        Determination of Optimal Storage Condition for Pre-packed Hanwoo Loin

        The aim of this study was to determine the optimal storage condition of pre-packed Hanwoo beef without freezing. Hanwoo loin was purchased from a local distributor at 48 h after slaughter, then sliced in 1.5±0.5 cm thickness, and packed in a polyethylene (PE) tray covered with linear low-density polyethylene (LLDPE) film. The studied factors to set the optimal storage condition were chamber temperature (5, 2.5 and -1oC for 14 d), cooling method (direct and indirect cooling system), and ultraviolet (UV) light irradiation for beef surface sterilization (0, 30, 60, and 120 min). The changes of pH, thiobarbituric acid reactive substances (TBARS) and number of aerobic bacteria were measured during storage. Beef samples stored in -1oC showed the minimal increasing rate in TBARS and microbial growth. After 15 d of storage, there was no significant difference in pH and TBARS values. However, the microbial population of beef stored in direct type cooling chamber (4.25±0.66 Log CFU/g) was significantly lower than that of beef stored in indirect type chamber (6.47±0.08 Log CFU/g) (p<0.05).

        Cross-Tolerance and Responses of Antioxidative Enzymes of Rice to Various Environmental Stresse

        In order to examine the cross-tolerance of two chilling-tolerant cultivars (Donganbyeo and Heukhyangbyeo) and two chilling-susceptible cultivars (Hyangmibyeo and Taekbaekbyeo) to salt, paraquat, and drought, changes of physiological response and antioxidant enzymes were investigated. The seedlings were grown in a growth chamber until the 4-leaf stage. The seedlings were exposed to chilling at $5^{\circ}C$ for 3 days. For drought treatment, the seedlings were subjected to drought by withholding water from plants for 5 days. For paraquat study, plants were sprayed with $300{\mu}M$ paraquat. For the salt stress, the seedlings were transferred to the Hoagland's nutrient solution containing 0.6% (w/v) NaCl for 4 days. Chilling-tolerant cultivars showed cross-tolerant to other stresses, salt, paraquat, and drought in physiological parameters, such as leaf injury, chlorophyll a fluorescence, and lipid peroxidation. The baseline levels of antioxidative enzyme activities, catalase (CAT) and peroxidase (POX) activities in chilling-tolerant cultivars were higher than in the chilling-susceptible cultivars. However, there were no differences in ascorbate peroxidase (APX) and glutathione reductase (GR) activities between chilling-tolerant and -susceptible cultivars in untreated control. CAT activity in chilling-tolerant cultivars was higher than that in chilling-susceptible cultivars during chilling, salt, and drought treatments, but not during paraquat treatment. However, other antioxidative enzymes, APX, POX, and GR activities showed no significant differences between chilling-tolerant and -susceptible cultivars during chilling, salt, paraquat, and drought treatments. Thus, it was assumed that CAT contribute to cross-tolerance mechanism of chilling, salt, and drought in rice plants.

        Mechanisms of Chilling Tolerance in Relation to Antioxidative Enzymes in Rice

        In order to examine the mechanistic basis for differential sensitivities to chilling and subsequent recovery between two rice (Oryza sativa L.) cutivars, a chilling-tolerant japonica type (Ilpumbyeo) and a chilling-susceptible indica type (Taebaekbyeo), changes of physiological responses and antioxidant enzymes were investigated. Both cultivars at 3 leaf stage were exposed at a low temperature of $5^{\circ}C$ for 3 days and subsequently recovered in a growth chamber at a $25^{\circ}C$ for 5 days with 250 mmol $m^{-2}$ $s^{-1}$. Physiological parameters such as leaf fresh weight, relative water content, cellular leakage, lipid peroxidation, and chlorophyll a fluorescence showed that the chilling tolerant cultivar had a high tolerance during chilling. However, the chilling-susceptible cultivar revealed severe chilling damages. The chilling-tolerant cultivar was also faster in recovery than the chilling-susceptible cultivar in all parameters examined. We analyzed the activity and isozyme profiles of four antioxidant enzymes which are: superoxide dismutase (SOD), caltalase (CAT), ascorbate peroxidase (APX), and glutation reductase (GR). We observed that chilling-tolerance was due to a result of the induced or higher antioxidant enzyme system, CAT and APX in leaves and SOD, CAT, APX, and GR in roots. Especially, we observed the most significant differences between the chilling-tolerant cultivar and -susceptible cultivar in CAT and APX activity. Also in isozyme profiles, CAT and APX band intensity in the chilling-tolerant cultivar was distinctively higher than in the chilling-susceptible cultivars during chilling and recovery. Thus, the cold stability of CAT and APX are expected to contribute to a tolerance mechanism of chilling in rice plants. In addition, the antioxidative enzymes activity in roots may be more important than in that of leaves to protect chilling damage on rice plants.

However, TBARS values of 60 or 120 min UV light irradiated beef samples were significantly higher than non-irradiated beef sample after 4 d of storage (p<0.05).

        Growth inhibition by metformin in YD-38 oral cancer cells derived from Korean

        Metformin (1,1-dimethylbiguanide hydrochloride), derived from French lilac (Galega officinalis), is a first-line drug prescribed for patients with type 2 diabetes. It has been reported to have anti-cancer effects in a variety of cancer cells. However, effects of metformin on oral cancer cells have not been clearly established. The main goal of this study was to investigate the effect of metformin on cell growth and apoptosis induction in oral cancer cells derived from Korean patients. The effect of metformin on cell growth and apoptosis induction in oral cancer cells was examined by inhibition of cell growth, DNA fragmentation analysis and immunoblotting in YD-38 human oral cancer cells derived from Korean patients. Treatment with metformin induced inhibition of cell growth depending on the metformin treatment time and concentration in YD-38 human oral cancer cells. Treatment with metformin induced nuclear fragmentation in YD-38 human oral cancer cells. Metformin promoted proteolytic cleavage of procaspase-3 with an increase in the amount of cleaved caspase-3. Cleaved PARP was increased by metformin in YD-38 human oral cancer cells. Treatment of YD-38 human oral cancer cells with metformin increased the level of Bax, but it decreased the level of Bcl-2. These results suggest that metformin can induce suppression of cell growth and cell apoptosis in YD-38 human oral cancer cells derived from Korean patients.

