• Yeast SREBP Cleavage Activation Requires the Golgi Dsc E3 Ligase Complex

        Stewart, Emerson ,V.,Nwosu, Christine ,C.,Tong, Zongtian,Roguev, Assen,Cummins, Timothy ,D.,Kim, Dong-Uk,Hayles, Jacqueline,Park, Han-Oh,Hoe, Kwang-Lae,Powell, David ,W.,Krogan, Nevan& Elsevier 2011 Molecular cell Vol.42 No.2

Summary

Mammalian lipid homeostasis requires proteolytic activation of membrane-bound sterol regulatory element binding protein (SREBP) transcription factors through sequential action of the Golgi Site-1 and Site-2 proteases. Here we report that while SREBP function is conserved in fungi, fission yeast employs a different mechanism for SREBP cleavage. Using genetics and biochemistry, we identified four genes defective for SREBP cleavage, dsc1-4, encoding components of a transmembrane Golgi E3 ligase complex with structural homology to the Hrd1 E3 ligase complex involved in endoplasmic reticulum-associated degradation. The Dsc complex binds SREBP and cleavage requires components of the ubiquitin-proteasome pathway: the E2-conjugating enzyme Ubc4, the Dsc1 RING E3 ligase, and the proteasome. dsc mutants display conserved aggravating genetic interactions with components of the multivesicular body pathway in fission yeast and budding yeast, which lacks SREBP. Together, these data suggest that the Golgi Dsc E3 ligase complex functions in a post-ER pathway for protein degradation.

Graphical Abstract

<ce:figure id='dfig1'></ce:figure>

Highlights

► Yeast SREBP is proteolytically activated by a different mechanism than mammalian SREBP ► Deletion collection screen identified four dsc genes required for fission yeast SREBP cleavage ► Dsc proteins form a Golgi E3 ligase complex that resembles Hrd1 E3 ligase in ERAD ► Yeast SREBP cleavage requires activities of the ubiquitin-proteasome pathway

      • NEO-NARRATIVE OR GRAMMARLESS LANGUAGE

        Stewart Ruth G 관동대학교 1992 關大論文集 Vol.20 No.1

This paper introduces, explains and critiques Neo-Narrative(or Grammarless Language), an attempt in American literature to expand the frontiers of language usage in literary and imaginative literature. While many people feel television is the cause of a decrease in reading, others feel the cause is the language itself, whichhas not kept up with the vast changes in other areas of society. Language must remain contemporary or it dies. Other arts have ahanged radically but literature is still using 18th Century grammar. The question is : is 18th Century grammar adequate for the 20th Century in which we live? Writers such as Joyce, Woolf and Faulkner expanded literary language with free association and stream of consciousness writing. Neo-Narrative goes one step beyond these authors. The primary proponents of Neo-Narrative are Arlene Zekowski and Stanley Berne. They have attempted, in their imaginative literature, to change the structure of language by both shortening and lengthening the sentence while basically ignoring rules regarding subjects, verbs and predicates-which they feel place a barrier between the author's emotion and the reader's understanding. In Zekowski's writing language is primary. Periods are used instead of commas so that each word has a value all its own. This she says "leads our sensibilities into the multiple (not linear) consciousness-awareness levels of our time." Mr. Berne's "prose" extends the sentence using language, without the benefit of grammar, to express unfettered thought. Both of these authors strive for immediacy. One critique is that the authors writing is form over substance, yet the authors write of important things-the earth, history and manking. Another question is, is this writing onlyto illustrate a theory? The authors say the writing came first. If this new writing frees us to a deeper understanding of one another, however, I feel it is worthwhile. In summary, Neo-Narrative is writing which attempts to emphasize language itself and present the reader with immediate reality uncensored by grammar. Neo-Narrative goes boyond the development of free association and stream of consciousness. It is a post-modern grammarless "Open Structure" breakthrough into yet another realm of literature.

      • Tolerance of adzuki bean to preplant-incorporated, pre-emergence, and post-emergence herbicides in Ontario, Canada

Stewart, Christie L.,Nurse, Robert E.,Gillard, Chris,Sikkema, Peter H. The Korean Society of Weed Science and The Turfgra 2010 Weed Biology and Management Vol.10 No.1

