        Stenophagy as a Pathway for the Occurrence of Unique Coral Fatty Acid Biomarkers in Higher Branches of the Marine Tree of Life: The Nudibranch Armina maculata Case

        Tetracosapolyenoic fatty acids (TPA), namely tetracosapentaenoic (24:5n-6) and tetracosahexaenoic (24:6n-3) acids, are chemotaxonomic biomarkers of octocorals (Cnidaria, Octocorallia) in the marine environment. The present study confirms the occurrence of TPA on a marine non-cnidarian taxon, the nudibranch mollusc Armina maculata. This discovery is explained by the specialized feeding regime (stenophagy) of this sea slug that preys upon the octocoral Veretillum cynomorium. The prevalence of 24:5n-6 and 24:6n-3 in the body of A. maculata was demonstrated through the analysis of specimens starved for 30 days in captivity. Since the time frame is superior to the residence time of ingested octocorals, the present findings provide empirical evidence that the reported TPA are allocated from nudibranch tissues. Here we found support for previous claims that the presence of TPA in the marine tree of life is not restricted to its lower branches, as stenophagous trophic interactions may allow its transfer to or posterior biosynthesis in more evolved taxa.

        Levosimendan is a calcium sensitiser, acting by increasing the sensitivity of the heart cells to calcium, thus increasing cardiac contractility without a rise in intracellular calcium. Levosimendan exerts its positive inotropic effect by binding to cardiac troponin C in a calcium-dependent manner. It also has a vasodilatory effect, by opening adenosine triphosphate (ATP)-sensitive potassium channels in vascular smooth muscle to cause smooth muscle relaxation. The combined inotropic and vasodilatory actions result in an increased force of contraction, decreased preload and decreased afterload. Moreover, by opening also the mitochondrial (ATP)-sensitive potassium channels in cardiomyocytes, the drug exerts a cardioprotective effect. Levosimendan is indicated for inotropic support in acutely-decompensated severe congestive heart failure. Having in mind the crescent use of this drug in patients with severe heart failure admitted to the ICU, it is important to assess the security and the different outcomes and cross-reference this information with the various comorbidities found in each patient. Therefore, it is possible to identify in which patients the use of levosimendam is preferred over the other various positive inotropic drugs available in the ICU, in order to establish a valuable treatment guideline.

        Fatal cases of Theileria annulata infection in calves in Portugal associated with neoplastic-like lymphoid cell proliferation

        This study was carried out to investigate fifteen cases of acute lethal infection of calves (≤ 4 months of age) by the protozoan parasite Theileria (T.) annulata in the south of Portugal. Calves developed multifocal to coalescent nodular skin lesions, similar to multicentric malignant lymphoma. Infestation with ticks (genus Hyalomma) was intense. Theileria was seen in blood and lymph node smears, and T. annulata infection was confirmed by isolation of schizont-transformed cells and sequencing of hypervariable region 4 of the 18S rRNA gene. At necropsy, hemorrhagic nodules or nodules with a hemorrhagic halo were seen, particularly in the skin, subcutaneous tissue, skeletal and cardiac muscles, pharynx, trachea and intestinal serosa. Histologically, nodules were formed by large, round, lymphoblastoid neoplastic-like cells. Immunohistochemistry (IHC) identified these cells as mostly CD3 positive T lymphocytes and MAC387 positive macrophages. A marker for B lymphocytes (CD79αcy) labeled very few cells. T. annulata infected cells in these nodules were also identified by IHC through the use of two monoclonal antibodies (1C7 and 1C12) which are diagnostic for the parasite. It was concluded that the pathological changes observed in the different organs and tissues were caused by proliferation of schizont-infected macrophages, which subsequently stimulate a severe uncontrolled proliferation of uninfected T lymphocytes.
