        공립 보육시설 적용 태양열냉난방시스템의 에너지성능 평가

        This study examined the energy performance of a solar heating and cooling system for an public child care center by using the dynamic simulation program (TRNSYS). This solar heating and cooling system incorporates evacuated tube solar collectors of 229.5 m2, storage tank of 15 m2, 348.8 kW auxiliary heater and single-effect LiBr/ H2O absorption chiller of 20RT nominal cooling capacity. As a result, the average daily efficiency of the heating and cooling collector were 36% and 34% respectively. The average monthly efficiency of the heating and cooling collector were 32% and 28%, and the share of the solar heating and cooling system for loads were 45% and 26%. And the average yearly collector efficiency of the system was 27%. and the share of the system for loads was 37%.

        반복유산을 경험한 384부부의 세포유전학적 연구

        During the years 1984 to 1989, in order to determine of chromosome abnormalities are associated with recurrent spontaneous abortions, cytogenetic studies were performed 384 couples. Abnormal karyotypes were found in 51(13.3%) couples. There was no apparent relation with the number of abortions. The abnormalities were as follows: 17(4.4%) balanced translocation; 15(3.9%) mosaicisms; 17(4.4%) pericentric inversion; 2(0.5%) addition or isochromosome. Chromosome abnormalities were observed in 34(67%) of the wives and 17(33%) of the husbands. In addition, we detected polymorphic variants of chromosomes in 89(23.2%) subjects. Reciprocal translocations(13/17) were more common than the robertsonian type(4/17). All of the mosaicisms were associated with the sex chromosomes in 10 females and 5 males subjects. Pericentric inversions were most common in chromosome 9. Compared to previously studied general populations, significantly higher frequencies of translocations, mosaicisms and inversions were found in couples with repetitive spontaneous abortion. This suggests that couples should have chromosome studies after two or more abortions.

        Leavening Ability of the Isolate Saccharomyces cerevisiae MF10003 in Bakery Dough

        건포도 발효 배양액으로부터 이산화탄소 생성능이 우수한 효모를 분리하였다. 분리 효모는 18S rDNA ITS 유전자 상동성을 비교하여 Saccharomyces cerevisiae로 동정하고 Saccharomyces cerevisiae MF10003로 명명하였다. 분리효모 S. cerevisiae MF10003은 풍미 발생 야생효모인 S. ellipsoideus KCTC7243에 비하여 각각 1.9배 및 3.1배 우수한 이산화탄소 생성능 및 dough 발효 팽창능을 나타냈었다. 분리 효모는 glucose, fructose, sucrose, maltose를 기질로 하여 4시간 이내에 이산화탄소 발생력을 보여 주었다. An effective leavening yeast was isolated from raisin broth. The isolate was identified as Saccharomyces cerevisiae by comparing the homology of 18S rDNA ITS sequences and named as S. cerevisiae MF10003. S. cerevisiae MF10003 showed a 1.9-fold and 3.1-fold increase in CO₂ production and leavening ability, respectively, compared with the wild yeast S. ellipsoideus KCTC7243, and the dough had a rich and volatile flavor. When glucose, sucrose, fructose, and maltose were added to the culture broth as a carbon and energy source, CO₂ was produced in 4 hr.

        Effects of Energy Substrates on In Vitro Fertilization of the Mouse Oocytes with Cumulus Mass and their Developments

        난구세포는 lactate와 pyruvate를 쉽게 생성하고, 이로 인해 배양액내 에너지원의 농도를 변화시켜 난자의 수정과 배양에 영향을 주는 것으로 알려져 있다. Glucose, lactate 및 pyruvate의 농도가 상이한 M16, MTF 및 CZB배양액에서 난구세포를 포함한 생쥐난자의 체외 수정과 발달을 관찰하여, 이들 기질의 영향에 대하여 살펴보고 배양액의 유용성을 재검토하고자 하였다. Glucose를 제거한 배양액 (CZ2 배양액)에서 수정율과 배반포 형성율은 다른 배양액에 비해 유의하게 감소되었으나 (p<0.05), 생쥐 난관액과 동일한 기질 농도로 조성된 MTl (난간액이 난구세포를 포함하고 있을 때) 및 MT2배양액 (난관액이 난구세포를 포함하고 있지 않을 때)과 glucose를 포함한 modified CZB배양액에서는 영향이 없었다. 이와 같은 결과로 기질의 농도를 생리적 수준으로 조정한 배양액의 이용은 난구세포를 포함한 생쥐 난자의 체외수정과 그 발달을 향상시키지 못하고, glucose의 제거는 악영향을 나타내는 것으로 사료된다. Cumulus cells have possibly influence on fertilization of mouse oocytes and their subsequent development in vitro, because they readily produce lactate and pyruvate and can modify the concentration of substrates in the medium. In vitro fertilization of mouse oocytes with cumulus mass and their developments in five media which were differently composed in concentrations of glucose, lactate and pyruvate were observed. In the absence of glucose (CZ2 medium) decreased (p<0.01) the percentage of fertilization and embryos reaching the blastocyst stage. But, in the same concentration of glucose, lactate and pyruvate as mouse oviductal fluid with (MT1 medium) and without (MT2 medium) cumulus mass and modified CZB medium containing glucose (CZ1 medium) had no effects (p>0.05). These studies indicate that the adjustments of energy substrates concentration to the physiological level did not improve the fertilization of mouse oocytes with cumulus mass and their development in vitro, and the deletion of glucose showed adverse effects.

