      • 흰쥐 중추신경계내 난소로 투사하는 미주신경로에 관한 연구

        본 실험은 부교감신경의 하나인 미주신경이 난소를 지배하는 신경으로 관여하고 있는지를 pseudorabies 바이러스를 이용하여 난소신경로와 난소주사후 미주신경절단을 통하여 조사한 연구 보고이다. 이를 위하여 Sprague Dawley계 암흰쥐를 대상으로 pseudorabies 바이러스를 난소에 주사한 무리와 난소 주사후 미주신경을 절단한 무리에서 뇌를 적출하여 pseudorabies 바이러스에 대한 면역조직화학 염색을 시행하여 비교하였다. 본 실험결과 미주신경 중추신경로내의 상위신경핵들이 pseudorabied 바이러스에 대하여 양성반응이 줄어들었거나 관찰되지 않는 차이를 보였다. 즉 적색핵, 종말판혈관기관, 줄무늬체, 침상핵과 이마엽겉질은 부분적으로 난소의 미주신경로에 관여하고 있으며 미주신경등쪽핵, 고립로핵, 최후영역, 청색반점, 팔옆핵, 코리케퓨즈핵, 흑색질 및 시각교차위핵은 양성반응이 관찰되지 않아 미주신경으로 투사하는 부교감신경핵으로 조사되었다. The mammalian ovary has been known as receiving its innervation by sympathetic and sensory neurons of the peripheral nervous system from the brain. Recently, there were several functional reports that the vagus nerves were also regulating the ovarian function, but the vagus nerve had not been identified by clear morphological evidence. A viral transneuronal tracing technique has been used to demonstrate the morphological evidence for the central vagal involvement in ovarian innervation in brain areas. Bartha strain of pseudorabies virus injection was made into the ovary of Sprague Dawley rats. In experimental group, the vagus nerve of the same injection side was removed right after ovarian injection. At five days after initial injection, all the rats were sacrificed and brains were processed for immunohistochemistry. Several central nuclei including hypothalamic paraventricular nucleus showed strong bilateral positive labelings after unilateral injection in control rats, but the positive labelings were disappeared or decreased in several hypothalamic nuclei and nuclei of the vagus nerve. I n conclusion, these results provide the morphological evidence that vagus nerve has neural connection to ovary and by which the central nervous system may maintains the state of ovulation and reproduction as a possible parasympathetic routes in mammals.

        Antiadipogenic Activity of Solvent-partitioned Fractions from Limonium tetragonum in 3T3-L1 Preadipocytes

        갯질경이(Limonium tetragonum)는 질경이과에 속하는 여러해살이 풀로 습지에 자생하는 염생식물의 일종이며, 항산화, 항종양 및 간보호 효능이 있는 것으로 알려져 있다. 본 연구에서는 갯질경이 추출물로부터 용매 극성에 따라 분획한 분획물(H₂O, n-BuOH, 85% aq. MeOH 및 n-Hexane)을 이용하여 지방세포내 중성지방 생성 및 지방세포 분화조절 인자 발현에 미치는 영향을 검토하였다. 마우스 유래 지방전구세포3T3-L1을 지방세포로 분화하여 Oil Red O염색법으로 지방 세포 분화정도를 확인한 결과, 갯질경이 분획물에 의해 지방세포의 형성이 농도의존적으로 억제되었다. 또한 지방생성조절에 관여하는 전사인자 PPARγ, C/EBPα 및 SREBP-1c의 발현을 mRNA와 단백질 수준에서 확인한 결과 갯질경이 분획물 처리시 지방세포 분화 인자의 발현이 유의적으로 감소하였다. 지방세포 분화에 관여하는 것으로 알려진 MAPK 신호 전달 경로를 확인한 결과 갯질경이 분획물 처리군에서 p38, ERK 및 JNK의 인산화가 억제되었다. 용매 분획물중에서 H₂O 및 n-Hexane 분획물이 가장 우수한 지방생성 억제활성을 나타내었는데 이는, 분획물 중 페놀 또는 지방 유도체에 의한 것으로 사료된다. 본 연구 결과로부터 갯질경이 분획물의 MAPK 신호전달 경로 억제를 통한 항비만 효과를 확인하였으며, 나아가 건강기능성 식품 소재로서의 개발 가능성이 기대된다. Limonium tetragonum, an edible halophyte that grows on salt marshes in Korea, is thought to possess various health benefits (e.g., antioxidant, antitumor, and hepatoprotective). In the present study, different solvent partitioned subfractions, water (H₂O), buthanol (n-BuOH), 85% aqueous methanol (85% aq. MeOH), and hexane (n-hexane), from crude extract of L. tetragonum were tested for their ability to prevent adipogenesis in differentiating 3T3-L1 preadipocytes. The treatment of differentiating 3T3-L1 preadipocytes with L. tetragonum subfractions (LTFs) resulted in suppressed adipogenesis and reduced expression of adipogenesis-related transcription factors such as peroxisome proliferator-activated receptor gamma (PPARγ), CCAATT/enhancer-binding protein alpha (C/EBPα), and sterol regulatory element- binding protein 1c (SREBP-1c) at both mRNA and protein levels. In addition, the LTF treatment notably decreased the levels of phosphorylated p38, extracellular signal-regulated kinase (ERK), and c-Jun N-terminal kinase (JNK) of the mitogen-activated protein kinase (MAPK) pathway in association with PPARγ-linked adipogenesis. Among all the tested LTFs, H₂O and n-hexane were the most effective in lowering lipid accumulation and regulating the adipocyte differentiation via PPARγ pathway. Taken together, the results indicated that the H₂O and n-hexane LTFs contain bioactive compounds that may exhibit significant antiadipogenesis activity by downregulation of the PPARγ pathway and inactivation of the MAPK signal pathway in 3T3-L1 preadipocytes.

