      • In vitro 방법에 의한 천연물의 지용성분 중 면역활성제 개발 연구

        To develope activating agents of immune system from natural products, we focused on the activation of T and macrophage cells. We chose 35 kinds of candidates to do this. We used in vitro method such as Fetal Thymic Organ Culture(FTOC) system and bacterial phagocytic method to do test the inducing ability for T cell activation and phagocytosis of peritoneal macrophages. The results obtained from above way are as follows: Atractylis Rhizoma, Japanese touchwood and Astragali Radix showed T cell activating effect, particullary Astragali Radix is affecting most strongly between these 3 samples. Wherease Polyporus and corni Fructus which is known to be T cell activator are affecting on the decrease of fetal thymic T cells. Malvae Semer used as negative control showed predicted results. From the flow cytometric analysis, the results from Atractylis Rhizoma, Japanese Touchwood. Astragali Radix, Polyporus and Corni Fructus showed large population of CD4 positive helper T cells. Malvae Semer affect on the increasing of CD8 positive T cells, as opposed to the samples known to be T cell activating agents. From the analysis of phagocytic activityof peritoneal macrophage cells, Angelicae Gigantis Radix, Polyporus and Codonopsi Radix showed the increasing effect on phagocytic activity. Codonopsi Radix is particullary affecting most powerfully. Phellodendri Cortex which is known to be the activating agent on macrophage showed negative effect.

      • 해바라기, 아몬드, 땅콩의 給與가 생쥐의 增體量 臟器重量 및 血液像에 미치는 影響

        本 試驗은 해바라기, 아몬드 및 땅콩의 種實을 기초사료에 5%수준으로 혼합하여 생쥐에 給與하였을 때 미치는 飼養成績, 臟器重量 및 血液像등을 調査한 結果를 要約하면 다음과 같다. 1. 終了時 體重은 아몬드나 땅콩을 혼합하여 給與하였을 때가 해바라기를 給與하였을 때보다 암수 共히 높았다(p<.05) 2. 內部臟器의 重量에서 肝臟과 腎臟은 해바라기, 아몬드를 給與한 생쥐가 암수 共히 基礎飼料 給與區보다 높았다. 3. RBC, Hb, PCV, MCV 및 MCHC는 해바라기나 아몬드 및 땅콩의 給與에 따른 뚜렷한 影響이 없었고 WBC는 해바라기를 給與하였을 때 다른 處理區보다 높았다. 4. 해바라기를 給與하였을 때 r-GTP와 Total cholesterol含量은 암수 共히 높았으나 BUN含量은 낮았다. This experiment was conducted to investigate the effect of substituting ration with sunflower seed, almond and peanut on the performance, internal organs weight and hemocytological constitution of mouse The cxperimental design was divided into 4 treatments: control, 3 experimental groups added 5% sunflower seed, 5% almond, and 5% peanut to 95% basic diets of mouse The results were summarized as follows: The final body weight of mouse fed the diets substituting ration with almond or peanut seed was significantly(P<0.05) higher compared to those of mouse fed sunflower seed, In the internal organs weight, liver weight or kidney weight of mouse fed the diets sugstituting ration with sunflower or almond seed were higher the those of mouse fed the basic diets There were no difference in RBC, Hb, PCV, MCV, and MCHC ansong mice fed sunflower, almond, and peanut. But WBC in mouse fed sunflower seed was higher the that ao the other treantment groups r-GTP and total cholesterol level of mouse fed sunflower seed were high, but BUN was low

      • Activation of Fc Epsilon RI Inhibits the Pyruvate Kinase through Direct Interaction with the Gamma-Chain

