대장암 세포주에서 주박 추출물의 유기용매 분획물의 항성장 활성

본 연구는 한국 고유의 전통주인 막걸리 제조시 생성되는 주박 추출물과 유기용매 분획물을 총 85종 분리하여, 이들의 대장암 세포주에 대한 항성장 활성과 활성기전을 연구하였다. 이들 중 세포생존율 연구결과에 따라 3가지 종류의 분획물(KSD-E1-3, KSD-E2-3, KSD-E4-3)을 선별하였다. 이 중 가장 활성이 높은 KSD-E1-3 분획물에 의한 유전체 수준에서의 유전자 발현변화를 확인하기 위하여 oligo DNA microarray 실험을 수행하였다. 그 결과, 2배 이상 발현이 증가된 유전자들 가운데 항암 유전자인 NAG-1, ATF3, DDIT3 그리고 p21을 선별하여 추가 실험을 진행하였다. 세포생존율 결과에 따라 선별된 3가지 종류의 분획물(KSD-E1-3, KSD-E2-3 그리고 KSD-E4-3)에 의한 4가지 종류의 유전자의 발현을 확인한 결과, KSD-E1-3에 의하여 모든 유전자의 발현이 높게 증가되었다. KSD-E1-3에 의한 유전자의 발현이 전사조절인자 p53에 의존적인지 확인하고자 p53 null HCT116 세포주를 이용하여 RT-PCR한 결과, NAG-1, ATF3, 그리고 DDIT3 유전자 p53에 비의존적이었으며, 반면 p21 유전자는 p53에 의해서만 발현이 증가되는 것을 확인하였다. 이러한 연구 결과는 주박 추출물이 다양한 기작에 의해 항암 유전자의 발현을 유도함으로써 암 세포에 대한 항성장 활성을 보여주고 있음을 나타낸다. In the present study, we prepared eighty-five different kinds of lees extracts and their solvent fractions and investigated their anti-proliferative activities against human colorectal cancer HCT116 cells. HCT116 cells were treated with eighty-five solvent fractions of lees extracts and then cell viability was measured using MTS assay. Among the treated solvent fractions, three solvent fractions (KSD-E1-3, KSD-E2-3, and KSD-E4-3) were selected based on cell viability assay. In addition, we performed an oligo DNA microarray analysis to analyze the gene expression changes by treatment of KSD-E1-3 in HCT116 cells. Among the up-regulated genes, we selected 4 genes (NAG-1, ATF3, p21, and DDIT3) and performed RT-PCR using gene-specific primers. Among the treated solvent fractions, KSD-E1-3 dramatically induced the expressions of the four selected genes. In addition, we investigated whether the up-regulations of those genes were dependent on the transcription factor p53’s presence using p53 null HCT116 cells. The results indicate that the up-regulations of NAG-1, ATF3, and DDIT3 are not dependent on the p53 presence, whereas p21 is dependent on the p53 presence. These findings may help to understand the molecular mechanisms of the anti-proliferative activity mediated by rice wine lees in human colorectal cancer cells.
주박 추출물과 이들의 유기용매 분획물에 의한 항염증 활성

