        PROM2 promotes gemcitabine chemoresistance via activating the Akt signaling pathway in pancreatic cancer

        In recent years, the deoxycytidine analogue gemcitabine (2′,2′,-difluorodeoxycytidine) has become the first-line chemotherapeutic agent for patients with pancreatic cancer. However, due to the intrinsic resistance of pancreatic cancer cells, gemcitabine-based chemotherapy yields limited disease control, with >85% disease progression at 6 months from diagnosis. Therefore, elucidating the mechanisms of chemoresistance is a critical step in improving cancer therapy, especially for the treatment of pancreatic cancer. We show PROM2, a transmembrane glycoprotein, is ubiquitously upregulated in pancreatic cancer cell. We also found higher PROM2 expression is associated with shortened overall and disease-free survival times in patients diagnosed with pancreatic cancer. We provide evidence that PROM2 promotes chemoresistance to gemcitabine both in vivo and in vitro. Mechanistically, we demonstrate that PROM2 could directly interacted with Akt and activates the Akt signaling pathway, which thus inhibiting gemcitabineinduced apoptosis. As further evidence, we show PROM2 expression and Akt phosphorylation both promote gemcitabine chemoresistance, and cause poorer survival in clinical samples with pancreatic cancer. Combining gemcitabine with the Akt inhibitor MK-2206 facilitated significant tumor shrinkage and dramatically elevated the survival status in mice xenografted with pancreatic cancer cells. Our findings not only establish PROM2 as a novel positive regulator of the Akt signaling pathway and a candidate prognostic indicator of gemcitabine response, but also provide a neo-therapeutic approach for patients resistant to gemcitabine treatment.

      • Online Mean Kernel Learning for Object Tracking

        Features for representing the target are the fundamental ingredient when constructing the appearance model in the tracking problem. Only one type of features is utilized to represent the target in most current algorithms. However, the limited representation of a single feature might not resist all appearance changes of the target during the tracking process. To cope with this problem, we propose a novel tracking algorithm - Mean Kernel Tracker (MKT) - to robustly locate the object. The MKT combines three complementary features - Color, HOG (Histogram of Oriented Gradient) and LBP (Local Binary Pattern) - to represent the target. And Extensive experiments on public benchmark sequences show MKT performs favorably against several state-of-the-art algorithms.

        Upregulation of adiponectin by Ginsenoside Rb1 contributes to amelioration of hepatic steatosis induced by high fat diet

        Background: Ginsenoside Rb1 (GRb1) is capable of regulating lipid and glucose metabolism through itsaction on adipocytes. However, the beneficial role of GRb1-induced up-regulation of adiponectin in liversteatosis remains unelucidated. Thus, we tested whether GRb1 ameliorates liver steatosis and insulinresistance by promoting the expression of adiponectin. Methods: 3T3-L1 adipocytes and hepatocytes were used to investigate GRb1's action on adiponectinexpression and triglyceride (TG) accumulation. Wild type (WT) mice and adiponectin knockout (KO)mice fed high fat diet were treated with GRb1 for 2 weeks. Hepatic fat accumulation and function as wellas insulin sensitivity was measured. The activation of AMPK was also detected in the liver andhepatocytes. Results: GRb1 reversed the reduction of adiponectin secretion in adipocytes. The conditioned medium(CM) from adipocytes treated with GRb1 reduced TG accumulation in hepatocytes, which was partlyattenuated by the adiponectin antibody. In the KO mice, the GRb1-induced significant decrease of TGcontent, ALT and AST was blocked by the deletion of adiponectin. The elevations of GRb1-induced insulinsensitivity indicated by OGTT, ITT and HOMA-IR were also weakened in the KO mice. The CM treatmentsignificantly enhanced the phosphorylation of AMPK in hepatocytes, but not GRb1 treatment. Likewise,the phosphorylation of AMPK in liver of the WT mice was increased by GRb1, but not in the KO mice. Conclusions: The up-regulation of adiponectin by GRb1 contributes to the amelioration of liver steatosisand insulin resistance, which further elucidates a new mechanism underlying the beneficial effects ofGRb1 on obesity

        Resilient Filtering for Delayed Markov Jump Neural Networks via Event-triggered Strategy

        This paper deals with the event triggered filtering problem for a class of delayed discrete-time Markov jump neural networks, where a resilient filter with parameter uncertainties is adopted. The aim of this paper is to design a suitable filter which ensures that the filtering error system is stochastically stable and satisfies a prescribed mixed passivity and H∞ performance. Sufficient conditions for solvability of such a problem are developed. Basedon the obtained conditions, an explicit expression of the desired resilient filter is proposed. Finally, an example is presented to show the usefulness of the proposed scheme.