        The downstream signaling components of high-affinity IgE receptor (FcεRl) were studied using yeast two-hybrid screening of the cDNA library constructed from RBL-2H3 cells. The cytoplasmic part of the γ-chain but not that of the β-chain was found to interact with pyruvate kinase in the yeast. The in-vitro-translated pyruvate kinase also specifically interacted with the bacterially expressed glutathione-S transferase fusion protein of the cytoplasmic part of the γ-chain. When RBL-2H3 cells were challenged with antigen, the activity of pyruvate kinase gradually decreased, reaching the minimum activity around 5 min after the activation, and then slowly returned to the normal level. The dose-response curve (antigen vs. pyruvate kinase activity) plotted at 5 min after stimulation showed that the pyruvate kinase was dose-dependently inhibited and the maximum inhibition was reached at the concentration of 0.1 ㎍/㎖ of antigen. Direct interaction between FcεRl and pyruvate kinase was also demonstrated by co-immunoprecipitation in RBL-2H3 cells. These data suggest that pyruvate kinase is functionally linked with FcεRl and might exert an important role in controlling cellular functions following the activation of FcεRl.

        Voltage-gated $K^+$ channels contributing to temporal precision at the inner hair cell-auditory afferent nerve fiber synapses in the mammalian cochlea

        To perform auditory tasks such as sound localization in the space, auditory neurons in the brain must distinguish sub-millisecond temporal differences in signals from two ears. Such high temporal resolution is possible when each neuron in the ascending auditory pathway fires brief action potential at very accurate timing. Various pre- and postsynaptic machineries ensuring such high temporal precision of auditory synaptic transmission have been identified. Of particular, in this review, the role of $K^+$ channels in shortening the duration of synaptic potentials will be discussed. First, the contribution of $K^+$ channels to AP firing of general auditory neurons will be discussed. Then, the focus will be moved to the inner hair cell (IHC)-auditory afferent nerve fiber (ANF) synapses, the first synapses of ascending auditory pathway. Molecular and immunohistological techniques have revealed various $K^+$ channels in the cell bodies and their processes of ANFs. Since the development of patch-clamp recordings from the ANF dendrites in 2002, it became possible to monitor the IHC-ANF synaptic transmission in greater detail. As revealed in brain auditory synapses, several different $K^+$ channels appear to participate in reducing the duration of synaptic potentials at the IHC-ANF synapses. In addition, $K^+$ channels at the ANF dendrites might act as potential targets of efferent feedback from the brain. The hypothesis is that, upon loud sound exposure, efferent neurotransmitters released onto the ANF dendrites activate certain $K^+$ channels and prevent excitotoxicity of ANFs. Therefore, $K^+$ channels of the ANF dendrites might provide potential sites of pharmacological actions to prevent noise-induced hearing loss.

        Isolation and Activity Evaluation of Peptides with Anti-hypertensive Activity from Commercial Enzymatic Flounder Fish Hydrolysate

        The potent antioxidant and anti-hypertension activities have evidenced gastric enzymatic hydrolysates from flounder fish and their derived peptides. However, peptide composition and functional effect in various enzymatic hydrolysates differ by enzyme types, hydrolyzed times temperatures, etc. Therefore, we determined potential anti-hypertensive effect of hydrolysates produced from flounder fish using commercial enzymes such as Protamex, Flavourzyme, and Kojizyme which are common food grade proteases and characterized on its derived peptides. In this study, Protamex-mediated hydrolysate showed a more potent anti-hypertension effect than other commercial enzymes. Protamex-mediated hydrolysate was fractionated into three ranges of molecular weight (above 10 kDa (FPH-I), 5-10 kDa (FPH-II), and below 5 kDa (FPH-III)). The FPH-III exhibited the strongest anti-hypertensive effect, and it was revealed that three active peptides, valine-phenylalanine-serine-glycine-tryptophan-alanine-alanine (VFSGWAA), leucine-histidine-phenylalanine (LHF) and tryptophan-proline-tryptophan (WPW) were contained. The activities were confirmed via angiotensin-converting enzyme (ACE) inhibition and molecular docking simulation. Among the three peptides, LHF and WPW have a molecular structure stability against the gastrointestinal digestion. LHF showed a significant anti-hypertension effect at 9 h after oral administration in spontaneously hypertensive rats (SHRs). Therefore, we suggest that Protamex-mediated hydrolysate would be an excellent anti-hypertensive agent due to the existence of stabilized functional peptides, including LHF and WPW.

