      • <i>Citrus bergamia</i> Risso Elevates Intracellular Ca <b>2+</b> in Human Vascular Endothelial Cells due to Release of Ca <b>2+</b> from Primary Intracellular Stores

Kang, Purum,Han, Seung Ho,Moon, Hea Kyung,Lee, Jeong-Min,Kim, Hyo-Keun,Min, Sun Seek,Seol, Geun Hee Hindawi Publishing Corporation 2013 Evidence-based Complementary and Alternative Medic Vol.2013 No.-

<P>The purpose of the present study is to examine the effects of essential oil of <I>Citrus bergamia</I> Risso (bergamot, BEO) on intracellular Ca2+ in human umbilical vein endothelial cells. Fura-2 fluorescence was used to examine changes in intracellular Ca2+ concentration [Ca2+]i . In the presence of extracellular Ca2+, BEO increased [Ca2+]i , which was partially inhibited by a nonselective Ca2+ channel blocker La3+. In Ca2+-free extracellular solutions, BEO increased [Ca2+]i in a concentration-dependent manner, suggesting that BEO mobilizes intracellular Ca2+. BEO-induced [Ca2+]i increase was partially inhibited by a Ca2+-induced Ca2+ release inhibitor dantrolene, a phospholipase C inhibitor U73122, and an inositol 1,4,5-triphosphate (IP3)-gated Ca2+ channel blocker, 2-aminoethoxydiphenyl borane (2-APB). BEO also increased [Ca2+]i in the presence of carbonyl cyanide m-chlorophenylhydrazone, an inhibitor of mitochondrial Ca2+ uptake. In addition, store-operated Ca2+ entry (SOC) was potentiated by BEO. These results suggest that BEO mobilizes Ca2+ from primary intracellular stores via Ca2+-induced and IP3-mediated Ca2+ release and affect promotion of Ca2+ influx, likely via an SOC mechanism.</P>

Linalool elicits vasorelaxation of mouse aortae through activation of guanylyl cyclase and K(+) channels.

Kang, Purum,Seol, Geun Hee Pharmaceutical Society of Great Britain 2015 Journal of pharmacy and pharmacology Vol.67 No.5

<P>ObjectivesThe aim of this study was to investigate the cardiovascular relaxing properties of monoterpene alcohol (-)-linalool (LIN), a principal component of several aromatic plants. MethodsWe assessed the effects of LIN on vascular contractility in mouse aortae and evaluated its underlying mechanisms of action. Key findingsWe found that LIN dose-dependently relaxed the vascular tonus of mouse thoracic aortae induced by prostaglandin F-2 alpha (PGF(2), 3m). This effect, however, was reduced by pretreatment with the nitric oxide synthase inhibitor L-NAME (30m). Treatment with the inhibitor of soluble guanylyl cyclase ODQ (2m) or the K+ channel blocker TEA (10mM) partially blocked LIN-induced vasorelaxation. Moreover, addition of TEA after incubation of the rings with L-NAME and ODQ partially blocked LIN-induced vasorelaxation. Furthermore, LIN was able to partially antagonize CaCl2-induced contractions in high potassium (80mM) Krebs' solution, whereas LIN did not affect Ca2+ release from endoplasmic reticulum Ca2+ stores. ConclusionOur findings indicate that LIN may induce endothelium-dependent vasorelaxation in mouse thoracic aortae by activating soluble guanylyl cyclase and K+ channels.</P>

        강푸름(Purum Kang),정금미(Kum Mi Jung),민선식(Sun Seek Min),설근희(Geun Hee Seol) 대한스트레스학회 2012 스트레스硏究 Vol.20 No.3

