      • cDNA Cloning and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes-Jocheon

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA. More investigation works including gene expression, immunological analysis etc. will be carried continuously without hesitation after this presentation.

The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

      • 중부지방의 난대성 수종 기후 적응도 분석 : 세종특별자치시 수산리 배재대학교 수목실습장을 대상으로

        This study was carried out to monitor the growth of warm-temperate species that are planted in the Sejong practice field of Pai Chai university and evaluated the degree of adaptation of the species in the midlands. Trees less than two years were sampled and investigated for the analysis of the climatic characteristics of the practice field. Also, this survey identified the growth characteristics and climate adaptation of warm-temperate woody species. This general forests, unlike significance in terms of the experiment excludes the pressure of the outside within the same conditions were limited to the temporal scope only the initial growth characteristics of the short forested study period. In the future study, the continuous survival rate, investigation and analysis of the physico-chemical resistance in various aspects of soil analysis should be need to look at the trends and characteristics of the additional growth. 본 연구는 세종특별자치시 수산리 배재대학교 수목실습장에 식재한 난대성 수종의 생장량을 측정하여 중부지방의 적응정도를 파악하고자 하였다. 배재대학교 수목실습장의 기후특성 분석을 통해서 수목의 생장량과의 연관성을 파악하였다. 배재대학교 세종 수목실습장에 식재한 수목을 종별로 표본추출하여 생장량을 측정하였다. 중부내륙지방에 식재한 난대성 2년 미만의 수령에 해당하는 수종의 생장특성 및 기후적응여부를 조사하였다. 연구기간이 짧아 수목의 초기 생장특성만을 시간적 범위로 한정하였다. 수목의 적응여부를 살펴보기 위한 과정으로 생장량과 고사율만을 살펴보았다는 데서 한계점이 있어 차후 연구에서는 지속적인 예찰을 통해 추가적인 생장특성의 변화추이를 살펴보고, 토양의 이화학성 분석 등 다양한 관점에서의 조사가 이루어져야 할 것이다.

      • 大學生의 커뮤니케이션패턴과 政治行爲

        The Purpose of this research is to investigate the role of communication in the political behavior of college students in Korea. The relationships between the communication patterns of college students, and their political knowledge, political orientation and political activities were examined. 700 samples were randomly selected among the students of seven colleges; four universities in Seoul area and three in rural areas. It was found that: 1. All communication media affected the political behavior of college students. 2. The direction of the influence was mostly negative to the orientation toward the subjects. 3. Especially, the irregular communication media(Dae Ja Bo. printed matters, and rumor) strongly affected the political orientation and the political activities of college students. It is suggested that the activation of the existing media with more freedom of press are necessary to minimize the unhealthy impact of the irregular communication media upon the large number of audience.

      • 오스테나이트계 TP347 스테인레스강의 파괴저항특성 평가

        The objective of this paper is to evaluate the material properties of TP347 stainless steel and their associated welds and the welded joint of the carbon steel and the stainless steel manufactured for primary coolant system of Ulchin 3 & 4 nuclear power plants. The effect of various parameters such as pipe size, welding method, chemical composition, crack plane orientation, metallography and fractography on the material properties were discussed. Test results showed that the effect of pipe size on fracture toughness was negligible while the effect of welding method on fracture toughness was significant. In addition, the drop of fracture toughness in the field fabrication weld of TP347 stainless steel is probably due to the large amount of σ-phase precipitated on the δ-ferrite boundary and the large size dimple.

