      • A quantitative shotgun proteomic study of rice grain development

        Although a great deal of rice proteomic research has been conducted, there are relatively few studies specifically addressing the rice grain proteome. The existing rice grain proteomic research has focused on the identification of deferentially expressed proteins. Here, we performed comparative shotgun proteomic analysis of rice grain development to construct an in-depth proteome reference map, to reveal the expression patterns of the identified proteins, and to detect proteins that are expressed deferentially during grain development. A Korean rice variety, Ilpumbyeo was used. Proteins were extracted from rice grains 10, 20, and 30 days after flowering, as well as from mature grains. The protein expression patterns were revealed by a quantitative shotgun proteoemic analysis. By merging all of the identified proteins in this study, we identified 4,172 non-redundant. A Genome Ontology category enrichment analysis for the 4,172 proteins revealed that 52 categories were enriched, including the carbohydrate metabolic process, transport, localization, lipid metabolic process, and secondary metabolic process. The relative abundances of the 1,784 reproducibly identified proteins were compared to detect 484 differentially expressed proteins during rice grain development. Clustering analysis and Genome Ontology category enrichment analysis revealed that proteins involved in the metabolic process were enriched through all stages of development, suggesting that proteome changes occurred even in the desiccation phase. Interestingly, enrichments of proteins involved in protein folding were detected in the desiccation phase and in fully mature grain.

      • Shotgun proteomics approach to identify candidate proteins related with drought stress in Brassica rapa ssp. pekinensis (inbred line ‘Chiifu’)

        As the drought is getting worse, Lot of studies related to drought stress in plant have been conducted. Recently whole genome sequencing of Brassica rapa ssp which is important vegetable crop to East Asians has been completed to enable Omic research. It is known that the drought damages occur in the early stage of plant development. Here, we performed shotgun proteomics analysis of B. rapa to observe the morphological characters, monitor the expression patterns of the identified proteins during drought stress, and detect the proteins related to drought stress. The three week old B. rapa grown in density of single plant in a single pot were used. Drought stress were treated as that a single plant in soil was removed from the pot and the plant with soil was exposed to air and light without watering. Leaves were immediately harvested before drought treatment, 24hr after drought treatment, and 48hr after drought treatment. The protein expression patterns were monitored by a quantitative shotgun proteomics analysis. Extracted proteins were separated in 1D-SDS-PAGE then the gel sliced into seven pieces. Chopped gels were ingel-digested. Peptides were assigned to mass spectrometry (Q-Exactive). The ms/ms spectra were analyzed through Proteome Discoverer. By combining all of the identified proteins in the seven sliced gel samples, total B. rapa proteome reference map was completed. Protein expression patterns were investigated by comparing the quantity of protein. With shotgun proteomic approach, we evaluated the changes in the quantity and finally discovered the candidate proteins related with drought stress.

        중단간, 만생 중간찰벼 ‘진상2호’

        The Jinsang 2 cultivar is a new late-maturing, low amylose rice variety with a medium to short culm created by the Seedpia Inc. breeding company in 2012. This variety was derived from a cross between Junam, a high-yield and lodging tolerant source, and DM3-8-72-9, which was derived from a cross between Donjinchal and Milkyqueen, with a low amylose content and superior agronomic traits. A general single seed descent method was applied to develop Jinsang 2 in the F3-F4 generations. During the F5-F8 generations, the breeding lines were selected through a pedigree breeding method. A promising line, JS6-B-52-8-3-5-1-B-B-B, was selected and named HJ-6 in 2011. This variety headed on the 25th of August, which was 14 days later than Hwayoung. The culm and panicle length of the variety was 71.6 cm and 19.9 cm, respectively. The milled rice from the variety had a dull and medium- to short-grain, and the 1,000-grain weight was around 20.0 g. It had a low amylose content of 12.2% and a low protein content of 6.8%. The yield performance of Jinsang 2 milled rice was about 5.36 MT/ha from the ordinary culture replicated yield trial test.