        The structure activity relationship of flavonoids for anti-allergic actions was studied by determining the IC_(50) values for the degranulation. The hexosaminidase release from RBL-2H3 cells (degranulation marker) was employed as an estimate for the anti-allergic actions. Among 22 flavonoid compounds tested, luteolin, apigenin, diosmetin, fisetin, and quercetin were found to be most active with IC_(50) values less than 10 μM.

        비상시 수계전환 수리해석을 위한 A-PDA모형의 적용

        상수관망 시스템은 중요 사회기반시설로 안정적이고 안전한 공급을 추구하나, 내/외적인 요인들로부터 발생하는 각종 비정상상황으로부터 자유롭지 못하다. 비정상상황으로 인한 용수공급에 차질이 생길 시, 수계전환에 따른 비상급수는 매우 효율적인 대비책 중 하나로서 수리해석을 통해 공급능력 평가 및 관망의 설계 보완 방안을 제시할 수 있다. 하지만 기존의 수리해석 모형들은 무한수원의 가정을 적용하고 있는 바, 비상연계 수리해석 시 비현실적인 결과를 도출할 수 있다. 이에 배수지의 저류량과 유입량을 고려한 Advanced-Pressure Driven Analysis(A-PDA) 모형을 적용하여 A시의 수계전환 시나리오를 모의하였으며, 기존 PDA 모형의 결과와의 비교를 통하여 A-PDA 모형이 현실적인 결과를 도출하였음을 확인하였다. The water distribution system is an important social infrastructure providing customers with stable and safe water supply. The fact, however, is that it is not free from various abnormal situations arising from internal / external factors. In case of a water supply failure due to an abnormal situation, the emergency interconnected operation between adjacent blocks in the system is one of the most effective countermeasures. To simulate these types of emergency operations, existing hydraulic analysis model which are based on infinite source assumption may result in unrealistic results such as negative pressure at demand nodes. Having more realistic simulation results from these situations, it is required to use an A-PDA model considering the reservoir volume and the inflow volume of the reservoir. With this consideration, the A-PDA model can simulate the condition of the limited amount of water supply. We apply the A-PDA model to the emergency interconnected operation between adjacent blocks in a city in Korea and find that more realistic results can obtained comparing with the results obtained from the PDA model which is based on the unlimited water supply.

        Anatomical Correlates of Neuropsychological Deficits Among Patients With the Cerebellar Stroke

        <P><B>Objective</B></P><P>To investigate the anatomical correlates of the neuropsychological deficits in patients with the cerebellar stroke.</P><P><B>Methods</B></P><P>We screened patients who were admitted to the National Rehabilitation Center with the cerebellar stroke between October 2012 and November 2016. The patients with the cerebellar stroke who underwent neuropsychological testing for which the Seoul Neuropsychological Screening Battery (SNSB) or the SNSB-II were enrolled. The neuropsychological function capacities were compared in accordance with the stroke type (hemorrhagic vs. ischemic) and the location (right/left anterior, right/left posterior intermediate, right/left posterior lateral lobe, and vermis). Mean z-scores were computed to compare the patient performances with the population averages.</P><P><B>Results</B></P><P>Twenty-six patients (15 with ischemic stroke and 11 with hemorrhagic stroke) with a mean age of 54.8±16.6 years were assessed 8.8±9.2 months after the stroke. Differences in the neuropsychological functioning according to the stroke type were not observed. All of the numerical subtests of the stroke patients showed significantly poorer performances compared with the population averages (mean z-score <0), and some of the subtests revealed abnormal performances in attention-, visuospatial function-, memory-, and frontal/executive function-related tasks (mean z-score <−1). The patients with the presence of a lesion in the right posterior intermediate lobe of the cerebellum showed a poorer performance in the subtests evaluating the executive function including the Korean-version Stroop Test (p=0.04), the Digit Symbol Coding Test (p=0.01), and the Korean-version Trail Making Test (p=0.02) compared with the patients without that lesion.</P><P><B>Conclusion</B></P><P>The present study confirms that the cerebellar stroke affects the neuropsychological functioning which is associated with the anatomical site of stroke.</P>
