      • A systemic assessment of QA-based food safety management system of Chinese edible oil manufacture in view of context characteristics

        Purpose - This study aims to have a systematic assessment of Chinese edible oil company, including: 1) evaluating the performance of the Food Safety Management System (FSMS) of a Chinese edible oil company; 2) assessing the FSMS performance in view of the context features of Chinese food sectors. Design/methodology/approach - This study implies a structured interview with Quality Assurance (QA) manager of one edible oil company to find the fundamental information about the companies. Then a tool used to diagnose the performance of FSMS is conducted to diagnose the levels of the core and assurance activities, as well as the context factors and output of FSMS. Finding - Our study finds that the edible oil company is operating in a moderate to low-risk context. Following the legislation, they imply QA-based FSMS. The results turn out to be that the control activities are operating at advanced level, while the assurance activities are at average level. Originality/value - This project will give insight in how context factors impact the set up and operation of FSMS and will help Food safety regulation, supporting (branch of industry) organizations, and Food company to gain more effective and efficient FSMS.

        Expression of recombinant anti‐colorectal cancer large single‐chain monoclonal antibody in insect cells

        Colorectal cancer is the third most commonly diagnosed cancer in the world. Monoclonal antibody (mAb) CO17‐1A recognizes the tumor‐associated antigen GA733‐2, a cell surface glycoprotein highly expressed in colorectal carcinoma cells which is applicable for preventing and curing colorectal cancer. In this study, we tried to produce a new recombinant anti‐colorectal cancer large single chain (lsc) mAb based on mAb CO17‐1A in the baculovirus‐insect cell protein expression system. Two kinds of recombinant lsc mAbs were generated where variable light chain (VL) and heavy chain (HC) of mAb CO17‐1A were fused together by an interchain linker. The only difference between the two mAbs is based on fusion of an ER retention signal (KDEL) at its C‐terminus of HC. Polymerase chain reaction analysis verified the presence of both recombinant genes in the bacmid for generating viral expression vectors in insect cells. Western blot confirmed the expression of lsc mAbs in baculovirus‐infected insect cells. Cell enzyme linked immunosorbent assay (ELISA) showed that the mAbs from cell lysates bound to SW480 and SW620 human colorectal cancer cells. These results indicate that the baculovirus insect expression system can produce anti‐colorectal lsc mAb recognizing human colorectal cancer cells.

        대안모형을 이용한 B2B 전환비용에 대한 실증연구 -한국의 중국 수출 기업 사례-

        본 연구는 중국 수출 기업의 B2B 관계마케팅 전략에 따른 전환비용의 효과를 기업 성과, 만족, 가치, 그리고 신뢰 측면에서 고찰하였다. 특히, 전환비용에 대한 만족, 가치, 신뢰, 기업성과 간의 이론적 차이를 대안모형을 통해 최적모형 선택에 초점을 둔다. 총 329명의 B2B 관계교환에 참여하는 한국 수출 기업을 중심으로, 본 연구의 전반적인 결과는 다음과 같다. 첫째, 대안모형 선택과정을 통해 전환비용 연구의 이론적 틀을 강화시키고, 시사점을 제시하였다. 둘째, 연구방법론 측면에서는 기존 선행연구들이 주로 구조방정식을 이용한 모형검증에 초점을 둔 반면, 본 연구에서는 구조방정식의 핵심인 대안모형을 제시하고, 실증분석을 실행한 후, 연구결과에 따라 최적모형을 선택하였다. 즉, 대안모형을 통해 중국 수출 기업의 관계마케팅 활동을 전환비용 측면에서 최적화시킬 수 있는 대안모형이 무엇인가에 대해 선택했으며, 구조방정식을 이용한 주요 구성개념의 이론적 관계를 고찰하였다. 이에 기반으로 방법론적 측면에서도 연구 시사점을 강화하였다. This study investigates the structural roles of satisfaction, value, trust, and firm performance on switching costs, particularly in the context of Korean exporters to China. Specially, our study focuses on the selection of the optimal model using alternative models such as direct, performance, trust-oriented, and research models. We use a total of 329 responses to analyse alternative models. First, the findings show that the research model is selected as the optimal model for supporting theoretical frameworks and prior studies. Second, research has mainly focused on the traditional SEM approach to investigate model robustness, whereas the current study focuses on the possibility of alternative models due to the availability of relationships among constructs. In line with these observations, we provide insights for selecting the best model when Korean firms need to strengthen their competitiveness through relationship exchanges with Chinese partners. By adding new aspects to the discussion about how to establish alternative ways, finally, our results speak to both theorists and managers.

