Characterization and genetic analysis of a plastochron 1-6 mutant in rice.

Plastochron and phyllotaxy are important traits to determine plant architecture. A plastochron 1-6 mutant was derived from japonica rice cultivar Koshihikari treated with ethylmethane sulfonate (EMS). The plastochron 1-6 mutant showed reduced plant height, shorter internode length, smaller and more leaves than Koshihikari, its wild type. In addition, the mutant has abnormal panicle, abnormal seed and upper node tillers. The F1 plants between mutant and Koshihikari were normal. In F2 population, segregation ratio between the wild type and mutant was fitted to 3:1. This genetic analysis indicated that plastochron 1-6 is controlled by a single recessive gene. Bulked segregant analysis revealed that the gene was located on chromosome 10. Through sequencing analysis, we found that plastochron 1-6 mutant had a single nucleotide transition occurred in the first exon of LOC_Os10g26340 (encoding P450 CYP78A11). A study on low-velocity impact damage of Z-pin reinforced laminates

Based on a low-velocity impact test, four main modes of low-velocity impact damage, including matrix cracking, de-laminating, fiber failure and matrix crushing, are taken into account. By using the proper failure criterion, the low-velocity impact damage of z-pin reinforced laminates can be realized. The results of FEM simulation, which indicate that a z-pin makes the area of delamination reduced by approximately 50%, are in good agreement with the experimental C-scan results.

Background: The calcium-binding S100A4 protein is involved in epithelial to mesenchymal transition, oncogenic transformation, angiogenesis, cytoskeletal integrity, mobility and metastasis of cancer cells. This study aimed to clarify the roles of S100A4 in genesis and progression of glioma. Materials and Methods: S100A4 expression was examined by real-time RT-CPR and Western blot in glioma and paired normal brain tissue (n=69), and compared with clinicopathological parameters of tumors. In addition, glioma U251 cells transfected with an S100A4-expressing plasmid were examined for proliferation by MTT, apoptosis by Annexin V-FITC, and migration and invasion with Transwell chambers. Results: Increased S100A4 mRNA expression was found in gliomas, compared with paired non-tumor tissue (p<0.001). Gradual elevation of overexpression of S100A4 was observed with increasing glioma grade (p<0.001). Astrocytoma showed lower S100A4 mRNA expression than oligodendrogliomas, with glioblastomas having highest values (p<0.001). Similar results were obtained for S100A4 protein, a positive link being found between mRNA and protein expression in gliomas (p<0.001). There was higher growth, lower apoptosis, stronger migration and invasion of S100A4 transfectants than control and mock transfected cells (p<0.001). Conclusions: These findings indicate that up-regulated S100A4 expression is positively linked to pathogenesis, progression and histogenesis of glioma by modulating proliferation, apoptosis, migration and invasion.
Application of Human Papillomavirus in Screening for Cervical Cancer and Precancerous Lesions

Cervical cancer is a commonly-encountered malignant tumor in women. Cervical screening is particularly important due to early symptoms being deficient in specificity. The main purpose of the study is to assess the application value of cervical thinprep cytologic test (TCT) and human papillomavirus (HPV) detection in screening for cervical cancer and precancerous lesions. In the study, cervical TCT and HPV detection were simultaneously performed on 12,500 patients selected in a gynecological clinic. Three hundred patients with positive results demonstrated by cervical TCT and/or HPV detection underwent cervical tissue biopsy under colposcopy, and pathological results were considered as the gold standard. The results revealed that 200 out of 12,500 patients were abnormal by TCT, in which 30 cases pertained to equivocal atypical squamous cells (ASCUS), 80 cases to low squamous intraepithelial lesion (LSIL), 70 cases to high squamous intraepithelial lesion (HSIL) and 20 cases to squamous cell carcinoma (SCC). With increasing pathological grade of cervical biopsy, however, TCT positive rates did not rise. Two hundred and eighty out of 12,500 patients were detected as positive for HPV infection, in which 50 cases were chronic cervicitis and squamous metaplasia, 70 cases cervical intraepithelial neoplasia (CIN) I, 60 cases CIN II, 70 cases CIN III and 30 cases invasive cervical carcinoma. Two hundred and thirty patients with high-risk HPV infection were detected. With increase in pathological grade, the positive rate of high-risk HPV also rose. The detection rates of HPV detection to CIN III and invasive cervical carcinoma as well as the total detection rate of lesions were significantly higher than that of TCT. Hence, HPV detection is a better method for screening of cervical cancer at present.

