        MTA1 Overexpression Induces Cisplatin Resistance Innasopharyngeal Carcinoma by Promoting Cancer Stem Cells Properties

        The metastasis-associated gene 1 (MTA1) oncogene hasbeen suggested to be involved in the regulation of cancer progression. However, there is still no direct evidence that MTA1 regulates cisplatin (CDDP) resistance, as well as cancer stem cell properties. In this study, we found that MTA1 was enriched in CNE1/CDDP cells. Knock down of MTA1 in CNE1/CDDP cells reversed CSCs properties and CDDP resistance. However, ectopic expression of MTA1 in CNE1 cells induced CSCs phenotypes and CDDP insensitivity. Interestingly, ectopic overexpression of MTA1-induced CSCs properties and CDDP resistance were reversed in CNE1 cells after inhibition of PI3K/Akt by LY294002. In addition, MTA1 expression and Akt activity in CNE1/CDDP cells was much higher than that in CNE1 cells. These results suggested that MTA1 may play a critical role in promoting CDDP resistance in NPC cells by regulatingcancer stem cell properties via thePI3K/Akt signaling pathway. Our findings suggested that MTA1 may be a potential target for overcoming CDDP resistance in NPC therapy.

        Pathogen-Imprinted Polymer Film Integrated probe/Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub> MXenes Electrochemical Sensor for Highly Sensitive Determination of Listeria Monocytogenes

        As one of the most hazardous and deadliest pathogens, Listeria monocytogenes (LM) posed various serious diseases to the human being, thus designing effective strategy for its detection is of great significance. In this work, by preparing Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub> MXenes nanoribbon (Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub>R) as carrier and selecting thionine (Th) acted simultaneously as signal probe and functional monomer, a LM pathogen-imprinted polymers (PIP) integrated probe electrochemical sensor was design to monitor LM for the first time, that was carried out through the electropolymerization of Th on the Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub>R/GCE surface in the existence of LM. Upon eluting the templates from the LM imprinted cavities, the fabricated PIP/Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub>R/GCE sensor can rebound LM cells effectively. By recording the peak current of Th as the response signal, it can be weakened when LM cell was re-bound to the LM imprinted cavity on PIP/Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub>R/GCE, and the absolute values of peak current change increase with the increasement of LM concentrations. After optimizing three key parameters, a considerable low analytical limit (2 CFU mL<sup>-1</sup>) and wide linearity (10-10<sup>8</sup> CFU mL<sup>-1</sup>) for LM were achieved. In addition, the experiments demonstrated that the PIP/Ti<sub>3</sub>C<sub>2</sub>T<sub>x</sub>R sensor offers satisfactory selectivity, reproducibility and stability.

        METHOD OF HMI OPTIMIZATION DESIGN BASED ON FIXATION TRANSITION CHARACTERISTICS AND VISUAL ATTENTION TRAJECTORY: A DRIVER SIMULATOR STUDY

        To further enhance the role of connected vehicles with the Human-Machine Interface (HMI) in helping drivers in foggy weather, it is meaningful to optimize the HMI based on the driver’s needs. In order to explore the method of HMI optimization, this paper builds a driving simulation experimental test platform of freeway connected vehicle system, designing two experimental scenarios according to the technical conditions (without HMI or with HMI). After that, the paper uses Markov chain to explore the drivers’ fixation transition to identify the driver’s needs in different sections at different conditions. Besides, combined with the visual trajectory results, the paper provides suggestions for optimizing HMI. The results show the review rate of drivers in each section for the straight upper front area is very high. The highest value occurs in the heavy fog zone, suggesting that HMI can provide more road information. HMI should offer a prompt to the driver before entering the warning zone, and remind the driver of changes in the speed limit before entering the fog area. The prompt module of HMI should shorten the horizontal length. In conclusion, this paper aims to propose a general diagnosis method by diagnosing the self-designed HMI.

        Manufacturing of 3D Microlens Array Mold on Bulk Metallic Glass by Self-Aligned Multi-Ball Hot Embossing

        Bulk metallic glasses (amorphous alloys) exhibiting both high hardness and extraordinary thermoplastic forming ability, are perfect mold material for 3D optical microlens array. However, conventional 3D micro fabrication for metallic glass requires Si templates which are extremely costly and eco-unfriendly to machine and demold. In this study, we report a green and lowcost self-aligned multi-ball hot embossing process to create 3D micro dimple array on metallic glass by replicating spherical crowns of self-aligned precision balls. A 3D metallic glass micro dimple array with aperture of 867.2 μm and surface roughness of 8.2 nm was fabricated in 30 s. Different from conventional hot embossing that material full-filled into mold, the new process enables micro dimple fabrication with different specifications by freely controlling the down displacement of balls. Therefore, the relationship between down displacement and depth of micro dimple was revealed through exploring the material flow behavior by a verified FE model. In addition, to test the feasibility of the fabricated metallic glass mold, a PMMA compound eye structure was embossed and its focusing and imaging performance were subsequently evaluated. This research demonstrates a new manufacturing process for rapid fabrication of 3D microlens arrays metallic glass mold in a neat and economic way.