Ocimum basilicum L. (basil essential oil, BEO) is a plant used in traditional folk medicine for the treatment of pain, inflammation and hypertension. BEO-inhalation was used to examine the effects of the odor of BEO and BEO concentration dependency on pain and anxiety before spine surgery in patients with chronic low back pain. A total of sixty patients were randomly assigned to one of the four study groups (control, 0.1%, 1%, 5% BEO-inhalation group). Pain-visual analog scale (P-VAS), anxiety-visual analog scale (A-VAS), profile of mood states (POMS) test, blood pressure, heart rate, and respiratory rate were measured before and after inhalation of the odors for five minutes. Inhalation of BEO at 0.1%, 1% concentration caused significant decreases in P-VAS compared with control (p=.022, p=.022, respectively), meanwhile 5% BEO-inhalation significantly reduced in diastolic blood pressure compared with control (p=.023). However, there was no significant difference among the four groups in systolic blood pressure, heart rate, respiratory rate, A-VAS, POMS test. In patients with chronic low back pain before spine surgery, 0.1% and 1% BEO-inhalation is effective in alleviating pain and 5% BEO-inhalation reduce diastolic blood pressure. In conclusion, BEO-inhalation at specific concentrations can be useful method for the relief of pain and blood pressure control for patients with chronic low back pain before spine surgery. <i>Trans</i>-anethole prevents hypertension induced by chronic exposure to both restraint stress and nicotine in rats

Seo, Eunhye,Kang, Purum,Seol, Geun Hee Elsevier 2018 BIOMEDICINE AND PHARMACOTHERAPY Vol.102 No.-

<P><B>Abstract</B></P> <P>Chronic stress and smoking are major risk factors for hypertension, with stress also being a factor predisposing to smoking. Methods are needed to prevent and/or reduce hypertension induced by chronic exposure to both stress and nicotine. This study investigated whether <I>trans</I>-anethole would prevent hypertension induced by chronic exposure to both restraint stress and nicotine in rats. Rats received nicotine intraperitoneally for 21 days following restraint stress (2 h/day) and <I>trans</I>-anethole (62, 125, and 250 mg/kg) on days 4, 8, 12, 16 and 20. To confirm the preventive effects of <I>trans</I>-anethole, blood pressure and vascular tone were measured on the last day of the experiment, and compared with the results of nifedipine and aerobic exercise. The ability of <I>trans</I>-anethole, at doses of 125 mg/kg and 250 mg/kg, to prevent hypertension was comparable to that of aerobic exercise and nifedipine. Furthermore, nifedipine combined with aerobic exercise and <I>trans</I>-anethole reduced both blood pressure and vascular tone. These findings are the first to show that <I>trans</I>-anethole can prevent hypertension, suggesting that <I>trans</I>-anethole may be useful as a prophylactic antihypertensive agent.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

<i>Foeniculum vulgare</i> Mill. Protects against Lipopolysaccharide-induced Acute Lung Injury in Mice through ERK-dependent NF-kB Activation

        Hui Su Lee,Purum Kang,Ka Young Kim,Geun Hee Seol 대한생리학회-대한약리학회 2015 The Korean Journal of Physiology & Pharmacology Vol.19 No.2

<i>Foeniculum vulgare</i> Mill. (fennel) is used to flavor food, in cosmetics, as an antioxidant, and to treat microbial, diabetic and common inflammation. No study to date, however, has assessed the anti-inflam-matory effects of fennel in experimental models of inflammation. The aims of this study were to investigate the anti-inflammatory effects of fennel in model of lipopolysaccharide (LPS)-induced acute lung injury. Mice were randomly assigned to seven groups (n=7∼10). In five groups, the mice were intraperitoneally injected with 1% Tween 80-saline (vehicle), fennel (125, 250, 500<i>μ</i>l/kg), or dexame-thasone (1 mg/kg), followed 1 h later by intratracheal instillation of LPS (1.5 mg/kg). In two groups, the mice were intraperitoneally injected with vehicle or fennel (250<i>μ</i>l/kg), followed 1 h later by intra-tracheal instillation of sterile saline. Mice were sacrificed 4 h later, and bronchoalveolar lavage fluid (BALF) and lung tissues were obtained. Fennel significantly and dose-dependently reduced LDH activity and immune cell numbers in LPS treated mice. In addition fennel effectively suppressed the LPS-induced increases in the production of the inflammatory cytokines interleukin-6 and tumor necrosis factor-alpha, with 500<i>μ</i>l/kg fennel showing maximal reduction. Fennel also significantly and dose-dependently reduced the activity of the proinflammatory mediator matrix metalloproteinase 9 and the immune modulator nitric oxide (NO). Assessments of the involvement of the MAPK signaling pathway showed that fennel significantly decreased the LPS-induced phosphorylation of ERK. Fennel effectively blocked the inflammatory processes induced by LPS, by regulating pro-inflammatory cytokine production, transcription factors, and NO.
