<P>Photochemistry in retinal proteins (RPs) is determined both by the properties of the retinal chromophore and by its interactions with the surrounding protein. The initial retinal configuration, and the isomerization coordinates active in any specific protein, must be important factors influencing the course of photochemistry. This is illustrated by the vast differences between the photoisomerization dynamics in visual pigments which start 11-<I>cis</I> and end all-<I>trans</I>, and those observed in microbial ion pumps and sensory rhodopsins which start all-<I>trans</I> and end in a 13-<I>cis</I> configuration. However, isolating these factors is difficult since most RPs accommodate only one active stable ground-state configuration. Anabaena sensory rhodopsin, allegedly functioning in cyanobacteria as a wavelength sensor, exists in two stable photoswitchable forms, containing all-<I>trans</I> and 13-<I>cis</I> retinal isomers, at a wavelength-dependent ratio. Using femtosecond spectroscopy, and aided by extraction of coherent vibrational signatures, we show that <I>cis</I>-to-<I>trans</I> photoisomerization, as in visual pigments, is ballistic and over in a fraction of a picosecond, while the reverse is nearly 10 times slower and kinetically reminiscent of other microbial rhodopsins. In this paper, we propose novel optics design for media inside near-field recording (NFR) using hemispherical solid immersion lens (HMS). To obtain strong advantage of data protection and high data capacity simultaneously, HMS based near field optics using aplanatic position of super hemisphere is designed. In this design, to improve small optical tolerance of this aplanatic position, additional aspheric lens surface is added on top of the HMS and it is combined with zoom optics which composed of two single lenses having low numerical aperture (NA). Also, to compensate chromatic aberration which happens seriously in optics using blue laser diode, diffractive optical element is used. Using zoom optics, additional aspheric lens surface, and diffractive optical element together, wavefront aberration and chromatic aberration are effectively reduced in broad range of cover layer thickness and wavelength variation. In addition, in this paper, effect of gap induced aberration is investigated by analyzing different behavior of each TM and TE wave for designed media inside dual-layer NFR optics.

Expectation propagation is a general approach to fast approximate inference for graphical models. The existing literature treats models separately when it comes to deriving and coding expectation propagation inference algorithms. This comes at the cost of similar, long-winded algebraic steps being repeated and slowing down algorithmic development. We demonstrate how factor graph fragmentization can overcome this impediment. This involves adoption of the message passing on a factor graph approach to expectation propagation and identification of factor graph sub-graphs,which we call fragments, that are common to wide classes of models. Key fragments and their corresponding messages are catalogued which means that their algebra does not need to be repeated. This allows compartmentalization of coding and efficient software development.

The identification of Plasmodium falciparum enoyl acyl-carrier protein reductase (pfENR) is considered as a potential biological target against malaria. Trema orientalis is considered a rich source of phytochemicals useful in malaria treatment. This study evaluated the in-vitro inhibitory activity of the extract and isolated compounds of T. orientalis leaf; the isolated compounds and the analogues of the most active compound were subjected to in-silico molecular docking studies on pfENR. The methanolic extract of T. orientalis was subjected to repeated chromatographic separation which led to the isolation of some compounds. The isolated compounds from the plant were examined for their antimalarial activity using β-hematin inhibition assay. Virtual screening via molecular docking and ADMET studies were conducted to gain insight into the mechanism of binding of ligand and to identify effective pfENR inhibitors. The isolated compounds and the analogues of the most active isolates were gotten from PubChem library for use in docking study. Hexacosanol and β-sitosterol showed inhibition of the β-hematin formation. The docking results showed that hexacosanol, β-sitosterol and the analogues of β-sitosterol displayed binding energy ranging between -6.1 kcal/mol and -11.6 kcal/mol. Sitosterol glucoside has the highest docking score. Some of the ligands showed more binding affinity than known bioactive compounds used as reference. Analogues of β-sitosterol has been shown to be potential inhibitors of pfENR, therefore, the findings from this study suggest that sitosterol glucoside and ergosterol peroxide could act as antimalarial agents after further lead optimisation investigations.
