        Characterization of mitochondrial genomes of three new species: Leptocimbex praiaformis, L. clavicornis, and L. yanniae (Hymenoptera: Cimbicidae)

        Three new species of Leptocimbex, L. praiaformis, L. clavicornis, and L. yanniae, from China were described and illustrated. Nearly complete mitochondrial genomes were sequenced using next-generation sequencing. Compared to the ancestral organization, four tRNA genes were rearranged: trnM (+) and trnQ () are shuffled, while trnC ()andtrnY () are translocated from the trnW-trnC-trnY cluster to a location upstream of trnI. By mapping gene order onto phylogenies estimated from the mitochondrial genome, the evolutionary histories of gene rearrangement traits can be deciphered. All protein-coding genes are initiated by ATN codons and terminated with TAN, or T codons. By labeling the mutation on predicted secondary structures of rRNAs, the highly conserved motifs of rrnS (H769, H921, H939, H1506) also show phylogenetic signals. Phylogenetic relationships among the Symphyta were reconstructed using mitogenomes. Both maximum likelihood and Bayesian inference analyses highly support the monophyly of the genus Leptocimbex and the sister-group relationship between Leptocimbex and Labriocimbex + Trichiosoma.

        miR-195/miR-497 Regulate CD274 Expression of Immune Regulatory Ligands in Triple-Negative Breast Cancer

        Purpose: Immune suppression is common in patients with advanced breast cancer but the mechanisms underlying this phenomenon have not been sufficiently studied. In this study, we aimed to identify B7 family members that were able to predict the immune status of patients, and which may serve as potential targets for the treatment of breast cancer. We also aimed to identify microRNAs that may regulate the expression of B7 family members. Methods: The Cancer Genome Atlas data from 1,092 patients with breast cancer, including gene expression, microRNA expression and survival data, were used for statistical and survival analyses. Polymerase chain reaction and Western blot were used to measure messenger RNA and protein expression, respectively. Luciferase assay was used to investigate direct microRNA target. Results: Bioinformatic analysis predicted that microRNA (miR)-93, miR-195, miR-497, and miR-340 are potential regulators of the immune evasion of breast cancer cells, and that they exert this function by targeting CD274, PDCD1LG2, and NCR3LG1. We chose CD274 for further investigations. We found that miR-195, miR-497, and CD274 expression levels were inversely correlated in MDA-MB-231 cells, and miR-195 and miR-497 expressions mimic inhibited CD274 expression in vitro. Mechanistic investigations demonstrated that miR-195 and miR-497 directly target CD274 3´ untranslated region. Conclusion: Our data indicated that the level of B7 family members can predict the prognosis of breast cancer patients, and miR-195/miR-497 regulate CD274 expression in triple negative breast cancer. This regulation may further influence tumor progression and the immune tolerance mechanism in breast cancer and may be able to predict the effect of immunotherapy on patients.

        Dual stimuli-responsive polymeric prodrug consisting of reversible covalent bonded celastrol for tumor targeted delivery

        In this work, pH/GSH-responsive amphiphilic polymeric prodrug (EDA-GLA/CE/2-FPBA) was successfully prepared and could self-assembled into micelles in an aqueous solution. The EDA-GLA/CE/2-FPBA micelles possessed high stability in physiological condition and were pH and GSH sensitive due to the reversible borate ester bonds and disulfide bonds within the prodrug polymer. The structures of the prodrug polymers were characterized by NMR, FTIR, UV–vis spectroscopy. Transmission electron microscopy and dynamic light scattering measurement indicated that the resulting micelles have desirable size distribution and regular spherical shape. Free active Celastrol can be released under low pH and high GSH environment; In vitro cellular uptake and growth inhibition assays suggested that the blank polymer micelles showed good biocompatibility. EDA-GLA/CE/2-FPBA micelles were more efficiently internalized by monolayer tumor cells and demonstrated superior tumor targeting effects as compared to free Celastrol control. These results demonstrated that the novel prodrug self-assembled dual-responsive nano-delivery platform was able to improve the bioavailability and tumor targeting activity of Celastrol, which provides a basis for further clinical applications of Celastrol and its derivatives.

        Nest-like Ag-doped NiMoO4/NF with rich oxygen vacancies as robust catalysts for highly efficient oxygen evolution

        The development of low-cost and highly efficient electrocatalysts for oxygen evolution reaction (OER)with good conductivity, high specific surface area and abundant oxygen vacancies is of great significancefor the future renewable energy conversion and storage system. Herein, Ag doped NiMoO4 materials onthe Ni foam (NiMoO4/Ag/NF) with rich oxygen vacancies were successfully prepared by hydrothermalstrategy and layer by layer assembly method. The Ag ions are doped in-situ to promote electron transferand compensate for the poor conductivity of metal oxides. The suitable air annealing treatment is used tocreate rich oxygen vacancies. The high specific surface area and one-dimensional characteristics of thecross mesoporous nanorod skeleton are responsible for the increased exposure of active centers andthe rapid charge transfer behavior. The NiMoO4/Ag/NF exhibits high activity for OER in alkaline solution(an overpotential of 275 mV at the current density of 20 mA cm2) and low Tafel slope (53.67 mV dec1),and a satisfying durability in 1.0 M KOH. This study provides an effective method for the preparation ofAg doped one-dimensional nanomaterials with excellent OER catalytic performance and electrochemicalstability.

        Neural adaptive pointing control of a moving tank gun with lumped uncertainties based on dynamic simulation

        This study focuses on the pointing control problem of a moving tank gun. Model uncertainty and foundation vibration, which may be nonlinear, coupled, or time-varying but bounded, are considered. First, the electrohydraulic servo system of a vertical stabilizer is constructed as a nonlinear dynamic system with lumped uncertainties. Second, a neural adaptive controller is proposed to improve the control performance of the vertical stabilizer. A backpropagation neural network is introduced to compensate for the uncertainties, and its weight and threshold values are self-tuned online. Third, a co-simulation model of the moving tank is established. Dynamic simulation verifies that the proposed controller exhibits better performance than typical controllers. Finally, the influence of hull foundation vibration on the proposed controller is analyzed. The pointing accuracy of a moving tank gun is verified to be controlled effectively by the proposed controller under different driving conditions. This work combines control theory with multi-body dynamics to provide a feasible solution for the pointing control problem of a moving tank with model uncertainty and foundation vibration.
