        Two New Metabolites with Cytotoxicities from Deep-Sea Fungus, Aspergillus sydowi YH11-2

        Two new compounds, 2, 3, 5-trimethyl-6-(3-oxobutan-2-yl)-4H-pyran-4-one (1) and (2R)-2, 3- dihydro-7-hydroxy-6, 8-dimethyl-2-[(E)-prop-1-enyl] chromen-4-one (2), together with six known compounds (3-8), were isolated from a deep-sea fungus, identified as Aspergillus sydowi, by a bioassay-guided method. Their structures were elucidated by spectroscopic methods and the cytotoxicities were evaluated by SRB method.

        Testing the effects of natural products on hair growth in stumptailed macauqe

        The effect of natural products(drug 1,drug 2) on hair growth of frontal scalp of ten stumptailed macaques was investigated in period of 12 months. Drug 1 conteins extracts of Angelica gigantis Radix etc. and drug 2 conteins extracts of Corni Fructus, etc. Photographs of close view of the frontal scalp were taken once every month and folliclogram analysis has been done at pretretment and on 5th, 10th and 12th month after treatment. Drug 1 showed only slight effect or no effect on hair growth and drug 2 exhibited a significant degree of hair regrowth, but the control group exhibited a definite degree of regrowth and increasing density of vellus hair. Also, the physiological parameters, such as body weight, blood pressure, heart rate, serum levels of androgens, hematological measures during the treatment of drug 1,drug 2,and vehicle, were within normal limits. It indicates the drug 2 is the possible appearence of new summit of hair growth.

        Bone Marrow Adiposity, Bone Mineral Density and Wnt/β-catenin Pathway Inhibitors Levels in Hemodialysis Patients

        Background: Marrow adipose tissue (MAT) is known to accumulate in patients with chronic kidney disease. This pilot study aimed to evaluate bone mineral density (BMD), MAT, visceral adipose tissue (VAT), and subcutaneous adipose tissue (SAT) using computed tomography (CT) scans and to explore correlations between bone parameters, circulating Wnt/β-catenin pathway inhibitor levels, and adipose tissue parameters. Methods: Single-center cross-sectional pilot study conducted in hemodialysis patients at the Centre Universitaire de Québec, Hôtel-Dieu de Québec hospital, Canada. CT-scan slices were acquired at the levels of the hip, L3 vertebra, and tibia. Volumetric and areal BMD, tibia cortical thickness, VAT and SAT area, and fat marrow index (FMI) were analyzed using the Mindways QCT Pro software. Blood levels of sclerostin, dickkopf-related protein 1 (DKK1), fibroblast growth factor 23, and α-Klotho were assessed. Spearman’s rho test was used to evaluate correlations. Results: Fifteen hemodialysis patients (median age, 75 [66–82] years; 80% male; dialysis vintage, 39.3 [27.4–71.0] months) were included. While inverse correlations were obtained between L3 FMI and BMD, positive correlations were found between proximal tibial FMI and vertebral and tibial BMD, as well as with tibial (proximal and distal) cortical thickness. VAT had a positive correlation with α-Klotho levels, whereas L3 FMI had a negative correlation with DKK1 levels. Conclusions: CT-scan allows simultaneous evaluation of bone and marrow adiposity in dialysis patients. Correlations between MAT and BMD vary depending on the bone site evaluated. DKK1 and α-Klotho levels correlate with adipose tissue accumulation in dialysis patients.

        Investigating Medical Cost and Mortality Among Psychiatric Patients Involuntary Admissions: A Nationwide Propensity Score-Matched Study

        Objective Involuntary admission to psychiatric inpatient care can protect both patients with severe mental illnesses and individuals around them. This study analyzed annual healthcare costs per person for involuntary psychiatric admission and examined categories of mental disorders and other factors associated with mortality.Methods This retrospective cohort study collected 1 million randomly sampled beneficiaries from the National Health Insurance Database for 2002–2013. It identified and matched 181 patients with involuntary psychiatric admissions (research group) with 724 patients with voluntary psychiatric admissions (control group) through 1:4 propensity-score matching for sex, age, comorbidities, mental disorder category, and index year of diagnosis.Results Mean life expectancy of patients with involuntary psychiatric admissions was 33.13 years less than the general population. Average annual healthcare costs per person for involuntary psychiatric admissions were 3.94 times higher compared with voluntary admissions. The general linear model demonstrated that average annual medical costs per person per compulsory hospitalization were 5.8 times that of voluntary hospitalization. Survival analysis using the Cox proportional hazards model found no significant association between type of psychiatric admission (involuntary or voluntary) and death.Conclusion This study revealed no significant difference in mortality between involuntary and voluntary psychiatric admissions, indicating involuntary treatment’s effectiveness.

        A Novel Hitting Frequency Point Collision Avoidance Method for Wireless Dual-Channel Networks

        In dual-channel networks (DCNs), all frequency hopping (FH) sequences used for data channels are chosen from the original FH sequence used for the control channel by shifting different initial phases. As the number of data channels increases, the hitting frequency point problem becomes considerably serious because DCNs is non-orthogonal synchronization network and FH sequences are non-orthogonal. The increasing severity of the hitting frequency point problem consequently reduces the resource utilization efficiency. To solve this problem, we propose a novel hitting frequency point collision avoidance method, which consists of a sequence-selection strategy called sliding correlation (SC) and a collision avoidance strategy called keeping silent on hitting frequency point (KSHF). SC is used to find the optimal phase-shifted FH sequence with the minimum number of hitting frequency points for a new data channel. The hitting frequency points and their locations in this optimal sequence are also derived for KSHF according to SC strategy. In KSHF, the transceivers transmit or receive symbol information not on the hitting frequency point, but on the next frequency point during the next FH period. Analytical and simulation results demonstrate that unlike the traditional method, the proposed method can effectively reduce the number of hitting frequency points and improve the efficiency of the code resource utilization.

