<P><B>Purpose:</B></P><P>Given that osimertinib is the only approved third-generation EGFR-TKI against <I>EGFR</I> activating and resistant T790M mutated non–small cell lung cancer (NSCLC), additional mutant-selective inhibitors with a higher efficacy, especially for brain metastases, with favorable toxicity profile are still needed. In this study, we investigated the antitumor efficacy of YH25448, an oral, mutant-selective, irreversible third-generation EGFR-TKI in preclinical models.</P><P><B>Experimental Design:</B></P><P>Antitumor activity of YH25448 was investigated <I>in vitro</I> using mutant <I>EGFR</I>-expressing Ba/F3 cells and various lung cancer cell lines. <I>In vivo</I> antitumor efficacy, ability to penetrate the blood–brain barrier (BBB), and skin toxicity of YH25448 were examined and compared with those of osimertinib using cell lines and PDX model.</P><P><B>Results:</B></P><P>Compared with osimertinib, YH25448 showed a higher selectivity and potency in kinase assay and mutant <I>EGFR</I>-expressing Ba/F3 cells. In various cell line models harboring <I>EGFR</I> activating and T790M mutation, YH25448 effectively inhibited EGFR downstream signaling pathways, leading to cellular apoptosis. When compared <I>in vivo</I> at equimolar concentrations, YH25448 produced significantly better tumor regression than osimertinib. Importantly, YH25448 induced profound tumor regression in brain metastasis model with excellent brain/plasma and tumor/brain area under the concentration–time curve value. YH25448 rarely suppressed the levels of p-EGFR in hair follicles, leading to less keratosis than osimertinib in animal model. The potent systemic and intracranial activity of YH25448 has been shown in an ongoing phase I/II clinical trial for advanced <I>EGFR</I> T790M mutated NSCLC (NCT03046992).</P><P><B>Conclusions:</B></P><P>Our findings suggest that YH25448 is a promising third-generation EGFR inhibitor, which may be more effective and better tolerated than the currently approved osimertinib.</P>

The luminescence intensity of $Tb^{3+}$ bound to picolinic acid, squaric acid, and 2,3-pyrazinedicarboxylic acid has been studied in water and water-ethanol mixtures. It has been found that the emission intensity of $Tb^{3+}$ in the complexes is enhanced with the increase of ethanol content in water-ethanol mixtures. Several factors affecting on the emission intensity of $Tb^{3+}$ in waterethanol mixtures are discussed.
Pore characteristics of Lotus‑Type Porous Copper Fabricated by Centrifugal Casting

Lotus-type porous copper was fabricated by a centrifugal casting. The long cylindrical pores aligned along single directionwas formed during unidirectional solidification in a hydrogen atmosphere under a centrifugal force. The relationship betweenprocessing parameters and pore characteristics in centrifugal casting was investigated in detail. Porosity and average porediameter increased with increasing distance from bottom of ingot and rotational velocity. The processing parameter in centrifugalcasing affected the pressure on liquid/solid interface and the solidification rate during unidirectional solidification. It is concluded that an appropriate pore characteristics of lotus-type porous copper can be obtained by control of processingparameters in the centrifugal casting.

Tyrophagus putrescentiae (Tp) as a storage mite inhabitats in stored grains, hay, and straw at agricultural areas. T. putrescentiae stimulates an immune response and triggers inflammatory cytokines release, and thus it is a source of allergen that sensitize and induce allergic reactions. Also, T. putrescentiae has been reported to cause asthma and atopic disease by cross-reactivity with Dermatophagoides pteronyssinus (Dp). The study on T. putrescentiae in human monocytic THP-1 cells is not enough to understand cytokine expression and pathological mechanisms. The aim of this study is to investigate the effect of T. putrescentiae extract (TpE) on production of inflammatory cytokines and expression of mRNA level in THP-1 cells. THP-1 cells are treated with TpE and supernatants were analyzed for the production of cytokines using enzyme-linked immunosorbent assay (ELISA). mRNA level in the culture cells was measured by a reverse transcriptase-polymerase chain reaction (RT-PCR). As a result of this study, TpE significantly induced secretion of interleukin-6, interleukin-8, and monocyte chemotactic protein-1 (MCP-1) in THP-1 cells in time- and dose-dependent manner. These results suggest that TpE may play a role in contributing to inflammatory disease through stimulation of immune cell. Further research of T. putrescentiae is needed to understand the elucidation of the pathogenic mechanism.

With power converters are applied to various kinds of production line, there are increasing in load that have need of consecutive operation from low speed to high speed. Thus, interests in power converter which is able to consecutive drive are increase.<br/> For forced commutated cycloconverter(FCC) which does not require a dc link and can be used with most AC loads type of variable frequency and be able to AC-AC direct conversion, we consider cycloconverter system that can control from 10 Hz to 500 Hz using Power-MOSFET.<br/> As a result, we can vary frequency from low frequency (10 Hz) to high frequency (500 Hz) and its voltage amplitude linearly.<br/>