Weed management options for adzuki-bean growers in Ontario, Canada are limited due to few herbicide registrations. Four field trials were conducted at three locations in south-western Ontario in 2007 and 2008 to determine the tolerance of adzuki bean to several preplant-incorporated (PPI), pre-emergence (PRE), and post-emergence (POST) herbicides. All the herbicides were applied at the doses registered for use in soybean. The application of pendimethalin, cloransulam-methyl, and halosulfuron-methyl (PPI), flumetsulam, cloransulam-methyl, and halosulfuron-methyl (PRE), and acifluorfen and fomesafen (POST)caused ${\leq}15%$ crop injury; however, the injury was transient and did not reduce the adzuki bean yield. The POST application of cloransulam-methyl and imazethapyr caused ${\leq}23%$ crop injury and reduced the biomass by ${\leq}50%$, but did not reduce the plant height or crop yield. Metribuzin, flumetsulam, atrazine, and pyroxasulfone (PPI), metribuzin, linuron, pyroxasulfone, and atrazine (PRE), and bentazon, imazethapyr plus bentazon, halosulfuron-methyl, and thifensulfuron-methyl (POST) caused ${\leq}61%$ crop injury. These treatments reduced the biomass, plant height, and crop yield. Based on these results, pendimethalin, cloransulam-methyl, and halosulfuron-methyl applied PPI, flumetsulam, cloransulam-methyl, and halosulfuron-methyl applied PRE, and acifluorfen and fomesafen applied POST might be potential weed management options for weed management in adzuki bean. Cloransulam-methyl and imazethapyr applied POST will need further evaluation due to phytotoxicity concerns. Metribuzin, flumetsulam, atrazine, and pyroxasulfone applied PPI, metribuzin, linuron, atrazine, and pyroxasulfone applied PRE, and bentazon, imazethapyr plus bentazon, halosulfuron-methyl, and thifensulfuron-methyl applied POST did not have an adequate margin of safety.

A Tobacco Cessation Intervention with Rural, Medically Underserved, Blue-collar Employees: A Quasiexperimental Study

Stewart, Telisa,Formica, Margaret K.,Adachi-Mejia, Anna M.,Wang, Dongliang,Gerrard, Meg Occupational Safety and Health Research Institute 2016 Safety and health at work Vol.7 No.4

Background: The aim of this study was to increase knowledge regarding the dangers associated with tobacco use, and decrease secondhand smoke exposure and tobacco use behaviors with an antitobacco messaging campaign among rural, medically underserved, blue-collar workers. Methods: A quasiexperimental study was conducted with employees at two worksites. One worksite received the intervention, which consisted of nine different antitobacco messages. Baseline and follow-up surveys were conducted at each worksite to assess change in knowledge and behavior; the data were compared across the two worksites. Results: Two hundred twenty-two and 243 participants completed baseline and follow-up surveys at the intervention and comparison sites, respectively. A statistically significant difference was seen over time between the worksites on knowledge of the dangers of tobacco (p < 0.0001); the mean knowledge score increased at the intervention site, but remained unchanged at the comparison site. In general, non-smokers at both worksites appeared to try to decrease exposure to secondhand smoke over the follow-up period. Repeated measures analysis indicated that there were no differences in motivation to quit (p = 0.81), interest in quitting (p = 0.40), thinking about quitting (p = 0.53), or several tobacco-use behaviors over time among smokers at the intervention and comparison worksites. There were slight increases over time in the proportion of smokers who do not allow smoking in their homes/vehicles at the intervention worksite, although not statistically significant. Conclusion: Participants at the intervention worksite increased their knowledge regarding the dangers of tobacco use and secondhand smoke exposure. Among current tobacco users, the intervention appeared to increase family rules regarding secondhand smoke exposure in their homes and vehicles.

      • The Case For The Inclusion of A Course on the Bible In The English Language And Literature Curriculum

        Stewart,Ruth G 관동대학교 1991 關大論文集 Vol.19 No.1

The case for the inclusion of a course in Bible in the English Language and Literature Department curriculum is based on the fact that while the Bible is a religious book it is also a book of fine literature. As a religious book it belong to Christians and Jews. As a book of great literature it belongs to everyone. The Bible contains examples of nearly every literary genre, many of which are outstanding. The moral and ethical values of the Bible often make these excellent literary examples great literature. The Bible is a primary source of western cultural values. Anyone studying English literature must have a good understanding of western culture if they are to understand and critique its literature. The references to the Bible, both direct and indirect, in English literature are so numerous it is obivous that many western authors assume the readers have and acquaintance with the Bible. Many poems and dramas are based on Biblical stories. The number of English words and sayings arising form the Bible compel the serious student of English language and literature to be, at least, aware of their origin thereby having a deeper understanding of their meaning. The beauty of the language of the KIng James Version of the psalms, poetry and prose sections of the Bible are thought to be some of the finest use of the English language in the world. Students deserve to study and appreciate the finest English language in its original setting. Most of all the combination of superb literature and superb language make it a book through which the English student may first hand learn how universal themes and good language together make great literature. While overall survey coures of western culture are good, in studying the Bible as a primary source, the students have the opportunity to make their own discoveries and connection, which is the best form of education. If our English language and literature students are to excel in their field, they must have a depth of understanding of western culture, language and literature which only a study of the Bible effords. The Bible has abundant material for a course on the Bible as and in literature. Texts in English are already available in Kore. Recognition of the importance and need for such a course in essential.