        A Simple Carbamidomethylation-Based Isotope Labeling Method for Quantitative Shotgun Proteomics

        In this study, we present a new isotope-coded carbamidomethylation (iCCM)-based quantitative proteomics, as a complementary strategy for conventional isotope labeling strategies, with providing the simplicity, ease of use, and robustness. In iCCM-based quantification, two proteome samples can be separately isotope-labeled by means of covalently reaction of all cysteinyl residues in proteins with iodoacetamide (IAA) and its isotope (IAA-$^{13}C_2$, $D_2$), denoted as CM and iCCM, respectively, leading to a mass shift of all cysteinyl residues to be + 4 Da. To evaluate iCCM-based isotope labeling in proteomic quantification, 6 protein standards (i.e., bovine serum albumin, serotransferrin, lysozyme, beta-lactoglobulin, beta-galactosidase, and alpha-lactalbumin) isotopically labeled with IAA and its isotope, mixed equally, and followed by proteolytic digestion. The resulting CM-/iCCM-labeled peptide mixtures were analyzed using a nLC-ESI-FT orbitrap-MS/MS. From our experimental results, we found that the efficiency of iCCM-based quantification is more superior to that of mTRAQ, as a conventional nonisobaric labeling method, in which both of a number of identified peptides from 6 protein standards and the less quantitative variations in the relative abundance ratios of heavy-/light-labeled corresponding peptide pairs. Finally, we applied the developed iCCM-based quantitative method to lung cancer serum proteome in order to evaluate the potential in biomarker discovery study.

        Kikuchi`s disease (KD), histiocytic necrotizing lymphadenitis, is a rare self-limited lymphadenopathy, which usually affects young women. KD has been reported to precede, coexist with or follow the diagnosis of other entities, such as systemic lupus erythematosus (SLE), adult-onset Still`s disease, Hashimoto`s disease, and viral infections. In a few cases, KD is associated with cutaneous lupus erythematosus (CLE), without systemic involvement. Herein, we report the first Korean case of KD associated with lupus erythematous profundus in a 9-year-old boy. (Korean J Dermatol 2012;50(4):341∼345)

        Inhibition of Na^(+), K^(+)-ATPase has been implicated in the pathogenesis of hypertension via its effect on smooth muscle reactivity and myocardial contractility. We recently demonstrated that translationally controlled tumor protein (TCTP) interacts with the 3rd cytoplasmic domain of Na^(+), K^(+)-ATPase α₁-subunit and acts as its cytoplasmic repressor. Therefore, we hypothesized that repression of Na^(+), K^(+)-ATPase by overexpressed TCTP might underlie the development of hypertension. In the present study, we confirmed that transgenic mice overexpressing TCTP developed systemic arterial hypertension at about 6 weeks after birth. Vascular smooth muscle of TCTP-overexpressing transgenic mice also displayed augmented contractile response to vasoconstrictors and attenuated relaxation response to vasodilators. These responses seem to be caused by reduced Na^(+), K^(+)-ATPase activity and increased intracellular calcium, suggesting that inhibition of Na^(+), K^(+)-ATPase by overexpression of TCTP is involved in the pathogenesis of hypertension. This study provides a new link between alteration of sodium pump activity and hypertension in vivo, and suggests that TCTP might be a therapeutic target for the treatment of hypertension.
