Efficient Construction of Quaternary Carbon: Stereocontrolled Synthesis of Novel Abacavir Analogue

This paper discusses the racemic and stereoselective synthetic route for novel 4'a -methyl and 6'a -methyl analogues of abacavir. The quaternary carbon at the 4'-position of carbocyclic nucleoside was installed successfully via a Claisen rearrangement. The stereocontrolled construction of a methyl group in the 6'a -position was directed through the Felkin-Anh rule. A Bis-vinyl compound 9 was cyclized successfully using Grubbs catalyst II to provide a carbocycle nucleus for the target compound. The synthesized compound 15 showed moderate anti-HIV activity (EC50 = 10.67 m M, MT-4 cell lines).

Geckos can run even on an inverted surfaces with exceptional speed, strength and agility for their size, representing in many respects an ideal model system for the study of terrestrial locomotion. By measuring the ground reaction force(GRF)s generated by animals when walking upright, climbing vertically or walking on an. inverted surface, we can understand animals adhesion mechanism and locomotion dynamics. We used a sensors array to measure the GRFs. When geckos walked on vertical and inverted surfaces, the lateral force and the fore-aft force play an important role.

This essay focus on the output-input ratio to establish corporate culture construction efficiency index system, with DEA, tries to figure out appraisal method and improvement suggestions, finally prove it by example analysis.

The synthesis of 4',5'-doubly branched carbocyclic nucleosides was accomplished in this study. The selective methylation in the 5'-position was made by Felkin-Anh controlled Grignard addition. The construction of the required 4'-quaternary carbon was carried out by using a [3,3]-sigmatropic rearrangement. Bis-vinyl 6 was successfully cyclized using a Grubbs' catalyst ll. The natural pyrimidine bases (cytosine, uracil, thymine) were efficiently coupled using a Pd(0) catalyst. When the synthesized compounds were examined for their activity against several viruses such as the HIV-1, HSV-1, HSV-2 and HCMV, the cytosine analogue 13 exhibited weak antiviral activity against the HCMV.
Synthesis and anti-HCMV activity of novel cyclopropyl phosphonic acid nucleosides

<P>A simple synthetic route for novel acyclic phosphonate nucleosides is described. The characteristic cyclopropyl moiety <B>8</B> was constructed employing the Simmons-Smith reaction as key step starting from simple acyclic 2-butene-1,4-diol. The condensation of the mesylate <B>11 </B>with natural nucleosidic bases (A,C,T,U) under nucleophilic substitution conditions (K<SUB>2</SUB>CO<SUB>3</SUB>, 18-Crown-6, DMF) and hydrolysis afforded the target nucleosides <B>16, 17, 18,</B> and <B>19</B>. In addition, the antiviral evaluations against various viruses were performed.</P>