      • 산촉매 하에서 [Co(NH₃)₄(C₂O₄)]+ 와 Fe(Ⅱ)와의 산화-환원 반응에 대한 반응속도와 메타니즘

        Kinetic study was carried out for the redox reaction of [Co(NH₃)₄(C₂O₄)]+ with Fe(Ⅱ) in acidic solution by UV/vis-spectrophotometric method. It has been found that the reaction order for H+ catalyst is the first order in lower H+ concentration range than 0.5mol/ℓ, while second order in the higher one than 0.5mol/ℓ. This redox reaction system is first order with respect to each of the two substracts. Accordingly overall reaction was the third and forth order. It was found that the H+ act as the first order, the values of rate constant, ΔH? and ΔS? is 2.2079×10 ? ℓ²·mol?. sec?, 13.40Kcal/mol and -17.33e. u., and H+ act the second order, these values is 2.4490×10? ℓ³·mol?·sec?, 15.39Kcal/mol and -20.60 e. u., respectively. The result of EHMO calculation appears to be the preferred trigonal bipyramidal intermediate. On the basis of kinetic data and EHMO calculation, we suggest that the redox reaction system proceed via innersphere mechanism.

        쿨백-라이블러 정보함수 이용한 단계 스트레스 가속수명모형의 지수성 검정

        In this paper, we propose goodness of fit test statistics for exponentiality in accelerated life tests data based on Kullback-Leibler information functions. This acceleration model is assumed to be a tampered random variable model. The procedure is applicable when the exponential parameter using the data from accelerated life tests is or is not specified under null hypothesis. And we compare the power of the proposed test statistics with Kolmogorov-Smirnov, Cramer von Mises and Anderson-Darling statistics in the small sample.

        반복적 부동화 스트레스가 신경활성 Steroid의 GABAА 수용체 조절작용에 미치는 영향

        신경활성 steroid들은 중추신경계의 말초성 benzodiazepine 수용체에서 합성되고, GABAA 수용체 복합체상의 독립적인 인지부위에 작용하여, GABAA 수용체에 대하여 효현활성을 나타내며, 스트레스에 대응하여 유리되는 내인성 조절자로 시사되고 있다. 그러나 신경활성 steroid의 신경전달조절 활성을 스트레스에 대한 반응도(reactivity)와 연관시켜 연구한 것은 아직 미흡한 실정이다. 이에 본 연구에서는 Sprague-Dawley rat에 2주간의 반복적인 부동화 스트레스를 부하한 후 반복적 스트레스에 의한 신경활성 steroid 활성의 변화를 검색하고자 하였다. 그리고 스트레스에 대하여 과도한 반응성을 보일 것으로 보고된 borderline-hypertensive rat을 대상으로 신경활성 steroid의 억제성 신경전달계인 GABA성 신경전달 조절활성을 비교 관찰함으로써, 반복적 부동화 스트레스에 대한 실험동물의 반응성에 대한 연구의 일환으로 신경활성 steroid의 신경조절작용을 검색하고자 하였다. 이를 위하여 Sprague- Dawley rat의 대뇌피질세포막에 있는 GABAA-benzodiazepine 수용체 복합체에서 대하여 신경활성 steroid인 pregnanolone의 [³H]flunitrazepam 결합반응을 항진효과를 비교, 검색하였다. 본 실험결과, Sprague-Dawley rat의 대뇌피질세포막에 있는 GABAA-benzodiazepine 수용체 복합체에서 신경활성 steroid인 pregnanolone은 [³H]flunitrazepam 결합반응을 항진시킴으로써 GABAA 수용체에 대하여 효현 활성을 나타내었으며, 2주간의 반복적인 부동화 스트레스에 의해 이러한 pregnanolone의 활성은 증강되었다. 한편, 스트레스에 대하여 과도한 반응성을 보인다고 알려진 borderline-hypertensive rat의 뇌조직에서도 Sprague-Dawley rat에 비하여 항진된 GABA성 신경조절활성이 관찰되었으며, 스트레스 부하에 대해서도 유사한 반응성을 나타내었다. 이상의 결과로부터 반복적인 부동화 스트레스는 스트레스에 대한 반응성의 일환으로 신경활성 steroid인 pregnanolone의 GABA성 신경전달 항진작용을 증강시킨다고 생각되었으며, 스트레스 유발 고혈압의 질환모델로 보고된 BHR에서는 동일한 스트레스에 대하여 과도한 반응성을 나타냄으로써 스트레스 관련 말초질환에서의 중추성 병인인자의 참여 가능성을 시사해주었다. Pregnanolone[5β-pregnan-3α-ol-one(pregnanolone)] is one of neuroactive steroids that are reduced metabolites of progesterone. Pregnanolone synthesis in the neuron or glia was regulated by peripheral benzodiazepine receptor agonist, which was consistently observed to be upregulated in the acutely stressed animals and humans. Pregnanolone is a potent positive modulator of the γ-aminobutyric acid(GABA) response that enhance the binding of [³H] flunitrazepam to the GABAA receptor. Recently, it was reported that chronic treatment with pregnanolone uncouples allosteric interactions between steroid and benzodiazepine recognition sites in cultured neurons. The present study was designed to assess the effect of repeated stress on the modulation of neuroactive steroid on the GABAA receptor. The effect of steroid on the ligands binding to GABAA receptor was investigated using cerebral cortices of unstressed and repeatedly immobilized Sprague-Dawley rats and borderline-hypertensive rats. Pregnanolone enhanced the binding of [³H]flunitrazepam to GABAA receptor in both of unstressed and repeatedly stressed Sprague-Dawley rats. However, repeatedly stressed rats showed significantly lower values in EC50 and higher values in Emax of enhancement binding of [³H] flunitrazepam than those of unstressed rats. Borderline-hypertensive rats, which are known to be overreactive to stress, showed higher basal binding of [³H]flunitrazepam to GABAA receptor, similar to those of stressed Sprague-Dawley rats. Borderline-hypertensive rats, in both control and stressed groups, showed similar pharmacological activity of pregnanolone with Sprague-Dawley rats. From these findings, it can be concluded that repeated immobilization stress enhanced the positive modulation of neuroactive steroid on the GABAA-receptor complex, and these pharmacological activity of neuroactive steroid might be a defensive reactivity to stress of individuals.

