        미국의 식물유래약용물질(Plant-Made Pharmaceuticals, PMPs)의 안전성 규제

        1. 식물유래 약용물질 (PMP)은 살아있는 식물에서 생산된 치료용 단백질로, 발효조에서 세포계통을 이용하는 것보다 적은 비용으로 높은 순도의 단백질을 생산할 수 있어 이에 대한 연구개발이 확대될 전망이다. 2. 식물유래 약용물질의 재배시험을 감독하는 주관부서는 미농무부(USDA)의 동식물 검역청(APHIS)이다. 3. PMP의 생산에는 알팔파. 옥수수, 벼 잇꽃과 담배 등이 이용되고 있다. 4. 현재 상업화된 PMP는 없으나 ProdiGene, Inc사 등 몇몇 생명공학 회사에서 이들의 연구개발이 진행되고 있다. 5. 2002년에 PMP 재배시험은 130에이커 규모로 34개 시험장에서 20건이 허가 되었으며 주별로는 애리조나, 네브라스카 등 14주에서 시험이 이루어 졌다. 6. 미국 USDA의 PMP 재배 지침에 의하면 PMP의 재배생산에는 안전성확보를 위한 별도의 전용장비를 필요로 하며 파종에서 생산까지 엄격하게 관리된다. 7. 미국 USDA의 APHIS는 2002년 PMP 옥수수를 생산하는 ProdiGene, Inc사에 대하여 2건의 포장시험허가 위반사항을 적발하여 벌금 조치를 한바있다. Plant-made pharmaceuticals(PMPs) are therapeutic agents(pharmaceutical proteins) produced in genetically engineered plants. To yield these valuable products plants offer several advantages that include large-scale production capacity at a reduced cost, equivalent purity/activity to other manufacturing systems, and freedom from contamination with animal pathogens, prions, or disease-causing germs. But there are risks of contamination of the food supply and unintended effects on ecosystems. In the U.S. PMPs are regulated under stringent requirement of the U.S. Department of Agriculture(USDA) and the Food and Drug Administration(FDA). In this article, contamination incident with PMP corn engineered by ProdiGene, Inc will be briefly introduced and regulations of PMPs will be discussed.

        Distribution and current vegetation of Cakile edentula, an invasive alien species in Korea

        Background: Cakile edentula (Bigelow) Hook. is a successful invader that has been propagating globally. In Korea, Cakile edentula was found in 2008 for the first time, in the east coast of Korean peninsula. Based on site surveys conducted thereafter, Cakile edentula has propagated in the east coast from Goseong to Jeongdongjin, Gangneung and in the west coast of city of Hwaseong of Gyeonggi-do Province, and Taeangun County of Chungcheongnam-do Province. Results: Cakile edentula has infiltrated into four different vegetation communities (syntaxa) (Calystegio soldanellaeSalsoletum komarovii Ohba, Miyawakiet Tx.1967; Elymo-caricetum Kobomugi Miyawaki 1967; Carex pumila; and Calystegia soldanella) belonging to phytosociological classes of saltwort (barilla) and coastal glehnia. Cakile edentula competes with dominant species such as the saltwort (barilla) and beach morning glory, causing qualitative changes in species composition. It also affects local ecosystems through its competition with indigenous species, causing destruction of the traditional landscape. Conclusions: However, competition of Cakile edentula with indigenous species and the resulting replacement of such species are limitedly reported. Potential competitors with Cakile edentula have not been found yet. In Northeast Asia, qualitative changes realized in local ecosystems due to invasion of Cakile edentula are significantly noticeable. These necessitate proper controls for this invasive alien species to protect and preserve coastal dune areas.

        Distribution characteristic of invasive alien plants in Jeju Island

        Background: This study was undertaken to analyze the distribution and ecological characteristics of invasive alien plant species on Jeju Island, and to provide basic data for their management and control. Results: A field research was conducted at 436 locations on Jeju Island. The field research identified nine species of invasive alien species growing on Jeju Island. Based on the distribution pattern, Hypochaeris radicata L., Rumex acetosella, and Ambrosia artemisiifolia L. were found to be distributed horizontally throughout Jeju Island, with vertical growth in two or more vegetation zones, from warm temperate to the subalpine zone. Widely distributed species penetrate various habitats, such as grasslands, ranches, roadsides, farmlands, and empty lots, and have an immensely negative impact on the ecosystem, including declining biodiversity on Jeju Island. Paspalum distichum var. indutum Shinners, Paspalum distichum L., Solanum carolinense L., and Aster pilosus Willd. were distributed in some areas as a biased distribution species, whereas Lactuca scariola L. and Solidago altissima L. were found only in certain areas as centralized distribution species. Conclusions: The centralized distribution species and biased distribution species of the invasive plants in the ecosystem of Jeju Island should be physically eliminated, keeping in mind the short- and mid-term perspectives and monitoring, and by considering expansion of additional distribution areas. Due to limitations of physical/chemical elimination, time, and cost, widely distributed species require to be eliminated and managed, mainly to restore the integrity of the ecosystem, by planting native species to reestablish the habitat.

