        MiRNA320a Inhibitor-Loaded PLGA-PLL-PEG Nanoparticles Contribute to Bone Regeneration in Trauma-Induced Osteonecrosis Model of the Femoral Head

        BACKGROUND: This study aimed to explore the effect of a nanomaterial-based miR-320a inhibitor sustained release system in trauma-induced osteonecrosis of the femoral head (TIONFH). METHODS: The miR-320a inhibitor-loaded polyethylene glycol (PEG)- Poly(lactic-co-glycolic acid) (PLGA)- Poly-L-lysine (PLL) nanoparticles were constructed using the double emulsion method. The TIONFH rabbit model was established to observe the effects of miR-320a inhibitor nanoparticles in vivo. Hematoxylin–eosin staining and microcomputed tomography scanning were used for bone morphology analysis. Bone marrow mesenchymal stem cells (BMSCs), derived from TIONFH rabbits, were used for in vitro experiments. Cell viability was determined using the MTT assay. RESULTS: High expression of miR-320a inhibited the osteogenic differentiation capacity of BMSCs in vitro by inhibiting the expression of the osteoblastic differentiation markers ALP and RUNX2. MiR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticles were constructed with a mean loading efficiency of 1.414 ± 0.160%, and a mean encapsulation efficiency of 93.45 ± 1.24%, which released 50% of the loaded miR-320a inhibitor at day 12 and 80% on day 18. Then, inhibitor release entered the plateau. After treatment with the miR-320a inhibitor nanoparticle, the empty lacunae were decreased in the femoral head tissue of TIONFH rabbits, and the osteoblast surface/bone surface (Ob.S/BS), osteoblast number/bone perimeter (Ob.N/B.Pm), bone volume fraction, and bone mineral density increased. Additionally, the expression of osteogenic markers RUNX2 and ALP was significantly elevated in the TIONFH rabbit model. CONCLUSION: The miR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticle sustained drug release system significantly contributed to bone regeneration in the TIONFH rabbit model, which might be a promising strategy for the treatment of TIONFH.         <P>A facile coagglomeration method for preparing a long alkyl chain modified graphene oxide (MGO)/MgCl2-supported Ti-based Ziegler-Natta catalyst was reported. The effects of MGO on the catalyst morphology and activity for ethylene polymerization were examined. The resultant polyethylene (PE)/MGO nanocomposites exhibited a layered morphology, with the MGO fillers being well dispersed and exhibiting strong interfacial adhesion to the PE matrix. The thermal stability and mechanical properties of the PE were significantly enhanced with the introduction of a small amount of the MGO filler. Thus, this work provides a facile approach to the production of high-performance PE.</P>

        Facile preparation of functionalized MoS<sub>2</sub>/polyethylene nanocomposites through in situ polymerization with MoS<sub>2</sub> containing Ziegler–Natta catalyst

        <P><B>Abstract</B></P> <P>A facile method for preparing polyethylene (PE)/octadecylamine-functionalized MoS<SUB>2</SUB> (ODA-MoS<SUB>2</SUB>) nanocomposites through in situ polymerization with an ODA-MoS<SUB>2</SUB>-MgCl-TiCl<SUB>4</SUB> catalyst was developed. The catalyst was synthesized simply by the treatment of ODA-MoS<SUB>2</SUB> with a Grignard reagent, followed by anchoring of a TiCl<SUB>4</SUB> catalyst. The resulting catalyst exhibited higher activity toward ethylene polymerization than did an ODA-MoS<SUB>2</SUB>-free catalyst. After polymerization, the resultant PE/ODA-MoS<SUB>2</SUB> nanocomposites reproduced the catalyst morphology, showing a flaked morphology. In addition, the thermal stability and mechanical properties of PE were significantly enhanced by the introduction of ODA-MoS<SUB>2</SUB> fillers.       • Historical Long-term Exposure to Pentachlorophenol Causing Risk of Cancer - A Community Study

        Background: Pervious studies suggested occupational workers exposure to pentachlorophenol (PCP) might contribute to increased risk of cancer. However, few studies have focused on associations between PCP and cancer risk at the community level. Objective: The present study was to explore the cancer risk for the community population living long-term in a PCP contaminated area. Methods: All the cancer cases diagnosed in 2009-2011 in Tongling City were collected. The cancer patients' residencies were geo-referenced in each district. The historical PCP usage for each district of Tongling was calculated as the PCP pollution index, which was further used to divide into PCP exposure categories. Standardized rate ratios (SRRs) of cancer incidence were applied to detect the cancer risk as exposure grade elevated. Correlation analysis was performed to analyze the relationship between PCP pollution and cancer incidence. Results: A total of 5,288 cancer cases (3,451 male and 1,837 female) were identified. PCP usage was correlated with the incidence of leukemia (r=0.88, P=0.002) for males, and with cancer of the esophagus for males (r=0.83, P=0.008) and females (r=0.71, P=0.020). Compared with the low exposure category, significant SRRs for total cancer sites was obtained for high PCP exposure category (SRR=1.61, 95%CI=1.59-1.62). Most SRR values of the cancer sites were significantly increased as exposure grade elevated and exposure time extended. Conclusion: The present study found that community residents living in the PCP contaminated area had increased risk of cancers. Leukemias, lymphomas and nasopharyngeal and esophageal cancers are most possibly associated with PCP exposure.