      • 터널굴착의 시공성 분석

        Efficiency of tunnel excavation using the jumbo drill is evaluated using the field data. To evaluate the efficiency, variations of excavation and overbreak quantities for different drilling patterns are related with rock qualities represented by rock mass rating(RMR). The drilling patterns consist of the number fo drill hole, and the geometry and depth of drill oles. Among effects of these patterns on excavation, the effect of the number of drill hole is mainly considered. On the basis of the observed relationships, it is intended to suggest the most appropriate drilling pattern for various rock qualities resulting the smallest amount of overbreak. In addition to this investigation, the construction time required for a unit advance of excavation is also investigated when the jumbodrill is used. From the result, the standard advance rate and the efficency of the drilling equipment are evaluated, respectively.

      • Er:YAG laser 조사에 의한 치은연하치석 제거효과

        The purpose of this in vitro study was to evaluate the efficiency of Er:YAG laser on calculus removal and the morphologic changes and hardness of the irradiated surface at different power settings. This experiment used human teeth which were extracted due to periodontal disease and had a band of calculus. Forty root slabs (5x5mm^2) were made and divided into control group and irradiated groups. Experimental groups were as follows;Control group (root planing), Group 1 (irradiated with laser at 30mj), Group 2(irradiated with laser at 60mJ). and Group 3(irradiated with laser at 100mJ). Twelve root slabs embedded in resin block were used in each group. Er:YAG laser was applied under water irrigation with the tip held perpendicular to the root surface in contact mode. The treatment time was measured until the calculus was removed completely under naked eyes. The efficiency of calculus removal was evaluated by the time for removal. Morphological changes of laser irradiated site were observed under SEM and the surface hardness was measured using a VH tester. The results were as follows; 1. The efficiency of laser scaling was increased with increasing the energy level of irradiation. (p<0.05) 2. The morphological changes such as carbonization, crater and scale-like defects in the irradiated root surface were frequently observed with increasing the energy level. 3. The surface hardness tended to increase at 60mJ and 100mJ irradiated groups than that of control group. From the results evaluting on the efficiency, morphological change, and surface hardness, lower energy level was suggested f'or the clinical application of the Er:YAG laser in scaling.
