        Mercury Resistance and Removal Mechanisms of Pseudomonas sp. Isolated Mercury-contaminated Site in Taiwan

        A new strain of Pseudomonas sp. was isolated from mercury (Hg)-contaminated sites in Taiwan. This bacterium removed more than 80% of Hg present in the culture medium at 12 h incubation and was chosen for further analysis of the molecular mechanisms of Hg tolerance/removal abilities in this Pseudomonas sp. We used RNA-seq, one of the next-generation sequencing methods, to investigate the transcriptomic responses of the Pseudomonas sp. exposed to 60 mg/L of Hg<sup>2+</sup>. We de novo assembled 4,963 contigs, of which 10,533 up-regulated genes and 5,451 down-regulated genes were found to be regulated by Hg. The 40 genes most altered in expression levels were associated with tolerance to Hg stress and metabolism. Functional analysis showed that some Hg-tolerant genes were related to the mer operon, sulfate uptake and assimilation, the enzymatic antioxidant system, the HSP gene family, chaperones, and metal transporters. The transcriptome were analyzed further with Gene Ontology (GO) and Cluster of Orthologous Groups (COGs) of proteins and showed diverse biological functions and metabolic pathways under Hg stress.

        Long-term structural analysis and stability assessment of three-pinned CFST arches accounting for geometric nonlinearity

        Due to creep and shrinkage of the concrete core, concrete-filled steel tubular (CFST) arches continue to deform in the long-term under sustained loads. This paper presents analytical investigations of the effects of geometric nonlinearity on the long-term in-plane structural performance and stability of three-pinned CFST circular arches under a sustained uniform radial load. Non-linear long-term analysis is conducted and compared with its linear counterpart. It is found that the linear analysis predicts long-term increases of deformations of the CFST arches, but does not predict any long-term changes of the internal actions. However, non-linear analysis predicts not only more significant long-term increases of deformations, but also significant long-term increases of internal actions under the same sustained load. As a result, a three-pinned CFST arch satisfying the serviceability limit state predicted by the linear analysis may violate the serviceability requirement when its geometric nonlinearity is considered. It is also shown that the geometric nonlinearity greatly reduces the long-term in-plane stability of three-pinned CFST arches under the sustained load. A three-pinned CFST arch satisfying the stability limit state predicted by linear analysis in the long-term may lose its stability because of its geometric nonlinearity. Hence, non-linear analysis is needed for correctly predicting the long-term structural behaviour and stability of three-pinned CFST arches under the sustained load. The non-linear long-term behaviour and stability of three-pinned CFST arches are compared with those of twopinned counterparts. The linear and non-linear analyses for the long-term behaviour and stability are validated by the finite element method.