      • Prognostic Values of Various Clinical Factors and Genetic Subtypes for Diffuse Large B-cell lymphoma Patients: A Retrospective Analysis of 227 Cases

        Aim: To analyze the significance of different clinical factors for prognostic prediction in diffuse large B-cell lymphoma (DLBCL) patients. Methods: Two hundred and twenty-seven DLBCL patients were retrospectively reviewed. Patients were managed with cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) regimen or rituximab plus the CHOP (RCHOP) regimen. Results: Lactate dehydrogenase (LDH), ${\beta}2$-microglobulin (${\beta}2$-M), B symptoms, Ann Arbor stage and genetic subtypes were statistically relevant in predicting the prognosis of the overall survival (OS). In the CHOP group, the OS in patients with germinal center B-cell-like (GCB)(76.2%) was significantly higher than that of the non-GCB group (51.9%, P=0.032). With RCHOP management, there was no statistical difference in OS between the GCB (88.4%) and non-GCB groups (81.9%, P=0.288). Conclusion: Elevated LDH and ${\beta}2$-M levels, positive B symptoms, Ann Arbor stage III/IV, and primary nodal lymphoma indicate an unfavorable prognosis of DLBCL patients. Patients with GCB-like DLBCL have a better prognosis than those with non-GCB when treated with the CHOP regimen. The RCHOP treatment with the addition of rituximab can improve the prognosis of patients with DLBCL.

        Laboratory Confirmation of A Suspicious Meningococcal Meningitis Death Case

        A suspicious meningococcal meningitis death case was reported to the Beijing CDC. The blood specimen was analyzed via multi-PCR and MLST. 6 isolates from close contacts were analyzed via PFGE and MLST. According to the results of the above analyses, the cause of this case was identified as a serogroup A Neisseria meningitidis, which, in terms of sequence typing, belonged the ST7 group.

        Effects of Insect Hormones on the Replication of Nucleopolyhedrovirus

        An experimental study was undertaken to quantify the effects of insect hormones on the replication of nucleopolyhedrovirus (NPV). The results demonstrated that TCID/ sub 50/ at 72 h post-infection (hpi) rose systematically from 0.55$\times$10$^{8}$ /m1, for untreated cells, up to 1.67$\times$10$^{8}$ / ml at 3$\mu$g/ml, then dropped down to 1.45$\times$10$^{8}$ /m1 at 4 $\mu$g/ml, by adding ecdysone to the culture medium for Bm-N cells infected with a wild-type Bambyx mori. nucleopolyhedrovirus (BmNPV). The optimum enhancement of about 3 times on budded virus (BV) titer at 72 hpi was given at 3 $\mu$g/ml of ecdysone. While the polyhedra number had no obvious variation within the range of concentrations from 0 to 4 $\mu$g/ml. By addition of juvenile hormone analogue (JHA) into the media with this concentration range, the BmNPV TCID/ sub 50/ and polyhedra number at 72 hpi did not show significant changes. Also, the addition of either 3 $\mu$g/ml of ecdysone or 3 $\mu$g/ml of JHA to the culture media did not appear to affect the TCID/ sub 50/ and polyhedra number significantly in infected Sf-21 cells with the autographa californica nucleopolyhedrovirus (AcMNPV).

        Heterologous expression of two Physcomitrella patens group 3 late embryogenesis abundant protein (LEA3) genes confers salinity tolerance in arabidopsis

        Salinity stress is a major limiting factor in agriculture and adversely affecting the whole plant. As a halophyte, the moss Physcomitrella patens, has been suggested to be an ideal model plant to study salinity tolerance and adaption. Two abiotic stress-responsive Group 3 Late Embryogenesis Abundant protein genes had been identified in P. patens and named as PpLEA3-1 and PpLEA3-2, respectively. Functions of these two genes were analyzed by heterologous expressions in Arabidopsis, driven either by their native P. patens promoters or by the 35S CaMV constitutive promoter. Phenotype analysis revealed that pLEA3::LEA3, pLEA3::LEA3::GFP and 35S::LEA3::GFP transgenic lines had stronger salinity resistance than that in the wild type and empty-vector control. Further analysis showed that the contents of proline and soluble sugar were increased and the malondialdehyde (MDA) were repressed in these transgenic plants after exposure to salinity stress. Our observations indicate that these two Group 3 PpLEA genes played a role in the adaption to salinity stress.

        Heavy Metals Immobilization in Soil with Plant-growth-promoting Rhizobacteria and Microbial Carbonate Precipitation in Support of Radish Growth

        The application of plant-growth-promoting rhizobacteria (PGPR) supports the growth of plants in contaminated soil while ureolytic bacteria can immobilise heavy metals by carbonate precipitation. Thus, dual treatment with such bacteria may be beneficial for plant growth and bioremediation in contaminated soil. This study aimed to determine whether the PGPR Pseudomonas fluorescens could work in synergy with ureolytic bacteria to assist with the remediation of cadmium (Cd)- and lead (Pb)-contaminated soils. Pot experiments were conducted to grow radish plants in Cd- and Pb-contaminated soils treated with PGPR P. fluorescens and the results were compared with dual inoculation of P. fluorescens combined with ureolytic Staphylococcus epidermidis HJ2. The removal rate of the metals from the soil was more than 83% for Cd and Pb by the combined treatment compared to 17% by PGPR alone. Further, the dual treatment reduced the metal accumulation in the roots by more than 80%. The translocation factors for Cd and Pb in plant tissues in both treatments remained the same, suggesting that PGPR combined with the carbonate precipitation process does not hamper the transfer of essential metal ions into plant tissues from the soil.