        Assessment of Myocardial Bridge by Cardiac CT: Intracoronary Transluminal Attenuation Gradient Derived from Diastolic Phase Predicts Systolic Compression

        Objective: To study the predictive value of transluminal attenuation gradient (TAG) derived from diastolic phase of coronary computed tomography angiography (CCTA) for identifying systolic compression of myocardial bridge (MB). Materials and Methods: Consecutive patients diagnosed with MB based on CCTA findings and without obstructive coronary artery disease were retrospectively enrolled. In total, 143 patients with 144 MBs were included in the study. Patients were classified into three groups: without systolic compression, with systolic compression < 50%, and with systolic compression ≥ 50%. TAG was defined as the linear regression coefficient between intraluminal attenuation in Hounsfield units (HU) and length from the vessel ostium. Other indices such as the length and depth of the MB were also recorded. Results: TAG was the lowest in MB patients with systolic compression ≥ 50% (-19.9 ± 8.7 HU/10 mm). Receiver operating characteristic curve analysis was performed to determine the optimal cutoff values for identifying systolic compression ≥ 50%. The result indicated an optimal cutoff value of TAG as -18.8 HU/10 mm (area under curve = 0.778, p < 0.001), which yielded higher sensitivity, specificity, positive predictive value, negative predictive value, and diagnostic accuracy (54.1, 80.5, 72.8, and 75.0%, respectively). In addition, the TAG of MB with diastolic compression was significantly lower than the TAG of MB without diastolic compression (-21.4 ± 4.8 HU/10 mm vs. -12.7 ± 8 HU/10 mm, p < 0.001). Conclusion: TAG was a better predictor of MB with systolic compression ≥ 50%, compared to the length or depth of the MB. The TAG of MB with persistent diastolic compression was significantly lower than the TAG without diastolic compression.

        A tubular linear motor structure suitable for large thrust

        In response to the problem that the motor shaft of conventional tubular linear motor is prone to bending, this paper proposes a new tubular linear motor structure for large thrust demand. Bending of the motor shaft can lead to collisions between structures and affect motor performance, and traditional air-gap structures cannot effectively handle this problem. The stator of the new motor is divided into an inner and outer cylinder structure. The armature windings are arranged between the inner and outer cylinders. The permanent magnets are arranged on the motor shaft. After analysis, the structural proposed in this paper can well stop the bending of the motor shaft. It also effectively improves the problem of large fluctuations in the thrust of linear motors. Finally, it is concluded that the peak motor thrust is maximized when the material with low permeability is chosen for the inner cylinder.

        Electromagnetic Counter-recoil Mechanism Based on Adaptive Sliding Mode Control

        Traditional counter-recoil machines always have problems of difficulty in adjusting the movement process flexibly, maintenance difficulties, and so on. To end these, this paper proposes a new electromagnetic counterrecoil scheme using cylindrical linear motors. Firstly, the mathematical expression of counter-recoil movement under the linear motor is formulated, and the ideal movement trajectory is designed using the piecewise polynomial of velocity based on acceleration. To obtain a better performance, of the motor controller, a composed approach of an adaptive sliding mode control based on barrier function (BFASMC) and the finite-time disturbance observer (FTDO) is introduced into the modeling and computation. The stability of the controller is proved by establishing the Lyapunov function. The new adaptive laws based on the barrier function effectively make up for the shortcomings of the exponential function and improves control chattering. At last, the new scheme is simulated. The results show that it has a strong robustness and anti-interference ability.

        On $\pi\mathfrak{F}$-embedded subgroups of finite groups

        A chief factor $H/K$ of $G$ is called $\mathfrak{F}$-central in $G$ provided $(H/K)\rtimes (G/C_{G}(H/K))\in\mathfrak{F}$. A normal subgroup $N$ of $G$ is said to be $\pi\mathfrak{F}$-hypercentral in $G$ if either $N=1$ or $N\neq1$ and every chief factor of $G$ below $N$ of order divisible by at least one prime in $\pi$ is $\mathfrak{F}$-central in $G$. The symbol $Z_{\pi\mathfrak{F}}(G)$ denotes the $\pi\mathfrak{F}$-hypercentre of $G$, that is, the product of all the normal $\pi\mathfrak{F}$-hypercentral subgroups of $G$. We say that a subgroup $H$ of $G$ is $\pi\mathfrak{F}$-embedded in $G$ if there exists a normal subgroup $T$ of $G$ such that $HT$ is $s$-quasinormal in $G$ and $(H\cap T)H_G/H_G\leq Z_{\pi\mathfrak{F}}(G/H_G)$, where $H_G$ is the maximal normal subgroup of $G$ contained in $H$. In this paper, we use the $\pi\mathfrak{F}$-embedded subgroups to determine the structures of finite groups. In particular, we give some new characterizations of $p$-nilpotency and supersolvability of a group.