        맥거핀 효과가 애니메이션 캐릭터의 무동기에 미치는 영향 분석 : 드림웍스 애니메이션 <쿵푸 팬더>를 중심으로

        본고에서는 애니메이션 <쿵푸 팬더>에 나타난 맥거핀 효과를 분석함으로 캐릭터 무동기에 연구했다. 맥거핀 효과의 애니메이션 서사적 합리적 설정은 궁금증을 유발하고 시청자의 욕구를 불러일으킬 것이다. 애니메이션에서 맥거핀 효과는 캐릭터들이 쫓아오거나 피할 수 있는 목표로 삼아 캐릭터들의 욕구를 자극하고 욕망이 동기를 부여한다. 맥거핀 효과는 캐릭터의 무동기 상태에 행동을 촉발시켜 캐릭터에 접근하거나 행동을 회피한다. 맥거핀 효과는 캐릭터의 접근과 회피 행동에 대한 억제와 촉진작용을 한다. 애니메이션에서는 맥거핀 효과를 이용해 캐릭터가 움직여 스토리를 발전시킬 수 있다. 마지막으로 전형적인 맥거핀 효과를 가진 애니메이션 <쿵푸 팬더>를 중심으로 분석했으며, 연구에서 제기된 맥거핀 효과가 캐릭터에 대한 영향을 체계적으로 검증했다. 앞으로 연구에서는 많은 애니메이션을 통해 맥거핀 효과가 캐릭터 무동기와 행동에 미치는 영향과 이에 따른 결과를 연구할 것이다. The paper analyzes the MacGuffin effect in the DreamWorks cartoon Kung Fu Panda , and studies the effect of MacGuffin s effect on the character s none-motivation and action. The reasonable setting of MacGuffin effect in the animation narrative will create suspense, satisfying or betraying the audience s expectations by creating a certain expectation or curiosity, thus arousing the audience s desire to watch. In the animation, the MacGuffin effect serves as a target for the character to pursue or avoid, stimulating the character s desire, which generates motivation. The MacGuffin effect plays a motivational inducing character in the unmotivated state, making the character have approaching or avoiding motivation, which determines the positive action or negative action. Finally, the cartoon Kung Fu Panda with typical MacGuffin effect is analyzed, and the mechanism of MacGuffin s effect on the character is systematically verified. In future studies, more animations will be used to study the influence of MacGuffin s effect on character none-motivation and actions, and the results of influence.

        Effect of Argon Gas Pressure on Residual Stress, Microstructure Evolution and Electrical Resistivity of Beryllium Films

        The residual stress in beryllium films fabricated on K9 substrates by using magnetron sputtering deposition is measured by using a curvature method and is theoretically estimated by using the Nix and Clemens (NC) model. The experimental results indicate that the 1.3-μm-thick film is always in a tensile state for pressure variations in the range from 0.4 to 1.2 Pa. When the sputtering gas pressure is increased, the average stress increases at first, after which it decreases by a remarkable amount. The observed descending trend of the tensile stress when the sputtering gas pressure is beyond 0.6 Pa is mainly attributed to the grain size in the film being larger than that in the film when the pressure is below 0.6 Pa. The maximal residual stress of 552 MPa at a sputtering gas pressure of 0.6 Pa is close to the tensile strength (550 MPa) of the corresponding beryllium bulk material and is about 8 times smaller than that calculated by using the N-C model. In addition, the surface morphologies of the as-fabricated films reveal fibrous grains while the cross-sectional morphologies are characterized by a coarsening of columnar grains. The measured electric resistivity of each film strongly depends on its porosity and the sizes of its grains.

        Overexpression of AtNHX1, a Vacuolar Na+/H+ Antiporter from Arabidopsis thalina, in Petunia hybrida Enhances Salt and Drought Tolerance

        PlantNa+/H+ antiporter plays a critical role in plant salt tolerance. In this study, AtNHX1, a vacuolar Na+ /H+ antiporter from Arabidopsis thalina, was introduced into Petunia hybrida using Agrobacterium-mediated methods. Polymerase chain reaction (PCR) and DNA gel blot analysis confirmed that AtNHX1 was successfully integrated into the P. hybrida genomes. Reverse transcription (RT)-PCR analysis indicated that AtNHX1 expressed highly in transgenic plants. It was found that the 35S::AtNHX1 transgenic P. hybrida plants showed much more resistant to high concentrations of NaCl and to water deprivation than the isogenic wild-type (WT) Petunia plants. The transgenic plants accumulated more Na+, K+, and proline in their leaf tissue than that of the WT Petunia plants and maintained high water contents and high ratio of K+/Na+. These results demonstrated that overexpression of the vacuolar Na+/H+ antiporter conferred both salt tolerance and drought tolerance to the transgenic Petunia plants.