      • Expression of GA733-Fc fusion protein as a vaccine candidate for colorectal cancer in transgenic plants

        Colorectal cancer is the third leading cause of cancer-related death in the Korea. It is eager to find a method for preventing and curing colorectal cancer. The tumor-associated antigen GA733 is a cell surface glycoprotein highly expressed in colorectal carcinoma. In this study, GA733 was fused to Fc fragment of human IgG as a vaccine candidate in plant expression system. The fusion protein GA733-Fc can improve their expression in plant cells and induce cellular and humoral immune responses. GA733-Fc is glycosylated, however the N-glycan structures of plant-derived GA733-Fc differ from their parental counterparts. Plant-derived specific N-glycoforms have the potential to induce immunogenicity and allergenic responses in human therapy. Thus, oligomannose glycan structure was tried to be generated through retaining GA733-Fc and GA733 in ER with KDEL sequence. Agrobacterium-mediated transformation was conducted to generate transgenic plant expressing GA733-Fc and GA733. Recombinant protein expression, accumulation and biological activity of GA733-Fc with KDEL (GA733-FcK), GA733-Fc without KDEL (GA733-Fc), and GA733 with KDEL (GA733K) in transgenic plants were compared by western blot, confocal immunofluorescence analysis, ELISA and mouse immunization, respectively.

        In the paper, a large customer satisfaction evaluation index system is constructed, which involves five broad categories including corporate image, service attitude service quality, and 21 individual indexes, for example popularity, Value for money and so on. By using fuzzy theory, the fuzzy evaluation model of large customer satisfaction is been established. And then, based on a questionnaire survey for the main districts and counties in Daqing, the paper completes large customer satisfaction evaluation of the China Netcom Corporation Daqing branch and gives the advantages and disadvantages in the customer service, in order to further improving the large customer satisfaction.

        Trust is risky. If the two sides is not enough high trust, or is asymmetirc in trust each other, it is easy totrigger a trust crisis, and which bring risks to the supply chain and its members. The guarantee mechanism of trust can manage and control trust risk. The paper firstly introduces the risk under the supply chain environment, analyses the risk of trust. Then, it analyzes the trust insurance mechanism based on the system constraints by using the game theory, as well as the insufficiency of system restraints.

        Expression of Anti-breast Cancer Monoclonal Antibody in Transgenic Plant

        BACKGROUND: Plant expression system for mass production of recombinant proteins has several advantages over other existing expression systems with economical and safety issues. Breast cancer is a cancer originating from breast tissue and comprises almost 25% of all cancers in women world widely. Lewis-Y antigen is difucosylated oligosaccharide and is carried by glycoconjugates at cancer cell surface. In this study, the anti-breast cancer mAb BR55, which recognizes the epitope Lewis-Y, was expressed in the plant expression system. METHODS AND RESULTS: We have developed plant system for production of mAb BR55 with or without KDEL (the ER retention signal). This ER retention signal was attached to C-terminus of protein to help retain the recombinant glycoprotein carrying oligomannose glycans and enhance glycoprotein accumulation. PCR analysis was performed and confirmed the presence of recombinant genes. Western blot validated that the recombinant proteins mAb BR55 with or without KDEL were expressed in transgenic plants, moreover, the expression level of the mAb BR55 with KDEL was higher compared to the mAb BR55 without KDEL. CONCLUSION: These results indicate that KDEL fusion is a good way to produce proteins and plant can be an ideal expression system to obtain proteins and enhance accumulation of proteins.
