        Synergistic Effect of Bone Marrow-Derived Mesenchymal Stem Cells and Platelet-Rich Plasma in Streptozotocin-Induced Diabetic Rats

        Background: Diabetic wounds are a major clinical challenge, because minor skin wounds can lead to chronic, unhealed ulcers and ultimately result in infection, gangrene, or even amputation. Studies on bone marrow derived mesenchymal stem cells (BMSCs) and a series of growth factors have revealed their many benefits for wound healing and regeneration. Platelet-rich plasma (PRP) may improve the environment for BMSC development and differentiation. However, whether combined use of BMSCs and PRP may be more effective for accelerating diabetic ulcer healing remains unclear. Objective: We investigated the efficacy of BMSCs and PRP for the repair of refractory wound healing in a diabetic rat model. Methods: Forty-eight rats with diabetes mellitus induced by streptozotocin were divided into four groups: treatment with BMSCs plus PRP, BMSCs alone, PRP alone, phosphate buffered saline. The rate of wound closure was quantified. A histopathological study was conducted regarding wound depth and the skin edge at 7, 14, and 28 days after surgery. Results: Wound healing rates were significantly higher in the BMSC plus PRP group than in the other groups. The immunohistochemistry results showed that the expression of platelet/endothelial cell adhesion molecule 1, proliferating cell nuclear antigen, and transforming growth factor-β1 increased significantly in the BMSC plus PRP group compared to the other treatment groups. On day 7, CD68 expression increased significantly in the wounds of the BMSC plus PRP group, but decreased markedly at day 14 compared to the controls.       • Multiresponse Surfaces Optimization Based on Evidential Reasoning Theory

        During process design or process optimization, it is quite common for experimenters to find optimum operating conditions for several responses simultaneously. The traditional multiresponse surfaces optimization methods do not consider the uncertain relationship among these responses sufficiently. For this reason, the authors propose an optimization method based on evidential reasoning theory by Dempster and Shafer. By maximizing the basic probability assignment function, which indicates the degree of belief that certain operating condition is the solution of this multiresponse surfaces optimization problem, the desirable operating condition can be found.

        ON THE EXISTENCE OF SOLUTIONS OF EXTENDED GENERALIZED VARIATIONAL INEQUALITIES IN BANACH SPACES

        In this paper, we study the following extended generalized variational inequality problem, introduced by Noor (for short, EGVI) : Given a closed convex subset K in q-uniformly smooth Banach space B, three nonlinear mappings T : $K\;{\rightarrow}\;B^*$, g : $K\;{\rightarrow}\;K$, h : $K\;{\rightarrow}\;K$ and a vector ${\xi}\;{\in}\;B^*$, find $x\;{\in}\;K$, $h(x)\;{\in}\;K$ such that <Tx-$\xi$, g(y)-h(x)> $\geq$ 0, for all $y\;{\in}\;K$, $g(y)\;{\in}\;K$. [see [2]: M. Aslam Noor, Extended general variational inequalities, Appl. Math. Lett. 22 (2009) 182-186.] By using sunny nonexpansive retraction $Q_K$ and the well-known Banach's fixed point principle, we prove existence results for solutions of (EGVI). Our results extend some recent results from the literature.

        Comparison between Field Test and Numerical Analysis for a Jacket Platform in Bohai Bay, China

        This paper presents a comparison between numerical analysis and field test on a real offshore platform in Bohai Bay, China. This platform is a steel jacket offshore platform with vertical piles. The field testing under wave-induced force and wind force etc. was conducted, in order to obtain the dynamic parameters of the structure, including the frequencies of the jacket platform, as well as the corresponding damping ratios and mode shapes. The natural excitation technology (NExT) combined with eigensystem realization algorithm (ERA) and the peak picking (PP) method in frequency domain are carried out for modal parameter identification under operational conditions. The three-dimensional finite element model (FEM) is constructed by ANSYS and an analytical modal analysis is performed to generate modal parameters. The analytical results were compared with experimental results. A good agreement was achieved between the finite element and analysis and field test results. It is further demonstrated that the numerical and experimental modal analysis provide a comprehensive study on the dynamic properties of the jacket platform. According to the analysis results, the modal parameters identification under ambient excitation can calibrate finite element model of the jacket platform structures, or can be used for the structural health monitoring system.

