We developed the educational purpose mobile application, named "Wild Flowers of Bukhansan National Park (version 1.0)", aiming for easy identification of wild flowers for students and visitors in the park. When visitors find a flower or part of plant in the park, visitors can search for its name utilizing the pictures and characters provided in their own smartphone mobile devices or tablet PCs. The application provides pictures of wild flowers in the park and character-based searching system based on 12 diagnostic features (e.g., growth form, leaf arrangement, flower symmetry, petal color, petal number, sepal number, etc). We adopted the complete floristic survey of Chung and Lee (1962) and added species that we confirmed their distribution in the park during the development of this application. In summary, number of vascular plants in this park was estimated to be 428 taxa including 100 families, 280 genera, 327 species, 1 subspecies, 50 varieties, and 5 formas. We provided a total of 588 pictures representing 358 taxa and each taxon includes multiple pictures in many cases. Included identification quizzes can be an efficient educational tool as well as fun activity for students and visitors who are learning plant species in Korea. Our next step will include GPS function in the application for indicating visitor's location and for providing previously reported sites of the species that we interested in the map of the park. The future application which includes GPS function will be a valuable tool for the monitoring of rare plants, plant researches related to the climate changes, etc. We currently provide Korean iPhone version only, and English version and both of android versions will be serviced soon.

In aerodynamic design of centrifugal compressors, impellers are designed through preliminary design and blade profile generation. In order to find out faults of the initially designed impellers, the detailed informations about internal flow phenomena such as pressure distribution, flow separation, blade loading, etc are essential. These informations can be acquired with flow measurements or computational flow analyses. In this study, we calculated 3-D viscous flow in 4 back-swept impellers which were designed in our laboratory, and analyzed the flow characteristics which influence the performance of impellers.

In present study, the unbalance in axial thrust and the change of volumetric efficiency when trimming the impeller diameter have been investigated. Present study is for the low specific speed multistage centrifugal pumps with balance piston as balancing mechanism. The effect of impeller trimming on the axial thrust balance has been compared in multistage pumps with horizontal axis and vertical axis. As a result of impeller trimmings, an additional axial thrust acting to pump inlet direction. Axial thrust unbalance by impeller trim has presented severely in vertical axis pumps than horizontal axis pump. The balance piston's diameter is required to be increasing proportion to impeller trim rate for maintaining axial thurst balance. And increase in piston length in parallel with increase in piston diameter was more effective to reduce axial thrust unbalance and volumetric efficiency drop as well.

Present paper describes on/off design performance of a 50KW turbogenerator gas turbine engine for hybrid vehicle application. For optimum design point selection, relevant parameter study is carried out. The turbogenerator gas turbine engine for a hybrid vehicle is expected to be designed for maximum fuel economy, ultra low emissions, and very low cost. Compressor, combustor, turbine, and permanent-magnet generator will be mounted on a single high speed (82,000 rpm) shaft that will be supported on air bearings. As the generator is built into the shaft, gearbox and other moving parts become unnecessary and thus will increase the system's reliability and reduce the manufacturing cost. The engine has a radial compressor and turbine with design point pressure ratio of 4.0. This pressure ratio was set based on calculation of specific fuel consumption and specific power variation with pressure ratio. For the given turbine inlet temperature, a rather conservative value of $1100^\circK$ was selected. Designed mass flow rate was 0.5 kg/sec. Parametric study of the cycle indicates that specific work and efficiency increase at a given pressure ratio and turbine inlet temperature. Off design analysis shows that the gas turbine system reaches self operating condition at N/$N_{DP}$ = 0.53. Bleeding air for turbine stator cooling is omitted considering low TIT and for a simple geometric structure. Various engine performance simulations including, ambient temperature influence, surging at part load condition. Transient analysis were performed to secure the optimum engine operating characteristics. Surge margin throughout the performance analysis were maintained to be over 80% approximately. Validation of present results are yet to be seen as the performance tests are scheduled by the end of 1998 for comparison.