        생태계교란식물의 부산광역시 분포 실태

        부산광역시에 분포하는 생태계교란식물에 대해 종 다양성과 분포현황을 규명하고자 연구를 수행하였다. 연구결과 부산광역시에는 10종의 생태계교란식물이 분포하고 있는 것으로 확인되었으며, 국내에서 높은 종 다양성을 보이는 지역으로 확인되었다. 집괴분석을 통해 미국쑥부쟁이 우점군락, 가시상추 우점군락, 돼지풀 우점군락, 애기수영 우점군락, 도깨비가지 우점군락 등이 높은 유사성을 나타내는 것으로 확인되었다. 생태계 교란식물은 우점군락 내에서 다른 생태계 교란식물이 함께 혼생하는 경향을 보여, 유사한 서식처를 공유하고 있는 것올 밝혀졌다. 부산시에서 가장 광범위한 분포경향을 보이는 종은 가시상추(16개 구)이며, 돼지풀(11개 구), 미국쑥부쟁이(11개 구), 애기수영(10개 구) 등이 넓은 분포를 보였다. 행정구역 중 가장 다양한 생태계교란식물이 출현한 지역은 강서구(8종)와 북구(8종)이며, 인위적 간섭이 높으면서 다양한 서식처가 존재하는 지역들로 확인되었다. 부산광역시의 종별 서식처 환경정보가 수집되었다. 본 연구를 통한 결과는 부산광역시 내 생태계교란식물의 현황파악과 더불어 효율적 관리와 확산 방지를 위한 기초자료로 활용될 수 있을 것으로 기대된다. This research aimed to identify the diversity and the distribution of invasive alien plant species in the metropolitan city of Busan, South Korea. According to our results, we discovered 10 species of invasive alien plants distributed in Busan, demonstrating that this urban area has a high domestic plant diversity. A cluster analysis identified that the dominant communities of Aster pilosus, Lactuca serriola, Ambrosia artemisiifolia, Rumex acetosella and Solanum carolinense were highly similar in species composition. Different species of invasive alien plants tended to occur together in dominant communities, indicating their preference for shared habitats. The most extensively distributed species in Busan were Lactuca serriola (16 districts), followed by Ambrosia artemisiifolia (11 districts), Aster pilosus (11 districts) and Rumex acetosella (10 districts). The administrative districts with the most diverse invasive alien plants were Gangseo-gu (8 species) and Buk-gu (8 species), which are both areas with high human interference and diverse habitats. Additional environmental information was collected for these species’ habitats in Busan. The results of this research can be used to assess the current status of invasive alien plants in Busan and can provide basic data useful for effectively controlling and preventing the spread of invasive plants.

        Capillary Characteristics of Water and Cations in Multi-layered Reclaimed Soil with Macroporous Subsurface Layer Utilizing Coal Bottom Ash

        Serious problems in reclaimed land agriculture are high soil salinity and poor vertical drainage, so desalinization in these soils is very difficult. Also, although desalinization is accomplished in reclaimed top soils, before long, soils are resalinized according to capillary rise of salts from the subsurface soils. To resolve these problems, multi-layered soil columns with subsurface layer of macroporous medium utilizing coal bottom ash (CBA) were constructed and the effects of blocked resalinization of these soils were investigated. In this experiment soil samples were collected from Munpo series (coarse-loamy, nonacid, mixed, mesic, typic Fluvaquents). The soil texture was silt loam and the EC was $33.9dS\;m^{-1}$. As for groundwater seawater was used and groundwater level of 1 cm from the bottom was maintained. The overall rate of capillary rise was $2.38cm\;hr^{-1}$ in soil 60 cm column, $0.25cm\;hr^{-1}$ in topsoil (30 cm) + CBA (5 cm) + subsurface soil (10 cm) column and $0.08cm\;hr^{-1}$ in topsoil (30 cm) + CBA (10 cm) + subsurface soil (10 cm) column. In multi-layered soil columns with CBA 20, 30 cm layer, wetting front due to capillary rise could not be seen in top soil layer. After 70 days capillary rise experiment water soluble Na+ accumulated in top soil of soil columns with CBA 20, 30 cm was diminished by 92.8, 96.5% respectively in comparison with Na+ accumulated in top soil of soil 60 cm column because CBA layer cut off capillary rise of salts from the subsurface soil. From these results we could conclude that the macroporous layer utilizing CBA placed at subsurface layer cut off capillary rise of solutes from subsurface soil, resulting in lowered level of salinity in top soil and this method can be more effective in newly reclaimed saline soil.