The tensile properties and microstructure of extruded and cold rolled 2A55 Al–Li alloys with different Cu content werestudied. After solution treatment, the strength of extruded sheets is slightly higher than that of cold rolled sheets due to strongdeformation texture and high density of substructure. However, the strength change order is different with the increase of Cucontent with T6-24 h. The extruded sample with higher Cu content have higher strength due to many dense T1precipitates. The δ′/θ′/δ′ composite precipitates and θ′ precipitates suppress the formation of T1precipitates, which cause cold rolled samplewith higher Cu content shows lower strength. Additionally, many fine T1precipitates nucleate at the sub-grain boundariesformed in the extrusion process, which have a significant effect on the mechanical property.

Acute respiratory distress syndrome is a life-threatening critical syndrome resulting largely from the accumulation of and the inability to clear pulmonary edema. Protectin DX, an endogenously produced lipid mediator, is believed to exert anti-inflammatory and pro-resolution effects. Protectin DX (5 μg/kg) was injected i.v. 8 h after LPS (14 mg/kg) administration, and alveolar fluid clearance was measured in live rats (n = 8). In primary rat ATII epithelial cells, protectin DX (3.605 × 10−3 mg/l) was added to the culture medium with LPS for 6 h. Protectin DX improved alveolar fluid clearance (9.65 ± 1.60 vs. 15.85 ± 1.49, p < 0.0001) and decreased pulmonary edema and lung injury in LPSinduced lung injury in rats. Protectin DX markedly regulated alveolar fluid clearance by upregulating sodium channel and Na, K-ATPase protein expression levels in vivo and in vitro. Protectin DX also increased the activity of Na, K-ATPase and upregulated P-Akt via inhibiting Nedd4–2 in vivo. In addition, protectin DX enhanced the subcellular distribution of sodium channels and Na, K-ATPase, which were specifically localized to the apical and basal membranes of primary rat ATII cells. Furthermore, BOC-2, Rp-cAMP, and LY294002 blocked the increased alveolar fluid clearance in response to protectin DX. Protectin DX stimulates alveolar fluid clearance through a mechanism partly dependent on alveolar epithelial sodium channel and Na, K-ATPase activation via the ALX/PI3K/Nedd4–2 signaling pathway.

Dichlorodiphenoxytrichloroethane (DDT), a persistent organic pollutant and hepatotoxicant, is used to control the malaria. However, scarce information exists on relevant effective inhibitors of DDT’ toxicity. The aim of this study was to investigate protective effects of natural antioxidants vitamin C (VC) or/and vitamin E (VE) on p,pʹ-DDT-induced genotoxicity and hepatotoxicity in human liver cells. p,pʹ-DDT exposure increased levels of chromatin condensation, comet parameter, micronucleus induction and DPC coefficient of HL-7702 cells in a dose-dependent manner. Also, an increase in mRNA levels of CYP1A1 as well as CYP3A4, and a decrease in UGT along with GST were observed. Interestingly, supplementation with VC or/and VE prevented p,pʹ-DDT-caused alterations in DNA damage and phase metabolism gene expressions, and the combination of VC and VE had a more protective effect than VC or VE alone. These findings illustrated that VC or/and VE could be beneficial for the alleviation of p,pʹ-DDT-incurred toxicity in human liver cells.
Characterization and genetic mapping of white spotted leaf (wspl) mutant in rice

Spotted leaf mutants which produce necrotic lesions spontaneously are important sources to study programmed celldeath in plant defense responses. A novel white-spotted leaf (wspl) mutant was induced from Ilpum, Korean japonica rice cultivar bythe treatment of ethyl methane sulfonate (EMS). The phenotype of wspl mutant differed from that of other spotted leaf mutants in thatnot only brown spots but also white lesion mimic spots were observed on the tip of the leaves from the vegetative stage. Strong NBT andDAB staining were observed on the older leaf of wspl mutant in microscopic ROS assay, and the chlorophyll content of wspl mutantmaintained longer than wild-type in the old leaves. Genetic analysis revealed that the wspl mutant trait was controlled by a singlerecessive gene and the locus of wspl gene was mapped on the long arm of chromosome 5 between the flanking markers S05100 andS05112 (4.1 Mb). Through the combination of the genetic mapping and SNP analysis, two candidate genes for white-spotted leaf wereidentified in the genic region. A novel phenotype of white-spotted leaf mutant has not yet been reported, thus further study of the wsplmutant will contribute to understanding of the molecular mechanisms involved in lesion mimic phenotype in rice.