      • Parameter Optimal Choice of Claw Pole Alternator based on Iron Loss Model

        Based on classical Berotti discrete iron loss calculation model, the iron loss analysis mathematical model of alternator was proposed in this paper. Considering characteristics of high speed and changing frequency of the alternator, Maxwell 3-D model was built to analyze iron loss corresponding to each running speed in alternator. Based on iron loss model of alternator at rated speed, the rotor claw pole size was made an optimization design. The optimization results showed that alternator's output performance had been improved. A new idea was explored in size optimization design of claw pole alternator.

        A triage strategy in advanced ovarian cancer management based on multiple predictive models for R0 resection: a prospective cohort study

        Objective: To present the surgical outcomes of advanced epithelial ovarian cancer (AEOC) since the implementation of a personalized approach and to validate multiple predictive models for R0 resection. Methods: Personalized strategies included: 1) Non-invasive model: preoperative clinico-radiological assessment according to Suidan criteria with a predictive score for all individuals. Patients with a score 0–2 were recommended for primary debulking surgery (PDS, group A), or otherwise were counseled on the choices of PDS, neoadjuvant chemotherapy (NAC, group B) or staging laparoscopy (S-LPS). 2) Minimally invasive model: S-LPS with a predictive index value (PIV) according to Fagotti. Individuals with a PIV <8 underwent PDS (group C) or otherwise received NAC (group D). Intraoperative assessment (with Eisenkop, peritoneal cancer index [PCI], and Aletti scores) and surgical results were prospectively collected. Results: Between September 2015 and August 2017, 161 pathologically confirmed epithelial ovarian cancer patients were included. A total of 52 (32.3%) patients had a predictive score of 0–2, and 109 (67.7%) patients had a score ≥3. Among these individuals, 41 (25.5%) patients received S-LPS. Finally, 110 (68.3%) patients underwent PDS (A+C), and 51 (31.7%) patients received NAC (B+D). The R0 resection rates in PDS and NAC patients were 56.4% and 60.8%, respectively. The area under the curve (AUC) of Suidan criteria was 0.548 for group (A+C). The AUC of Fagotti score was 0.702 for group C. The AUC of Eisenkop, PCI, and Aletti scores were 0.808, 0.797, and 0.524, respectively. Conclusion: The Suidan criteria were not effective in these AEOC patients. S-LPS was helpful in decision-making for PDS and should be endorsed in the future.

        Soft-switching buck inverter

        The soft-switching buck inverter, which is mainly applied to solve the contradictions between the switching frequency and the shoot-through problem along with the dead-time effect, is proposed in this paper. A detailed analysis of the relationship between the range of the duty ratio to realize soft-switching as well as the additional voltage stress and the current stress is conducted. In addition, a 1 kW prototype is designed and established. Results of simulations and experiments verify the validity of the theoretical analysis. Furthermore, the proposed soft-switching buck inverter has a strong practical significance in situations where medium and high power outputs are required.

        Resolution of Racemic Ketoprofen in Organic Solvents by Lipase from Burkholderia cepacia G63

        A lipase from the Burkholderia cepacia strain G63 immobilized on resin was used for the resolution of ketoprofen. To study its catalytic properties in enantioselective esterication, different alcohols and solvents were tested to select the most suitable acyl acceptor and reaction medium. Compared with the low activity of the free lipase, the enzyme activity and E value of the immobilized lipase were significantly enhanced. The enantioselectivity of the immobilized lipase could also be markedly improved by adding a small amount of 18-crown-6. RSM was employed to optimize the reaction parameters. The optimal reaction conditions were: reaction time 22.50 h, additives dosage 0.4322 g (0.33 mmol/mL), and substrate molar ratio 54.11:1. Under optimal conditions, the maximal E value was up to 10.01, which exhibited a better enantioselectivity than some commercial lipases, such as Novozym 435,Lipozyme RM IM and LipozymeTL IM.
