        Genetics and Molecular Biology of Alzheimer's Disease : Implication for Diagnosis and Treatment

        Amyloid is deposited at characteristic locations in brains from individuals with Alzheimer`s disease (AD), older individuals with Down`s Syndrome (DS), and aged mammals including rats. These deposits (tangles, plaques, vascular amyloid) contain amyloid A4 polypeptide (Mr 4.500) derived from a larger glycosylated membrane protein resembling a cell-surface receptor (Kang et al., Nature 325, 733, 1987). The gene for this A4 precursor is located on chromosome 21 at the border between bands q21 and q22.1 (in collaboration with B. Zabel, Univ Mainz) and much closer to the 21q22 Down`s phenotype region than previously reported (Tanzi et al., Science 235, 880, 1987). We detected an intragenic, restriction fragment length polymorphism for the A4 gene with 2 alleles which proved to be useful for tracking the inheritance of the A4 gene through pedigrees with familial Alzheimer`s disease (Broeckhoven et al., Nature 329, 153, 1987). Expression of the A4 gene is seen in the brain, heart, muscle, kidney, adrenal gland, lung, spleen, thymus, testes, pancreas but not in liver. Rat A4 mRNA is widely and abundantly expressed in neurons (Shivers et al. Abstracts of the Society for Neuroscience 13, 819, 1987). High expression was seen in cerebral cortex, hippocampus, olfactory areas, amygdala and cerebellum. Since most but not all of these brain regions represent sites of amyloid deposition in AD and DS brains, we conclude that high expression of A4 precursor is not the sole cause of amyloid formation and other factors common to amyloid-forming regions initiate amyloid generation. The location of the A4 amyloid polypeptide in the precursor sequence allows to predict the putative nature of these factors. Using immunocytochemistry the A4 glycoprotein precursor was detected in rat brain section as patches on or near the plasma membrane of cell bodies and their proximal dendrites (Shivers et al. Abstracts of the Society for Neuroscience 13, 819, 1987). These data suggest a role for A4 precursor in cell contact. The location of the A4 precursor in the plasma membrane renders the protein vulnerable to extracellular proteases whose attack could be eventuated by free-radical injury and lipid peroxidation to neuronal membranes in AD and aged DS. If in longer-lived mammals the mechanisms protecting against free-radical injury fail, brain areas where plasticity is thought to underlie cognitive functions may become primary sites for amyloid deposition, and account for amyloid formation in some aged mammalian brains. Breakdown of amyloid precursor resulting in amyloid formation at clinical target sites may be the link between neuronal dysfunction and AD pathology whereby amyloid precursor and its breakdown products become putative diagnostic markers. Prevention of amyloid formation by interference with the various putative breakdown processes is of therapeutic interest and can now be studied using anti-oxidants, protease-inhibitors and agents that maintain functional cell contacts.

        Crawler robot kinematics modeling by using a two-wheeled approach

        The aim of this paper is to analyze the kinematics of a small crawler robot. A mathematical model of kinematics based on a twowheeled approach is proposed. This model is experimentally verified using vision-based motion measurements of a crawler vehicle equipped with encoders and a remote control system. It is assumed that the vehicle moves along a few curves with different angular speeds of its wheels. Based on the model of motion and the values of these speeds, numerical simulations are investigated. The results obtained from numerical and experimental validation are presented and discussed. The comparison delivered some important conclusions.

        Pragmatic Strengthening is not Strong Enough: Meanings of Sequential Closed-Class Forms

