the first to describe the effects of Rg3 on “trigger” entry mechanism and intracellular survival S. Typhimurium. Conclusion: Rg3 could be considered as a supplement agent to prevent S. Typhimurium infection.

        Isolation, Molecular Characterization and Antibiotic Susceptibility Pattern of Vibrio parahaemolyticus from Aquatic Products in the Southern Fujian Coast, China

        Vibrio parahaemolyticus is a major gastroenteritis-causing pathogen in many Asian countries. Antimicrobial resistance in V. parahaemolyticus has been recognized as a critical threat to food safety. In this study, we determined the prevalence and incidence of antimicrobial resistance in V. parahaemolyticus in the southern Fujian coast, China. A total of 62 isolates were confirmed in retail aquatic products from June to October of 2018. The serotype O3:K6 strains, the virulence genes tdh and trh, antibiotic susceptibility and molecular typing were investigated. Then plasmid profiling analysis and curing experiment were performed for multidrug-resistant strains. The results showed that the total occurrence of V. parahaemolyticus was 31% out of 200 samples. Five strains (8.1%) out of 62 isolates were identified as the V. parahaemolyticus O3:K6 pandemic clone. A large majority of isolates exhibited higher resistance to penicillin (77.4%), oxacillin (71%), ampicillin (66.1%) and vancomycin (59.7%). Seventy-one percent (44/62) of the isolates exhibited multiple antimicrobial resistance. All 62 isolates were grouped into 7 clusters by randomly amplified polymorphic DNA, and most of the isolates (80.6%) were distributed within cluster A. Plasmids were detected in approximately 75% of the isolates, and seven different profiles were observed. Seventy-six percent (25/33) of the isolates carrying the plasmids were eliminated by 0.006% SDS incubated at 42°C, a sublethal condition. The occurrence of multidrug-resistant strains could be an indication of the excessive use of antibiotics in aquaculture farming. The rational use of antimicrobial agents and the surveillance of antibiotic administration may reduce the acquisition of resistance by microorganisms in aquatic ecosystems.

      • Computer modeling for frequency performance of viscoelastic magneto-electro-elastic annular micro/nanosystem via adaptive tuned deep learning neural network optimization

        The presented paper is the first attempt to apply deep learning for predicting the frequency characteristics of a magneto-electro-elastic (MEE) annular nano/microdisk (MEEAD). The optimum amount of the factors participating in the mechanism of the fully connected neural network are achieved through the optimizer based on the momentum. The positive side of the mentioned approach employed in this investigation would be due to its high accuracy along with lower epochs required for training the multi-layered network. This scrutinization would be semi-computational research that estimates the vibrational behavior of a MEEAD employing a non-classical continuum model known as the modified couple stress (MCS) model. First-order shear deformation theory (FSDT) and shell model would be provided for presenting their displacement fields. Then, Kelvin-Voight theory has been applied to model the viscoelastic foundation. Its non-classical governing equations, as well as related boundary conditions (BCs) of small-scaled MEEAD, would be achieved by considering the symmetric spinning gradient along with higher-order stress tensors for the strain energy. The provided non-classical theory would be able to capture the small scale in the MEEAD employing just one length scale of a material factor, then, the mathematical modeling of MEEAD according to the classical theory would be able to be recovered from the provided model by eliminating the material length scale factor. Ultimately, the non-classical governing equations would be solved by applying the generalized differential quadrature (GDQ) approach for multifarious BCs. Moreover, parametric research has been conducted to analyze the influences of the viscoelastic foundation, length scale factor, geometry of MEE, radial and circumferential mode number, radius ratio, and BCs on the frequency behavior of the MEEAD by applying MCST. The outcomes reveal that there would be a crucial radius ratio in which the relationship between these elements and crucial inserted voltage alters from direct to indirect relation.
