Myosin light chain 1 (MLC‐1) protein acts in the organization, dynamics and transport processes associated with the cytoskeleton. In this work, an MLC‐1 gene was cloned and characterized from the Indian meal moth, Plodia interpunctella (Lepidoptera: Pyralidae). The isolated PiMLC‐1 cDNA is 913 bp, including a 5′‐untranslated region (UTR) of 79 bp, 3′‐UTR of 381 bp and an open reading frame (ORF) of 453 bp encoding a polypeptide of 150 amino acids, which contains two calcium binding domains (EF‐hands). The deduced PiMLC‐1 protein sequence has 39–94% comparison with other individuals. The qPCR analysis revealed that PiMLC‐1 was expressed in the four developmental stages (egg, larva, pupa and adult) and in all tissues tested, suggesting that it plays an important role in development of P. interpunctella. Based on the MLC‐1 amino acids, phylogenetic analysis showed a similar topology with the traditional classification, suggesting the potential value of the MLC‐1 protein in phylogenetic inference.

Naphthoquinone pigments were extracted from purple gromwell (Lithospermum erythrorhizon) via ultrasoundenhanced subcritical water extraction (USWE). USWE was compared with supercritical CO2 fluid extraction (SCE) and Soxhlet extraction (SE) on the extraction yield of naphthoquinone pigments and the components and various activities of the extracts. The results show that the extraction yield of naphthoquinone pigments by USWE was the highest, up to 0.31%, while the yields achieved by SE and SCE were 0.21 and 0.19%, respectively. The 6major compounds were detected in these 3 extract samples but the USWE sample contained the fewest impurities. The extracts obtained with USWE showed the best inhibitory effects on Escherichia coli and Staphylococcus aureus, followed by that with SE. However, the extracts obtained with SE showed the highest DPPH scavenging capacity and reducing action.

The epidermal growth factor receptor (EGFR) is an important surface receptor with N-glycans in its extracellular domain, whose glycosylation is essential for its function, especially in tumor cells. Here, we demonstrated that polylactosamine is markedly increased in H7721 hepatocellular carcinoma cells after treatment with EGF, while it apparently declined after exposure to all-trans retinoic acid (ATRA). In the study of the enzymatic mechanism of this phenomenon, we explored changes in the expression of poly-N-acetyllactosamine (PLN) branching glycosyltransferases using RT-PCR. Among the four glycosyltransferases with altered expression, GnT-V was most elevated by EGF, while GnT-V and B3GNT2 were most declined by ATRA. Next, we conducted co-immunoprecipitation experiments to test whether B3GNT2 and EGFR associate with each other. We observed that EGFR is a B3GNT2-targeting protein in H7721 cells. Taken together, these findings indicated that the altered expression of B3GNT2 will remodel the PLN stucture of EGFR in H7721 cells, which may modify downstream signal transduction.

An efficient and selective method for the synthesis of ethyl 2-amino/aryloxy-3-aryl-4-oxo-5-phenyl-4,5- dihydro-3H-pyrrolo[3,2-d]pyrimidine-7-carboxylate derivatives has been developed. The main process involved the reaction of diethyl 1-phenyl-3-((triphenylphosphoranylidene)amino)-1H-pyrrole-2,4-dicarboxylate and aromatic isocyanates, followed by addition of amines/phenols in the presence of catalytic amount of sodium ethoxide or solid potassium carbonate.
Influence of Dietary Polyphenols on Platelet Aggregation
Ping Chen,Pei Qiu,Hao Chen 한국차학회 2015 한국차학회지 Vol.- No.S
Dietary polyphenols has been recognized in recent years as beneficial nutrients in cardiovascular disease prevention, especially for the role against athero-thrombogenesis. Various polyphenols including catechins and resveratrols exhibit an influence on platelet aggregation, which is part of the sequence leading to the formation of thrombus. However, polyphenols from various plant resources with different structures showed inconsistent effect in modulating platelet aggregation. In this review, we investigated the antiplatelet effect of different dietary polyphenols and explored its structure-activity relationship. Various studies related to antiplatelet polyphenols were well discussed with their molecular structure and configuration, thus clarified a comprehensive understanding of the mechanism involved in antithrombotic properties, and provided a theoretical basis for the use of natural ingredients in therapeutic intervention.

Fifty-four biological characters were used for quantitative classification of sorghum and sudangrass and their relationship. Discriminant analysis of all 240 individuals of 48 accessions suggested that 100% of originally grouped cases were correctly classified. Analysis of variance results showed that the differences among 48 accessions in characters were significant at the 0.01 level. Cluster analysis showed that the 48 accessions were obviously divided into two clusters by a Euclidean distance of 17.46 as the classification standards, i.e. the genus maize and genus sorghum. The 48 accessions were divided into four clusters by a Euclidean distance of 9.86, i.e. Sorghum-Sudangrass, Sorghum related species (S. propinquum and S. halepense), Zea mexicana and Z. mays. All accessions were divided into five clusters by a Euclidean distance of 8.91, namely group Ⅰ (Sudangrass-Chinese sorghum), group Ⅱ (Non-Chinese sorghum), group Ⅲ (Sorghum related species), group Ⅳ (Zea mexicana), and group Ⅴ (Z mays).

<P>Phosphorene, also known as single- or few-layer black phosphorus (FLBP), is a new member of the two-dimensional (2D) material family and has attracted significant attention in recent years for applications in optoelectronics, energy storage and biomedicine due to its unique physicochemical properties and excellent biocompatibility. FLBP is regarded as a potential biological imaging agent for cancer diagnosis due to its intrinsic fluorescence (FL) and photoacoustic (PA) properties and negligible cytotoxicity. FLBP-based photothermal and photodynamic therapies have emerged with excellent anti-tumour therapeutic efficacies due to their unique physical properties, such as near-infrared (NIR) optical absorbance, large extinction coefficients, biodegradability and reactive oxygen species (ROS) or heat generation upon light irradiation. Furthermore, FLBP is a promising drug delivery platform because of its high drug-loading capacity due to its puckered layer structure with an ultralarge surface area, and FLBP is size-controllable with facile surface chemical modification. Because of the marked advantages of FLBP nanomaterials in biomedical applications, an overview of the latest progress and paradigms of FLBP-based nanoplatforms for multidisciplinary biomedical applications is presented in this tutorial review.</P>

Background: MiRNAs belong to non-coding RNAs that are involved in cancer development. Acting as a mediator, they could regulate the expression level of numerous gens. However, the expression and function of miR-1299 in gastric cancer (GC) are not clear. Objective: To explore the role of miR-1299 in the process of GC. Methods: We detected the levels of miR-1299 in clinical samples of GC and investigated the role of miR-1299 in the regulation of the GC cells proliferation, apoptosis and metastasis. Luciferase reporter assay was employed to verify the target of miR-1299. Additionally, the proliferation, apoptosis and metastasis of AGS and SGC7901 cells were analyzed after the overexpression of miR-1299. Results: Our study showed the expression of miR-1299 was decreased in GC tissues and cell lines. It indicated that the cell proliferation, migration and invasion was inhibited, while the cell apoptosis was promoted by the over-expressed miR-1299. Also, we found that miR-1299 could directly target the 3'-untranslated region (3'UTR) of ARF6 genes. In addition, rescue assay demonstrated that miR-1299 overexpression promoted the cell apoptosis and inhibited cell growth, which could be attenuated by the overexpression of ARF6. Conclusions: These findings indicate that miR-1299 regulates cell progression in GC by targeting ARF6 genes, and suggest that miR-1299 may be a tumor suppressor in the GC progression.