        고품질 중간찰 향미벼 ‘골든퀸 3호’

        ‘Golden queen 3’, an aromatic rice variety, was developed by Seedpia inc. (Suwon, Korea) breeding company in 2014. This cultivarwas derived from a cross between ‘HJ-11’, a high-yield, aromatic line, and ‘JCH33M-15-1-9-1’, which has a low amylose content. Duringthe F3-F8 generation, breeding lines were selected using a pedigree breeding method. The most promising line (JS9-3-73-15-5-1-5-3-1-1) wasadvanced and named ‘SP005’ in 2013. This variety was headed on August 13, two days later than ‘Hwayeong’ in the ‘in middle plane’is commonly used to distinguish rice cultivation areas in Korea, so I think it s no problem to the understanding for readers (including farmers). The culm and panicle lengths of this variety were 75.7 cm and 20.1 cm, respectively. It has approximately 14.7 tillers per hill and 104.8spikelets per panicle, and the 1000-grain weight was 21.8 g. The yield performance was 5.47 MT/ha in the local adaptability test, and theamylose content of this variety was 12.5%. 2-Acetyl-1-pyrroline was detected as the major volatile ingredient in aromatic rice, and a 1 bpinsertion on the 14th exon (badh2.7 allele) was identified in the Badh2 gene.         한국 재래종 벼 유묘기 뿌리 발달 전장유전체연관분석

        Root development at the seedling stage is an important trait for the early growth of rice. In this study, the root length and weightof 211 Korean landrace rice at the seedling stage were evaluated, and the average root length and weight were 6.94 cm and 48.18 mg, respectively. Principal component analysis (PCA) of 211 accessions based on 70,849 SNPs presented three subgroups. Two quantitative trait loci (QTLs)associated with root length were detected on chromosomes 3 and 6 using genome-wide association study (GWAS). Four haplotypes wereidentified using associated SNPs for the two root length QTLs. The average root length of Hap2 and Hap 3 was 7.32 cm and 6.56 cm,respectively, and were significantly different. Gene expression analysis in the candidate regions of QTLs for root length at the seedling stageshowed that several genes were expressed in the root. QTLs associated with root length at the seedling stage identified in this study maybe applied to improve the root length trait and detect candidate genes.

        We are making continuous efforts to improve the agreeableness of a car HVAC system. As interior noise level of the car is improved and the requirement of the customer is enlarged, the Sound Quality improvement has been desired more. In this research we understood the influence of the factors at the HAVC SYSTEM on the sound pressure level and the sound quality by the use of Taguchi Method. We could enhance the sound quality through improvement of the main factors.

      • 이너스타그램: 인물 사진과 컬러를 이용한 감정 저널링

        이너스타그램(Innerstagram)은 포토 부스를 통해 감정을 추적하는 접근 방법으로, 사용자의 감정상태에 따라 그날 하루를 대표할 수 있는 색상을 입힌 인물 사진을 제공한다. 우리는 2 명의 참가자와 함께 이틀에 걸쳐 실시된 파일럿 테스트를 통해 Innerstagram 의 장점과 개선점을 조사하였다. 파일럿 테스트에는 다이어리 스터디(Diary Study)와 향후 개선을 위한 레트로스펙티브 인터뷰(Retrospective Interview)가 포함되어 있다. Innerstagram 은 꾸준히 마음을 추적하고자 하는 이들을 위해, 매일 일기를 작성하는 대신 사용할 수 있도록 디자인되었다. 우리는 Innerstagram 의 결과물을 인화하여 물리적으로 만질 수 있도록 설계하여 사용자가 계속 자기 추적을 이어가도록 동기를 부여하였고, 제품을 직접 사용하는 경험을 주어 이를 매일 떠올릴 수 있도록 하였다. 직접 사용하는 경험에 대한 흥미가 떨어진다 하더라도 색상을 입힌 사진의 기억은 장기간 유지될 것이다. 이 연구는 감정 추적용 도구의 디자인 특징과 Innerstagram 이 사용자의 지속적인 감정 추적을 독려하는 데에 어떤 역할을 할 수 있는지 그 기회를 보여주며 결론을 내린다.
