Plant regeneration from cultured hypocotyls may provide means for overcoming breeding barriers caused by heterostylic cross-incompatibility in interspecific crosses of common buckwheat(Fagopyrum esculentum Moench). Hypocotyles were excised from seed embryos of common buckwheat(cv. Miyazakizairai) and wild annual buckwheat(F. homotropicum Ohnishi) and cultured on MS medium supplemented with 0∼2.0 mg/l IAA and 0~2.0 mg/l BAP. Shoot was induced only on the medium containing 1.0 mg/l NAA and 1.0 mg/l BAP, shoot induction was promoted by 2.4-D and the frequency was lower in wild annual buckwheat than in common buckwheat. Roots were developed on White's medium containing 1.0 mg/l IBA. Whole plantlets were obtained following the production of adventitious roots from the shoots. When transferred from sterile culture condition to peat and soil medium, most plantlets were able to survive and grew to have flowers. Development and Genetic Analysis of Near Isogenic Lines for Opaqueness of Endosperm in Rice

Rice plants segregating for opaque endosperm were selected and tested in every generation from F5 to F9 of a CB243/IR50 cross. Six near isogenic lines(NILs) showing diference in opaqueness, of which three lines have opaque endosperm with theothers having translucent endosperm, were selected among F9 lines. No diferences were found in plant growth characteristicsexcept for 1,000-grain weight between opaque and normal translucent endosperm NILs. F2 plants of two crosses between opaqueand translucent endosperm NILs segregated into 1 homozygous opaque : 2 segregating : 1 homozygous translucent endosperm, andthis result indicates that the opaqueness of rice grain folows the single gene inheritance. Also genetic analysis showed that opaqueendosperm was recessive to translucence. Pedigree lines or F2 plants bearing opaque rice grain showed low amylose content(AC),while translucent grain showed high AC. It seems that the opaqueness of rice grain of a parental variety CB243 is closely relatedwith low AC.
In vitro culture of hybrid ovule between Fagopyrum esculentum Moench and F. homotropicum Ohnishi

Interspecific hybridization in buckwheat is of great importance to transfer desirable genes from wild species to cultivars as well as to broaden genetic variation in cultivars. In vitro ovule culture following the bud pollination was made to overcome postzygotic cross/self-incompatibility in interspecific hybridization between Fagopyrum esculentum Moench (common heterostylous buckwheat) and F. homotropicum Ohnishi (homostylous buckwheat). The percentage of fertilized ovaries of F. esculentum flowers (thrum-type) pollinated with F. homotropicum was 50%, while that of the reciprocal cross was as low as 10% and their ovaries were smaller. Vigorous pollen tube elongation following the bud pollination indicated the possibility for overcoming of breeding barriers in interspecific crosses between buckwheat species. Pre-embryo formation was observed 48 hours after pollination and ovules cultured up to 5 days after pollination tended to develop callus. But ovules isolated 11 days after pollination had apparently lost their viability. Hybrid plantlets were produced from only the ovules dissected 5 days after bud pollination. Regeneration rate was better in ovule culture on White medium than other mediums tested. Interspecific hybrid plants between F. esculentum and F. homotropicum were regenerated with the rate of 24.2% in average.
