EcoRI Methylase 의 Cysteine Residue 역할에 관한 연구

The chemical modification of cysteinyl residue of EcoRl methylase by NEM and DACM resulted in the loss of the enzyme activity following pseudo first order kinetics. Second order rate constants (K) for the inactivation of EcoRI methylase by NEM and DACM were 685 M^(-1)·min^(-1) and 99960 M^(-1)·min^(-1), respectively. The reaction orders with respect to NEM and DACM were determined from plot of log (100/t_½) against log (NEM) and log (DACM). When the datas are plotted as indicated above, slope of 0.95 and 1.25 are obtained, suggesting that inactivation is the result of the reaction of one cysteinyl residue per active site of EcoRI methylase. The inactivation of EcoRl methylase by DTNB was reversed upon addition of excess 2-mercaptoethanol. When the enzyme solution was preincubated with the substrates, the inactivation of the enzyme by NEM and DACM was protected. Furthermore, loss of SAM binding capacity of EcoRI methylase was detected by nitrocellulose filter binding assay. Based on the data, we suggest that cysteinyl residue of EcoRI methylase plays a very important role as a SAM binding residue.

Abstract: Treatment of EcoRI methylase with phenylglyoxal resulted in time and concentration dependent enzyme inactivation. The reaction followed pseudo first-order kinetics until 90 to 95% of the enzyme had been inactivated, and prolonged incubation with phenylglyoxal resulted in complete inactivation. Second order rate constant (K) for the inactivation of EcoRI methylase by phenylglyoxal was 666 M^(-1)·min^(-1). The slope determined from the plot of log (100/t^(½)) against log (phenylglyoxal) indicated that the reaction of 1 molecule of phenylglyoxal reagent in necessary for inactivation of each active site in EcoRI methylase. Preincubation with pUC19 or S-adenosylmethionine as substrates decreased significantly the rate of inactivation by phenylglyoxal. By nitrocellulose binding assay, phenylglyoxal was observed to reduce DNA binding capacity of the enzyme more rapidly than S-adenosylmethionine. Furthermore, DNA binding capacity of the enzyme was preserved when the enzyme was preincubated with pUC19 DNA. The cumulative results suggest that arginyl residue plays a very important role as a DNA binding residue in EcoRI methylase.

The flow around a ROV (Remotely Operated Vehicle) has been numerically investigated to improve resistance performance by modifying the hull form of the ROV. For the base hull form considered ill this study, the form drag rather than the friction drag is dominant to the total drag. Subsequently, the surfaces on which the local pressure highly acts have been modified to produce the streamlined-shape. Based on the surface modification, seven different hull forms have been chosen as candidates for drag reduction. Among the candidates, the semi-sphericalized housing and the streamlined-bow achieved greatest drag reduction comparing with the others. Consequently, tue hull form combined with the semi-sphericalized housing and tile streamlined-bow gave approximately 17% drag reduction at the design velocityof 3 knots.

The flow around a remotely-operated vehicle (ROV) has been investigated numerically to improve the resistant performance by modifying the hull form of the ROV. In the case of the base hull form considered in this study, form drag rather than friction drag was the dominant component of total drag. Subsequently, the surfaces that were most susceptible to local pressure effects were modified to give them a more streamlined shape. Eleven different hull forms were chosen to undergo surface modification for drag reduction. In addition, four different boot-tail appendages with different slant angles were installed at the stern to reduce the wake vortices that are induced by the local regions of very low pressure. Consequently, a total of 11 different hull forms for drag reduction were considered. The final hull form, which combined the hull for which surface modification resulted in the lowest drag with a boot-tail appendage with a 15-degree slant angle, resulted in a drag reduction of 20%.

The international SPICE (Software Process Improvement and Capability determination) Project ISO/IEC 15504(SPICE : Software Process Improvement and Capability determination) is an emerging International Standard on SPA(Software Process Assessment). A prime motivation for developing this standard has been the perceived need for an internationally recognized software process assessment framework that pulls together the existing public and proprietary models and methods. A SPICE assessment can be considered as one of representative SPA model since assessors assign ratings to indicators and metrics to measure the capability of software process. But this models doesn't provide a systematic measurement procedures and dynamic method for SPI(Software Process Improvement). Through the evaluation of SPICE is capable of providing a substantiated basis for using the notion of capability, as well as providing information for nacessary improvements to the standard using 6sigma process. As a result, this paper propose a measurement procedure and guidelines for application of 6sigma process to guarantee the reliability in SPI and suggest the structure to support SPI on overall organization.

What is the most important in articles of food is hygienic safety. Because food Is the most common thing in our everyday life, however, the importance of its hygienic safety and other many problems caused by food may be easily neglected. What is more, food is can be dangerous as much as it is directly related to human life and accidents from the same cause may have different effects on the victims according to physical and environmental differences of individuals. Thus PL action for food requires more thorough prevention and measure. Korea has been enforcing 'the Product Liability Act' since the 1/sup 1st/ of July 2002. Product Liability (PL) is liability of the manufacturer or the seller of a product to compensate for the death or injury of consumers or the loss of properties caused by the defect of the product. This study surveyed consumers' response to and the effects of the enforcement of the Product Liability Act, investigated how consumers perceived the importance of food safety and the risk of defective food based on PL standards and their experience in damage by food through a questionnaire survey, and analyzed collected data through empirical analyses (reliability analysis, factorial analysis, regression analysis and ANOVA t-test) using SPSS 10.0. Based on the results of analysis, the researcher proposed strategies for coping with the Product Liability Act in the food industry.

FPSO model tests of mooring line failure were carried out in the ocean basin at KRISO. The characteristics of the motions and mooring lines were investigated. The FPSO ship was moored using an internal turret and catenary mooring. The test model was 1/60 scale. The mooring lines were designed to satisfy the characteristics of the original mooring lines using the truncation method. The experiments were conducted under various environments: a safe wave, current and wind condition; single broken mooring line condition; and transient mode condition. The moment of the break was determined based on experimental test results. The results showed that the FPSO behavior and mooring line tensions were acceptable under the failure condition.