        This study focuses on the semantics of word patterns and schematic constructions. Examples of constructions with purportedly rich meanings are shown to convey readings less complex than is claimed in the literature. Many previous studies stressed these constructions’ idiosyncratic properties which have been held up as arguments in favor of their special construction status.pairings of (usually unique, conventionalized) form and (usually idiosyncratic) meaning. I wish to argue that although the constructions reviewed here do have clear meanings, their form and function are not as idiosyncratic, irregular, or unpredictable as they are portrayed in the literature. Indeed, the formal and functional properties of one construction analyzed here, the Incredulity Response Construction fit well within traditional characterizations of items located on the syntactic side of the lexicon-syntax continuum. I will attempt to demonstrate that the form of the construction is an iconic representation of its reading. Additionally, I question the reading itself, arguing that the incredulity that gave rise to the construction’s very name is not its semantic contribution. Instead, I propose a more general and abstract reading of incongruousness or “cognitive dissonance”. Finally, it is argued that there are no known mechanisms that could equip constructions with overly rich semantic content. One potential candidate, pragmatic strengthening, capable of endowing constructions with meanings does not go beyond fairly sparse readings already known to occur in grammatical forms.

        Detonation cell size model based on deep neural network for hydrogen, methane and propane mixtures with air and oxygen

        The aim of the present study was to develop model for detonation cell sizes prediction based on a deepartificial neural network of hydrogen, methane and propane mixtures with air and oxygen. The discussionabout the currently available algorithms compared existing solutions and resulted in a conclusionthat there is a need for a new model, free from uncertainty of the effective activation energy and thereaction length definitions. The model offers a better and more feasible alternative to the existing ones. Resulting predictions were validated against experimental data obtained during the investigation ofdetonation parameters, as well as with data collected from the literature. Additionally, separate modelsfor individual mixtures were created and compared with the main model. The comparison showed nodrawbacks caused by fitting one model to many mixtures. Moreover, it was demonstrated that the modelmay be easily extended by including more independent variables. As an example, dependency onpressure was examined. The preparation of experimental data for deep neural network training wasdescribed in detail to allow reproducing the results obtained and extending the model to differentmixtures and initial conditions. The source code of ready to use models is also provided

        Do Model Minorities Drink Wine or Beer? The Representation of Race, Elitism and American Identity

        Throughout the 2008 presidential campaign and into his presidency,President Barack Obama faced charges of elitism and questions about his patriotism and citizenship. These claims of elitism and foreignness locate Obama’s identity on familiar terrain for the Asian American experience. Obama’s biography, which is plotted through predominantly Asian locales such as Hawai‘i and Indonesia and Ivy League schools such as Columbia and Harvard, shares the elitist and exotic tropes associated with stereotypes that haunt the meaning of Asian bodies in America: Asian Americans as the model minority or as fundamentally foreign. This chapter explores how Obama’s Asian American grassroots supporters used Obama’s black-Asian intersection to organize around objectives that went beyond merely rallying around the candidate. Through YouTube, these supporters treated political engagement and cultural representation as a synonymous practice.

        The Temperament Risk Factor, Disease Severity, and Quality of Life in Patients with Psoriasis

        Background: Psoriasis exerts a profound negative impact on health-related quality of life (QoL). Although the severity of psoriasis is one potential variable that contributes to decreased QoL, previous studies have shown only weak or no association between measures of psoriasis severity and QoL. We hypothesized that this relationship is moderated by temperament factors. Objective: We aimed to verify whether the relationship between disease severity and QoL is moderated by a constellation of temperament traits (i.e., temperament risk factors) and whether this moderation takes place via cognitive- appraisal and coping processes. Methods: One hundred fifty patients with psoriasis vulgaris participated in the study. Psoriasis severity was assessed by a standardized measure, the Psoriasis Area and Severity Index (PASI), and the patients also completed a battery of psychological questionnaires assessing QoL, temperament, disease-related cognitive appraisals, and coping strategies. Results: A specific constellation of temperament traits was found to moderate the strength of the association between the PASI and QoL. This constellation of temperament traits was associated with certain disease-related cognitive appraisals (i.e., threat, obstacle/ loss, harm, profit, value) and emotion-focused coping strategies (i.e., self-blame, avoidance, resignation, seeking social support, and seeking information). Conclusion: The constellation of temperament traits is a crucial individual variable that strongly moderates the negative impact of psoriasis severity on QoL, potentially through the activation of nonadaptive cognitive appraisals and coping strategies in susceptible individuals.