        Identification of a novel splicing mutation within <i>SLC17A8</i> in a Korean family with hearing loss by whole-exome sequencing

        <P><B>Abstract</B></P> <P>Hereditary hearing loss (HHL) is a common genetically heterogeneous disorder, which follows Mendelian inheritance in humans. Because of this heterogeneity, the identification of the causative gene of HHL by linkage analysis or Sanger sequencing have shown economic and temporal limitations. With recent advances in next-generation sequencing (NGS) techniques, rapid identification of a causative gene <I>via</I> massively parallel sequencing is now possible. We recruited a Korean family with three generations exhibiting autosomal dominant inheritance of hearing loss (HL), and the clinical information about this family revealed that there are no other symptoms accompanied with HL. To identify a causative mutation of HL in this family, we performed whole-exome sequencing of 4 family members, 3 affected and an unaffected. As the result, A novel splicing mutation, c.763+1G>T, in the solute carrier family 17, member 8 (<I>SLC17A8</I>) gene was identified in the patients, and the genotypes of the mutation were co-segregated with the phenotype of HL. Additionally, this mutation was not detected in 100 Koreans with normal hearing. <I>Via</I> NGS, we detected a novel splicing mutation that might influence the hearing ability within the patients with autosomal dominant non-syndromic HL. Our data suggests that this technique is a powerful tool to discover causative genetic factors of HL and facilitate diagnoses of the primary cause of HHL.</P> <P><B>Highlights</B></P> <P> <UL> <LI> NGS is a powerful method to facilitate discovery and diagnosis of causative factors in genetic hearing loss. </LI> <LI> We performed whole exome sequencing to screen the genetic cause of autosomal dominant non-syndromic hearing loss. </LI> <LI> A probably pathogenic, novel splicing mutation was found in the <I>SLC17A8</I> gene. </LI> <LI> Our results provide essential information to understand hearing loss caused by a splicing mutation in the <I>SLC17A8</I> gene. </LI> </UL> </P>

      • Nucleation and growth of a helical nanofilament (B4) liquid-crystal phase confined in nanobowls

        <P>The B4 helical nanofilament (HNF) liquid crystal (LC) phase is a three-dimensional (3D) helical structure composed of 2D smectic layers. Because of the complex shape of the HNF phase, it is difficult to understand the generation mechanism of HNFs in the bulk as well as in the thin-film condition. Here, we directly investigated the nucleation and growth of HNFs in nanobowls. A combination of electron microscopy and X-ray diffraction was used to reveal the transitional surface structures, in which barrel-like structures as well as short HNFs with random handedness were observed, depending on the LC film thickness.       • Genetic Variants of IL-13 and IL-4 in the Korean Population: Polymorphisms, Haplotypes and Linkage Disequilibrium

        Asthma is an inflammatory airways disease characterized by bronchial hyperresponsiveness and airways obstruction, which results from a complex interaction of genetic and environmental factors. Interleukin (IL)-13 and IL-4 are important in IgE synthesis and allergic inflammation, therefore genes encoding IL-13 and IL-4 are candidates for predisposition to asthma. In the present study, we screened single-nucleotide polymorphisms (SNPs) in IL-13 and IL-4 and examined whether they are risk factors for asthma. We resequenced all exons and the promoter region in 12 asthma patients and 12 normal controls, and identified 18 SNPs including 2 novel SNPs. The linkage disequilibrium(LD) pattern was evaluated with 16 common SNPs, and haplotypes were also estimated within the block. Although IL-13 and IL-4 are localized within 27 kb on chromosome 5q31 and share many biological profiles, this region was partitioned into 2 blocks. One SNP and three SNPs were determined as haplotype-taggingSNPs (htSNPs) within IL-13 and IL-4 haplotype-block, respectively. No significant associations were observed between any of the SNPs or haplotypes and development of asthma in small number of Korean subjects. However, the genetic variants of IL-13 and IL-4 would provide valuable strategies for the genotyping studies in large population.

        Gene-Based Single Nucleotide Polymorphisms and Linkage Disequilibrium Patterns of 29 Asthma Candidate Genes in the Chromosome 5q31-33 Region in Koreans

        <P><I>Background and Methods:</I> Numerous genetic studies have mapped asthma susceptibility genes to a region on chromosome 5q31-33 in several populations. This region contains a cluster of cytokines and other immune-related genes important in immune response. In the present study, to determine the genetic variations and patterns of linkage disequilibrium (LD), we resequenced all the exons and promoter regions of the 29 asthma candidate genes in the chromosome 5q31-33 region. <I>Results:</I> We identified a total of 314 genetic variants, including 289 single nucleotide polymorphisms (SNPs), 22 insertion/deletion polymorphisms and 3 microsatellites. Standardized variance data for allele frequency revealed substantial differences in SNP allele frequencies among different ethnic groups. Interestingly, significant ethnic differences were observed mainly in intron SNPs. LD block analysis using 174 common SNPs with a frequency of >10% disclosed strong LD within most candidate genes. No significant LD was observed across genes, except for one LD block (CD14-IK block). Gene-based haplotype analyses showed that 1-5 haplotype-tagging SNPs may be used to define the six or fewer common haplotypes with a frequency of >5%, regardless of the number of SNPs. <I>Conclusion:</I> Overall, our results provide useful information for the identification of immune-mediated disease genes in the chromosome 5q31-33 region, as well as valuable evidence for gene-based haplotype analysis in disease association studies.</P><P>Copyright © 2006 S.