        ICR계 생쥐 1세포배를 이용한 수질의 평가

        To confirm the overcome of in vitro 2-cell block, ICR mouse I-cell embryos were cultured in CZB media. All embryos in CZB were overcome in vitro 2-cell block and 92% of embryos were developed to the blastocyst at day 4. However, in m-KRB group(control) only 20% of embryos were developed over 2-cell. Any embryos in m-KRB did not develop to the morular stage. Developments and degenerations of ICR mouse I-cell embryos were compared in CZB medium prepared with water of three quality:(l) Milli-Q ultrafiltration water(UF);(2) Milli-Q reverse osmosis water(RO);(3) tap water(TAP). The objective was to evaluate the potential of quality control using ICR mouse 1-cell embryos. The more water was purified, the better embryo developments were supported and the less embryos were degenerated. As a quality control system, the culture of ICR 1-cell mouse embryos in CZB was useful.

        미배양 양수세포에서 다운증후군과 에드워드증후군의 확인을 위한 FISH의 이용

        N/A FISH is suggested as a possible method to detect the numerical and structural abnormalities of chromosomes in interphase nucleus. We performed this study to discuss the clinical usefulness of FISH in uncultured amniocytes and to set up the cut-off value for further study. We collected amniotic fluid samples from patients whose chromosome studies were recommended due to screen positive for Down and Edword syndrome in triple marker test using maternal serum. The centromeric probe for chromosome 18 and the locus-specific probe for chromosome 21 were used and the results were compared to their karyotypes. We could find 2 cases of trisony 21 and 2 cases of trisony 18 and the other cases represented normal karyotypes. The accuracies were 91% for disomy 18, 89% for trisomy 18, 92% for disomy 21 and 88% for trisomy 21. Therefore FISH technique is a possible method to detect the chromosomal abnormalities in uncultured amniocytes and the use of locus-specific probe for chromosome 21 would be more useful for detecting the aneuploidy of chromosome 21 than 13/21 centromeric probe.

        미배양 태아제대혈액세포에서의 염색체이상 확인을 위한 FISH 의 이용

        목적: 미배양 태아제대혈액세포에서 FISH를 실시함으로써 신속하게 염색체 이상을 확인하고자 한다. 방법: 서울대학교병원 산부인과 및 함춘여성크리닉을 방문한 산모들 중 신속한 핵형분석이 필요한 경우 중 제대혈천자술을 시행한 14례의 환자들을 대상으로 염색체 18, 21, X, Y에 대해 FISH를 실시하였다. 결과 : 총 14례에서 염색체 18번과 21번에 대해 관찰한 결과, 1례의 에드워드증후군과 5례의 다운증후군을 관찰하였고 나머지는 정상으로 나타났다. 염색체 18번과 21번에 대한 정확도는 평균 91.0%와 92.4%로 나타났으며, 핵형분석과 비교한 결과는 모두 일치하였다. 1례의 45,X에서 염색체 X와 Y에 대한 FISH의 정확도는 평균 84.6%와 93.9%로 나타났으며, 핵형 분석 결과와 모두 일치하였다. 24시간 배양후 핵형분석에 실패한 9례는 48시간 이상 배양하여 핵형분석결과를 확인할 수 있었고, 3례의 다운증후군과 1례의 45,X, 3례의 46,XX 그리고 2례의 46,XY를 관찰할 수 있었다. 결론: FISH를 실시함으로써 미배양 태아제대혈액세포에서 48시간 이내에 염색체의 수적이상을 정확하게 확인할 수 있어 신속한 결과가 요구되는 경우와 배양이 용이하지 않아 핵형분석에 실패하는 경우 FISH가 매우 유용한 방법이 되리라 사료된다. Objective: To determine the fetal aneuploidy in fetal blood cells from cordocentesis. Methods: We analyzed their karyotype and performed fluorescence in situ hybridization(FISH) for chromosome 18, 21, X, and Y in 14 cases of fetal blood cells from cordocentesis at Department of Obstetrics & Gynecology, College of Medicine, Seoul National University and Hamchoon Women's Clinic. Results: In all cases we obtained the consistent results in both methods and were able to rapidly detect aneuploidy in uncultured fetal blood cells using FISH before karyotyping with culture for 48 hr. The averages for accuracy of FISH were from 84.6 % to 93.9%. Conclusion: In this study we suggest that the rapid detection in uncultured fetal blood using FISH is possible and that this diagnostic method will be clinically useful when rapid result would be demanded.
