Background: We investigated whether curcumin and genistein affect the MUC5AC mucin production from human airway epithelial cells that is induced by phorbol 12-myristate 13-acetate (PMA) or tumor necrosis factor-α (TNF-α). Methods: Confluent NCI-H292 cells were pretreated with each agent for 30 min and then stimulated with PMA or TNF-α for 24 hours. MUC5AC mucin production was measured by an ELISA. Results: (1) Curcumin dose-dependently inhibited the production of MUC5AC mucin that was induced by PMA or TNF-α; (2) Genistein inhibited PMA-induced MUC5AC mucin production. However, it did not decrease TNF-α-induced MUC5AC mucin production. Conclusion: These results suggest that curcumin and genistein inhibit the production of airway mucin induced by PMA.

We studied a efficient traffic signal system using RFID. This system consisted of 4-line main highway and 2-line farm load and designed for intelligent traffic signal system, which could make the traffic flow easily at a intersection of 4-line and 2-line loads. If the traffic is heavy on the main highway, the signal light of the highway continues with a green light for easy traffic flow. If the traffic is increased on the farm load, the signal of the highway will be changed with a red light for easily traffic flow of the farm load. In order to confirm a existence of the traffic on the farm load, RPID tag was installed in the car and RPID reader was installed in the traffic signal. following instructions give you basic guidelines for preparing camera-ready papers for the Journal of the KOSME. Methods: Confluent NCI-H292 cells were pretreated with each agent (carbenoxolone, prunetin, and silibinin) for 30 min and then stimulated with TNF-${\alpha}$ for 24 hours. The MUC5AC mucin gene expression and mucin protein production were measured by reverse transcription-polymerase chain reaction and enzyme linked immunosorbent assay, respectively. Results: Carbenoxolone, prunetin and silibinin inhibited the production of MUC5AC mucin protein induced by TNF-${\alpha}$; the 3 compounds also inhibited the expression of MUC5AC mucin gene induced by TNF-${\alpha}$. Conclusion: This result suggests that carbenoxolone, prunetin and silibinin can inhibit mucin gene expression and production of mucin protein induced by TNF-${\alpha}$, by directly acting on airway epithelial cells.

We investigated whether three flavonoids including daidzein, baicalein, hesperidin and ursolic acid, a triterpene acid, affect proliferation of MCF-7 human breast cancer cells stimulated by estrogenic compounds. Ursolic acid and baicalein inhibited proliferation of MCF-7 cells induced by PhIP, a food-derived carcinogen with estrogenic activity. Daidzein and hesperidin inhibited estradiol-induced proliferation of MCF-7 cells. These compounds should be further investigated for the possible involvement in signaling pathway after estrogen receptor binding in breast cancer cells.

<P>Introduction The BRAF V600E mutation is the most common genetic event in papillary thyroid carcinoma (PTC). The BRAF V600E mutational status has a significant diagnostic and prognostic role in PTC since it can be detected in 32%-87% of PTC by various molecular methods. Aim(s) A novel, fully automated real-time PCR-based Idylla test is assessed to detect the BRAF mutation in formalin-fixed paraffin-embedded (FFPE) thyroid samples. Methods 99 PTC and 11 nodular hyperplasia FFPE thyroid tissues are evaluated for the BRAF V600E mutation by the Idylla tests and compared with peptide nucleic acid-clamping PCR, real-time PCR and pyrosequencing. Results The sensitivity and specificity of the Idylla test to detect BRAF V600E are 98.8% and 100%, which is superior to real-time PCR and pyrosequencing. The concordance between Idylla and true positive is highest at 0.974. Conclusions This study validates that the Idylla test is a sensitive and specific method to detect BRAF V600E in FFPE thyroid tissues. A simple, quick and easy to handle Idylla test is a useful and reliable molecular technique to evaluate BRAF mutations.</P>

We conducted this study to investigate whether berberine significantly affects MUC5AC mucin gene expression and mucin production induced by epidermal growth factor (EGF), phorbol 12-myristate 13-acetate (PMA) or tumor necrosis factor-α (TNF-α) from human airway epithelial cells. Confluent NCI-H292 cells were pretreated with varying concentrations of berberine for 30 min and then stimulated with EGF, PMA or TNF-α for 24 h. MUC5AC mucin gene expression and mucin production were measured by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA), respectively. Berberine was found to inhibit the expression of MUC5AC mucin gene induced by EGF, PMA or TNF-α. Berberine also inhibited the production of MUC5AC mucin protein stimulated by the same inducers. This result suggests that berberine can regulate the expression of mucin gene and production of mucin protein, by directly acting on human airway epithelial cells.

Particulate matter (PM) has various systemic effects, such as respiratory, cardiovascular, endocrine, as well as having effects on the nervous systems. So far, there have been many epidemiologic studies, but studies related to the biological mechanisms are insufficient. We researched the effects of PM on lung epithelial cells with Next Generation Sequencing (NGS) and validated this with quantitative real-time polymerase chain reaction (qRT-PCR). We cultured the group treated with PM10 at a concentration of 50μg/mL and the untreated group for seven days in five lung cell lines: NCI-H358, HCC-827, A549, NCI-H292, BEAS-2B. Then, we extracted the RNA from the sample and performed NGS. As a result of NGS, various gene expressions were upregulated or downregulated. Among them, we selected the gene whose mean fold change was more than doubled and changed in the same direction in all five cell lines. Based on these genes, we selected the top 10 genes, either upregulated or downregulated, to validate with the qRT-PCR. There were the four genes that matched the NGS and qRT-PCR Results, all of which were upregulated genes(Table 1). The four genes are CYP1A1, CYP1B1, LINC01816, and BPIFA2. All four genes that matched the two Results were up-regulated genes and none of the down-regulated genes matched. CYP1A1 and CYP1B1 are known to cause lung cancer by metabolizing polycyclic aromatic hydrocarbons, and long non-coding RNA is also known to play an important role in lung cancer. Considering this, we thought that PM10 might be associated with lung cancer by activating CYP1A1, CYP1B1, and LINC01816.