        This paper aims for the transparency and visualization of the international intermodal cargo transportation throughout its whole process, achieving a comprehensive monitoring on the multiple transportation means such as by ocean, by air, by land or by rail. Based on Internet-of-Things-based distributed data acquisition technology and the cloud-computing-based big data analysis technology, this paper gives out a Multimodal Monitoring technology that can uniformly solve the comprehensive management of multiple transportation vehicles, which includes a service functionality model, a network hierarchy model and a technology system model. By building a Generic Target Monitoring System, it proves the multimodal monitoring resolution is able to effectively monitor the multiple transportation means and provide a fair good database platform of later analysis, distribution and optimization of those vehicles.

      • Effects of Different Raising Ways on the Early Growth and Development Performance of Nanjiang Yellow Goat

        A total of 106 two month-old Nanjiang yellow goats (male were 32, female were 34 and castrated were 40) were divided into treatment group and control group to determine the growth performance. The treatment group was reared in grazing mode, experimental group was pastured with supplement feeding. The results showed that the body weight of male in experiment group was heavier by 9.82%(P<0.05), 11.87%(P<0.01), 16.91%(P<0.01) and 23.99%(P<0.05) than that in control group in 3,4,5 and 6 month-age. The body weight of female in experimental group was heavier by 11.54%(P<0.05), 13.79%(P<0.01), 17.47%(P<0.01) and 21.15%(P<0.01) than that in control group in 3,4,5 and 6 month-old. The body weight of castrator in experimental group was heavier by 1.58%, 1.99%, 13.08%(P<0.01) and 15.76%(P<0.01) than that in control group in 3,4,5 and 6 month-age. The daily increasing weight and body measurements of experimental group is improved more than control group, especially in 6 month-age.

        Circ_0000554 is identified as a cancer‑promoting circRNA in colorectal cancer by regulating the miR‑1205/LASP1 axis

        Background: Colorectal cancer (CRC) is a prevalent malignant tumor with poor prognosis. Circular RNAs (circRNAs) are key regulators in the progression of CRC. Our study aimed to disclose the role of circ_0000554 in CRC. Methods: The expression of circ_0000554, miR-1205 and LIM and SH3 protein 1 (LASP1) was measured by quantitative real-time polymerase chain reaction (qRT-PCR). Cell proliferation, invasion and migration were monitored using cell counting kit-8 (CCK-8) assay, EdU assay, transwell assay and wound healing assay respectively. The protein levels of C-myc, matrix metallopeptidase 2 (MMP-2) and LASP1 were detected by western blot. Tumor formation assay in nude mice was conducted to explore the role of circ_0000554 in vivo. The association between miR-1205 and circ_0000554 or LASP1 was identified by dual-luciferase reporter assay and RNA immunoprecipitation (RIP) assay. Results: circ_0000554 was upregulated in CRC tissues and cells, high circ_0000554 expression was significantly linked to shorter overall survival. Downregulation of circ_0000554 restrained cell growth and metastasis while promoted apoptosis in vitro, and suppressed tumorigenesis of CRC in vivo. Furthermore, mechanism study and rescue experiments confirmed miR-1205 could be sponged by circ_0000554 and its inhibitor reversed the inhibitory effect of circ_0000554 silencing on CRC progression. LASP1 was a target gene of miR-1205 and the upregulation of LASP1 overturned miR-1205-induced effects on CRC cells. Circ_0000554 could elevate LASP1 expression via interacting with miR-1205.

        Numerical evaluation on the intraparticle transfer in butylene oxidative dehydrogenation fixed-bed reactor over ferrite catalysts

        Ferrite catalysts with high intraparticle pore volume and surface area are frequently used in theoxidative dehydrogenation of butylene. Whether the non-inclusion of intraparticle transfer limitation infixed-bed reactors for oxidative dehydrogenation over ferrite catalysts in previous studies is appropriateremains unclear. In this study, we attempt to verify this process using a multi-scale modelingtechnology. The multi-scale model consists of a porous medium model and a single particle model underthe oxidative dehydrogenation condition. This model can predict the influences of intraparticle transferon the main component distributions in reactors and demonstrate that the intraparticle transferlimitation is obvious.