본 연구에서는 전통주 제조시 부산물로 생산되는 주박과 누룩으로부터 추출물과 유기용매 분획물을 총 85종을 제조하고, 이들에 의한 항염증 활성을 연구하였다. 85종의 분획물 중 선별한 세 가지의 분획물(KSD-E1-3, KSD-E2-3, KSD-E4-3)에 의해서 LPS에 의해 염증이 유도된 RAW 264.7 세포주에서 nitric oxide 생산이 현저히 감소됨을 확인하였다. 또한, 세가지 분획물에 의해 염증유발 유전자인 COX-2, TNF-alpha, 그리고 iNOS 유전자의 발현이 감소되었다. 세 가지 분획물 중 KSD-E4-3에 의한 항염증 활성의 작용기전을 이해하기 위하여 oligo DNA microarray를 수행하였다. 마이크로어레이 결과 발현이 감소된 유전자 중 염증과 관련된 유전자 6개(IL-1F6, iNOS, IL-10, Fabp4, IL-1RN, CSF2)를 선택하여, RT-PCR과 정량적 real-time PCR을 수행하였다. 그 결과, 모든 유전자의 발현이 감소됨을 확인하였다. 결론적으로, 이러한 연구결과는 전통주 주박이 항염증 활성을 가지고 있는 식품이나 약품을 개발하는데 필요한 새로운 자원으로서 활용 가능함을 시사하는 것이다. In the current study, we prepared eighty-five different kinds of solvent fractions of rice wine lees and nuruk extracts and investigated their effects on cell viability and nitric oxide (NO) production in mouse RAW 264.7 cells. Among the treated solvent fractions, only three solvent fractions (KSD-E1-3, KSD-E2-3 and KSD-E4-3) significantly decreased NO production in LPS-activated RAW 264.7 cells without affecting cell viability. And, they also reduced the expression of pro-inflammatory genes such as COX-2, TNF-alpha and iNOS. To understand the molecular mechanisms involved in the inhibition of inflammation in (KSD-E4-3)-treated RAW 264.7 cells, we carried out oligo DNA microarray analysis using Agilent Mouse microarray. To confirm microarray data, 6 genes (IL-1F6, iNOS, IL-10, Fabp4, IL-1RN and CSF2) were selected and performed RT-PCR and quantitative real-time PCR analysis with gene specific primers. The results of RT-PCR and real-time PCR agreed with microarray data. Overall, our results suggest that rice wine lees can be a novel resource for the development of foods and drugs which possess anti-inflammatory activity.
프로바이오틱스 생산 exopolysaccharide에 의한 항염증 활성

본 연구에서는 다양한 식품원으로부터 7종류의 프로바이오틱스를 분리하여 16S rDNA 염기서열 분석을 통해 동정하였으며, 그 결과 Bacillus sp.와 Lactobacillus sp.로 확인되었다. 먼저 LPS로 활성화 된 RAW 264.7 세포주에 7종의 균주 배양액을 처리한 후, nitric oxide (NO) 생성을 측정하였다. 처리한 균주의 배양액 중 Bacillus sp. FG-1과 Lactobacillus sp. FG-6 균주의 배양액 처리군에서 현저하게 NO 생성이 저해되었다. 또한, 이들의 처리에 의해 COX-2, iNOS 그리고 TNF-α와 같은 pro-inflammatory 유전자의 발현이 감소되었다. 균주가 생산하는 여러 물질 중 exopolysaccharide (EPS)가 항염증 활성과 관련이 있는지를 검증하기 위하여 두 균주로부터 EPS를 분리하여, 이들이 NO 생성에 미치는 영향을 연구하였다. 그 결과, 두 균주가 생산하는 EPS가 NO 생성을 농도의존적으로 저해하였고, pro-inflammatory 유전자의 발현도 현저하게 감소시켰다. 이러한 연구결과는 EPS가 프로바이오틱스가 생산하는 여러 물질 중 항염증 활성의 핵심물질 중의 하나가 될 수 있음을 시사한다. The present study isolated seven different kinds of probiotics from various food sources and identified them with Bacillus sp. and Lactobacillus sp. by 16S rDNA sequencing. Their supernatants were prepared after a 24 hr culture, and their effects on nitric oxide (NO) production in mouse RAW 264.7 cells were investigated. Among the treated samples, the culture supernatants of two strains (Bacillus sp. FG-1 and Lactobacillus sp. FG-6) significantly decreased NO production in LPS-activated RAW 264.7 cells. Moreover, they dramatically reduced the expression of pro-inflammatory genes such as COX-2, iNOS, and TNF-α. To examine whether exopolysaccharide (EPS) is responsible for the anti-inflammatory effects of probiotics, EPS was purified from the culture supernatants of Bacillus sp. FG-1 and Lactobacillus sp. FG-6 strains. The EPS treatment produced by FG-1 and FG-6 strains decreased NO production in a dose-dependent manner in LPS-stimulated RAW 264.7 cells without affecting cell viability, while also reducing pro-inflammatory gene expression. Overall, these results suggest that EPS might be one of the key molecules responsible for the anti-inflammatory effects of probiotics.