        Antiserum Preparation of Recombinant Sweet Potato Latent Virus-Lotus (SPLV-Lotus) Coat Protein and Application for Virus-Infected Lotus Plant Detection

        Lotus is one of the most important aquatic vegetables in China. Previously, we detected sweet potato latent virus from lotus (SPLV-lotus) and found that it has highly significant sequence diversity with SPLV-sweet potato isolates (SPLV-sp). Here, we developed serological methods for the detection of SPLV-lotus in Chinese lotus cultivation areas. Based on the high sensitivity of SPLV-lotus coat protein antiserum, rapid, sensitive and large-scale diagnosis methods of enzyme-linked immunosorbent assay (ELISA) and dot blot in lotus planting area were developed. The established ELISA and dot blot diagnostic methods can be used to detect SPLVlotus from samples successfully. And our results also showed that the SPLV-lotus and sweet potato isolates appeared clearly distinction in serology. Our study provides a high-throughput, sensitive, and rapid diagnostic method based on serology that can detect SPLV on lotus, which is suggested to be included in viral disease management approach due to its good detection level.

      • Tas13D Inhibits Growth of SMMC-7721 Cell via Suppression VEGF and EGF Expression

        Objective: Taspine, isolated from Radix et Rhizoma Leonticis has demosntrated potential proctiective effects against cancer. Tas13D, a novel taspine derivative synthetized by structure-based drug design, have been shown to possess interesting biological and pharmacological activities. The current study was designed to evaluate its antiproliferative activity and underlying mechanisms. Methods: Antiproliferative activity of tas13D was evaluated by xenograft in athymic mice in vivo, and by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and cell migration assays with human liver cancer (SMMC-7721) cell lines in vitro. Docking between tas13D and VEGFR and EGFR was studied by with a Sybyl/Surflex module. VEGF and EGF and their receptor expression was determined by ELISA and real-time PCR methods, respectively. Results: Our present study showed that tas13D inhibited SMMC-7721 xenograft tumor growth, bound tightly with the active site of kinase domains of EGFR and VEGFR, and reduced SMMC-7721 cell proliferation (IC=34.7 ${\mu}mol/L$) and migration compared to negative controls. VEGF and EGF mRNAs were significantly reduced by tas13D treatment in a dose-dependent manner, along with VEGF and EGF production. Conclusion: The obtained results suggest that tas13D inhibits tumor growth and cell proliferation by inhibiting cell migration, downregulating mRNA expression of VEGF and EGF, and decreasing angiogenic factor production. Tas13D deserves further consideration as a chemotherapeutic agent.

        Evolutionary Rates and Phylogeographical Analysis of Odontoglossum Ringspot Virus Based on the 166 Coat Protein Gene Sequences

        Odontoglossum ringspot virus (ORSV) is a member of the genus Tobamovirus. It is one of the most prevalent viruses infecting orchids worldwide. Earlier studies reported the genetic variability of ORSV isolates from Korea and China. However, the evolutionary rate, timescale, and phylogeographical analyses of ORSV were unclear. Twenty-one coat protein (CP) gene sequences of ORSV were determined in this study, and used them together with 145 CP sequences obtained from GenBank to infer the genetic diversities, evolutionary rate, timescale and migration of ORSV populations. Evolutionary rate of ORSV populations was $1.25{\times}10^{-3}nucleotides/site/y$. The most recent common ancestors came from 30 year ago (95% confidence intervals, 26-40). Based on CP gene, ORSV migrated from mainland China and South Korea to Taiwan island, Germany, Australia, Singapore, and Indonesia, and it also circulated within east Asia. Our study is the first attempt to evaluate the evolutionary rates, timescales and migration dynamics of ORSV.

      • Robust Multi-response Parameter Design based on RSM

        This paper discusses multi-response robust parameter design problems based on response surface method. Most research effort on multi-response parameter design problem focuses much on finding out optimal parameters based on certain criteria or objectives. Research shows that optimal solution in terms of some criteria may not be robust. To achieve robust solution we should consider how sensitive the solution is when the factors change around it. A comparative study of methods for multi-response robust parameter design is conducted. Solution with consideration of robustness and optimality is proposed with applications of the example.

        STRONG CONVERGENCE THEOREMS FOR ASYMPTOTICALLY QUASI-NONEXPANSIVE MAPPINGS AND INVERSE-STRONGLY MONOTONE MAPPINGS

        In this paper, we consider an iterative scheme for finding a common element of the set of fixed points of a asymptotically quasi nonexpansive mapping and the set of solutions of the variational inequality for an inverse strongly monotone mapping in a Hilbert space. Then we show that the sequence converges strongly to a common element of two sets. Using this result, we consider the problem of finding a common fixed point of a asymptotically quasi-nonexpansive mapping and strictly pseudocontractive mapping and the problem of finding a common element of the set of fixed points of a asymptotically quasi-nonexpansive mapping and the set of zeros of an inverse-strongly monotone mapping.