        Introduction of Telomerase Reverse Transcriptase Gene into Epidermal Stem Cells Derived from Human Skins

        Telomerase extends the proliferation and prevent replicative senescence in most somatic cells. Whether it has similar function in human epidermal stem cells remains to be determined. In this study, the human telomerase reverse transcriptase (hTERT) cytalytic subunit was introduced into epidermal stem cells derived from human fetal skins. The expression of hTERT mRNA, protein and telomerase activity in the transduced cells was observed by real time PCR (RT-PCR) techniques, western blot analysis and telomeric repeat amplification protocol enzyme-linked immunosorbent assay (TRAP-ELISA) assay, respectively. The proliferation of the transduced cells was examined. The results showed the introduction of hTERT into the epidermal stem cells upregulated the expression of the hTERT gene and protein. And also the hTERT-transduced epidermal stem cells exhibited significantly elevated telomerase activity and proliferation. These works establish the base of further gene modification, monoclone screening and cell differentiation mechanism. The ability to maintain the biological characteristics and proliferation of epidermal stem cells could have important applications in wound healing and skin tissue engineering.

        Comparative Analysis of the Genomes of Bombyx mandarina and Bombyx mori Nucleopolyhedroviruses

        The Bombyx mandarina nucleopolyhedrovirus (BomaNPV) S1 strain can infect the silkworm, Bombyx mori,but is significantly less virulent than B. mori nucleopolyhedrovirus (BmNPV) T3 strain. The complete nucleotide sequence of the S1 strain of BomaNPV was determined and compared with the BmNPV T3 strain. The circular, double stranded DNA genome of the S1 strain was 126,770 nucleotides long (GenBank accession no. FJ882854), with a G+C content of 40.23%. The genome contained 133 potential ORFs. Most of the putative proteins were more than 96% identical to homologs in the BmNPV T3 strain, except for bro-a, lef-12,bro-c, and bro-d. Compared with the BmNPV T3 strain, however, this genome did not encode the bro-b and bro-e genes. In addition, hr1 lacked two repeat units, while hr2L, hr2R, hr3, hr4L, hr4R, and hr5 were similar to the corresponding hrs in the T3 strain. The sequence strongly suggested that BomaNPV and BmNPV are variants with each other, and supported the idea that baculovirus strain heterogeneity may often be caused by variation in the hrs and bro genes.

        Attenuated Secretion of the Thermostable Xylanase xynB from Pichia pastoris Using Synthesized Sequences Optimized from the Preferred Codon Usage in Yeast

        Xylanase has been used extensively in the industrial and agricultural fields. However, the low-yield production of xylanase from native species cannot meet the increasing demand of the market. Therefore, improving the heterologous expression of xylanase through basic gene optimization may help to overcome the shortage. In this study, we synthesized a high-GC-content native sequence of the thermostable xylanase gene xynB from Streptomyces olivaceoviridis A1 and, also designed a slightly AT-biased sequence with codons completely optimized to be favorable to Pichia pastoris. The comparison of the sequences` expression efficiencies in P. pastoris X33 was determined through the detection of single-copy-number integrants, which were quantified using qPCR. Surprisingly, the high GC content did not appear to be detrimental to the heterologous expression of xynB in yeast, whereas the optimized sequence, with its extremely skewed codon usage, exhibited more abundant accumulation of synthesized recombinant proteins in the yeast cell, but an approximately 30% reduction of the secretion level, deduced from the enzymatic activity assay. In this study, we developed a more accurate method for comparing the expression levels of individual yeast transformants. Moreover, our results provide a practical example for further investigation of what constitutes a rational design strategy for a heterologously expressed and secreted protein.

        The effect of chitosan (CS) coagulation bath on structure and performance of polylactic acid (PLA) microfiltration membrane

        Membrane hydrophilicity is a crucial factor in evaluating ultrafiltration processes. In this paper, chitosan(CS) was selected for the hydrophilic modification of the polylactic acid (PLA) membrane, and PLACS membranes wereprepared for the densification of the yeast solution. By non-solvent phase inversion method (NIPS), the PLACS microfiltrationmembranes were prepared by using chitosan (CS) acetic acid solution as the coagulation bath and glutaraldehydeas the crosslinking agent. PLACS membranes were characterized by water contact angle, porosity, pore sizedistribution, mechanical properties, ATR-FTIR, SEM, TGA and the ultrafiltration experiment. The viscosity of coagulationbath solution can severely influence the exchange rate of the solvent and the non-solvent as well during phaseinversion; therefore, it can regulate the precipitation kinetics and membrane morphology. The results showed that chitosan(CS) was presented as granular on the pore surfaces of the PLACS membranes. When chitosan (CS) contentincreased, gel rate became smaller and membrane forming process was prolonged; the porosity and pore size of thePLACS were increased compared to the polylactic acid (PLA) membrane, pure water flux increased from 90.31 L·m2·h1to 120.14 L·m2·h1, and yeast rejection rate increased from 75% to more than 90%.