        마이크로웨이브 폴리올 공정에서 금속 도핑 산화아연 나노클러스터의 합성

        금속이 도핑 된 산화아연 나노클러스터를 합성하기 위해 마이크로웨이브를 이용한 폴리올 공정은 빠르고 경제적인 합성 방법이다. 디에틸렌글리콜은 높은 분극률과 마이크로파의 흡수 능력이 뛰어나며, 높은 온도상승 비율과 반응시간을 짧게 해준다. 본 연구에서는 금속이 도핑 된 산화아연 나노클러스터를 합성하기 위해서 첨가되는 seed의 부피비를 다르게 하여 얻었으며, 전구체로는 아세트산 아연 2수화물, 도핑 금속은 아세트산 금속 염을 그리고 용매로서 디에틸렌글리콜을 사용하였다. 금속이 도핑된 산화아연 클러스터는 FE-SEM, XRD, Raman, PSA로 특성을 확인하였다. ZnO has attracted much attention such as photocatalysts, sensors, piezoelectricity and etc. At present, an economical and rapid synthesis route based on the efficient microwave polyol process is used to synthesized metal-doped ZnO nanoclusters. Diethylene glycol has a property of high polarizability, and is an excellent microwave absorbing agent, thus leading to a high heating rate and a significantly shorter reaction time. In this study, metal-doped ZnO nanoclusters are obtained with different seed volumes, when zinc acetate dihydrate is used as a precursor, and metal acetate hydrate is used as a doped-metal and diethylene glycol is used as a solvent. The obtained metal-doped ZnO nanoclusters were characterized by FE-SEM, XRD, Raman and PSA.

        In the electric railway systems, it is very important that we should design the system configuration, location and power capacity of substation. This paper presents the results of system configuration and system design of the DC traction power supply for the test line of Light Rail Vehicle. The voltage fluctuation of train and the power capacity of substation are calculated by computer simulation using the nodal equation, K.C.L/K.V.L, Ohm"s law and superposition theory.

        국내에서 분리한 Ornithobacterium rhinotracheale 균의 종란과 육계에서의 병원성

        본 시험은 국내에서 분리한 Ornithobacterium rhinotracheale(OR) 병원성 세균이 부화중인 종란과 일반 육계에서 얼마만큼 병원성을 보이는지 알아보기 위하여 수행하였다. 첫 번째 9일령 부화란의 yolk sac에 국내에서 분리한 3종의 OR strain을 접종하여 12일 동안 관찰한 결과, 66% 이상의 폐사율이 관찰되어 높은 병원성이 인정되었다. 두 번째로 3주령 일반 육계를 대상으로 5가지 다른 공격접종법[Intratracheal, Intravenous, Intramuscular, Aerosol, Newcastle Disease Virus(NDV)와 혼합 Aerosol]으로 접종한 다음 병원성을 관찰하였다. NDV와 OR균을 동시에 분무 접종한 계군에서만 특이적인 임상증상인 침울, 기침, 안면 종대와 함께 부검시 치즈양 또는 요구르트와 비슷한 염증성 삼출물이 기낭에 관찰되었다. 조직학적으로도 기낭상피세포의 변성과 탈락, 대식세포와 다형태성 관립구의 침윤, 부종 등의 기낭염이 확인되었으며, 이들 기낭을 이용한 면역 조직 화학적 염색법을 적용한 결과 다량의 OR균의 항원이 검출되었다. 그러나 OR균만 단독 처리한 닭에서는 일시적이고 경미한 조직학적 병변만이 관찰되었다. 결론적으로 NDV가 OR균의 감염에 따른 임상 증상과 병리조직학적 병변 유발에 주요한 역할을 하는 것으로 판단된다. Field strains of Ornithobacterium rhinotracheale (OR) were tested on their virulence in specific pathogen free (SPF) embryonated chicken eggs and 3-week-old broilers. When infected with three different OR isolates (OR-161, OR-240 and OR-295) through yolk sac infection route, all strains appeared to be highly pathogenic with responsible mortality 66% and 100% within 12 days post infection (DPI). To test the virulence of OR in the commercial broilers, 3 week-old broilers were grouped depends on the inoculation route of OR isolate (OR-295) through five different infection routes; group 1 (IT: intratracheal), group 2 (IM: intramuscular), group 3 (Ⅳ: intravenous), group 4 (aerosol) and group 5 [Mixed: NDV (LaSota)+OR aerosol]. Within 5 to 7 days after inoculation, only broilers given NDV+OR were slightly depressed and coughing, and had mild facial redness. Grossly, foamy and yellow-white yogurt like exudate in the air sacs, predominantly in the abdominal air sacs was present. In histology, infiltration of the air sac epithelium and lamina propria by macrophage and polymorphonuclear granulocytes was seen with cell debris and inflammatory cells, correlated with the presence of OR antigen, as demonstrated by immunohistochemistry. Field strains of OR were able to induce high mortality in the embryonated chicken eggs, whereas broilers were less susceptible to OR infection. Interestingly, in the absence of NDV infection, the four groups of OR single infection only different route showed minimal and temporary microscopic air sac lesions. Thus, Newcastle disease virus (LaSota strain) showed triggering effects on the OR infection in chickens.