Momentum exchange theory has been generally used for an analysis of the regenerative turbomachines due to Its direct description of the complicate circulatory flow. However, because Its application is limited only to linear region and Its model equations are Incomplete on three variables, it needs further refinements. In the present study it is unproved by introducing a central pivot of circulatory flow. Also, by assuming linear circulatory velocity distribution, mean radii of inlet and outlet flows through the Impeller are newly suggested. By applying control volume analysis to both linear region and the acceleration region. the governing equation on the circulatory velocity is derived As a result, systematic performance analysis on the entire region of the incompressible regenerative turbomachines can be earned out based on the proposed model equations.
Experimental study on the effect of blade angle on regenerative pump performance

<P>The regenerative pump is a kind of turbomachine that is capable of developing a high pressure rise at relatively low flow rates compared to the centrifugal and axial pumps. Although the efficiency of regenerative pumps is much lower than other turbomachines, they have still been widely used in many industrial applications for high heads at low flow rates. There are a few theoretical models to analyze the performance of regenerative pumps, though, the effect of the blade angle has not been included in any analysis model to date. In this study, the influence of the impeller blade angle and its shape on regenerative pump performance has been experimentally investigated. Straight blades with inclined blade angles of 0°, ± 15°, ± 30° and ± 45° were tested. In addition radial chevron impellers with chevron angles of 15°, 30° and 45° were also included in the present experiments. Hence a total of 10 blade configurations were examined. The pressure head, efficiency and the fluid temperature rise were measured at different flow rates and the results were expressed in appropriate non-dimensional coefficients. From the experimental results, it was found that the pressure head and the efficiency strongly depend on the blade angles as well as the blade geometry. Among all blade configurations tested in this study, the chevron blade exhibited the highest head with reasonably good efficiency. The comparative study shows that there is an optimum chevron angle of around 30° that yields the best performance. These experimental data may be used to include the effect of blade angles in the current performance analysis and design models to widen their applicability.</P>
접합전달을 이용한 Streptomyces natalensis ATCC27448의 형질전환 최적화 및 attB-site의 특성연구

상업적으로 중요한 macrolide계 항진균 학생물질인 natamycin을 생산하는 Streptomyces natalensis ATCC27448의 환자 유전학적인 연구를 위해 대장균으로부터 S. natalensis로 plasmid DNA를 직접 도입하는 형질전환법을 확립하였다. 이러한 S. natalensis의 형질전환은 oriT와 attP 단편을 가지고 있는, ${\Phi}C31$ 유래의 integration 벡터인 pSET152를 이용하여 Escherichia coli ET12567/pUZ28002을 DNA 공여체(donor)로 이용한 접합전달법(conjugal transfer)을 사용하여 확립하였다. 접합전달의 가장 높은 효율은 10 mM의 $MgCl_2$를 포함한 MS 배지에서, $6.25\times10^8$의 E. coli 공여체와 열처리를 하지 않은 S. natalensis의 포자를 사용하여 얻어졌다. 또 얻어진 접합전달체 (exconjugant)에 대하여 southern blot hybridization과 벡터가 삽입된 염색체부분의 염기서열분석을 통해 attB site와 pseudo-attB site를 확인하다. attB site의 경우에는 다른 방선균들처럼 S. natalensis 염색체의 pirin 상동체를 코드하는 ORF내에 존재하였으나 pseudo-attB site는 염색체내 다른 site (GenBank accession no. $YP\_117731$)에 존재하였고 그 염기서열은 attB 염기서열과 차이를 나타내었다. Streptomyces natalensis ATCC27448 produces natamycin, a commercially important macrolide antifungal antibiotic. For molecular genetic study of S. natalensis, we have developed a system for introducing DNA into S. natalensis via conjugal transfer from Escherichia coli. An effective transformation procedure for S. natalensis was established based on transconjugation from E, coli ET12567/pUZ8002 using a ${\Phi}C31$-derived integration vector, pSET152, containing oriT and attP fragments. The high frequency was obtained on MS medium containing 10 mM $MgCl_2$ using $6.25\times10^8$ of E.coli donor cells without heat treatment of spores. In addition, southern blot analysis of exconjugants and the sequence of plasmids containing DNA flanking the insertion sites from the chromosome revealed that S. natalensis contains a single ${\Phi}C31$ attB site and at least a secondary or pseudo attB site. Similar to the case of various Streptomyces species, a single ${\Phi}C31$ attB site of S. natalensis is present within an ORF encoding a pirin-homolog, but a pseudo-attB site is present within a distinct site (GenBank accession no. $YP\_117731$) and also its sequence deviates from the consensus sequences of attB sequence.
