      • MiR-454 Prompts Cell Proliferation of Human Colorectal Cancer Cells by Repressing CYLD Expression

        Previous studies have shown that miR-454 plays an important role in a variety of biological processes in various human cancer cells. However, the underlying mechanisms of this microRNA in colorectal cancer (CRC) cells remain largely unknown. In the present study, we investigated the miR-454 role in CRC cell proliferation. We found that miR-454 expression is markedly upregulated in CRC tissues and CRC cells compared with the matched tumor adjacent tissues and the FHC normal colonic cell line. Ectopic expression of miR-454 promoted the proliferation and anchorage-independent growth of CRC cells, whereas inhibition of miR-454 reduced this effect. Bioinformatics analysis further revealed cylindromatosis (CYLD), a putative tumor suppressor as a potential target of miR-454. Data from luciferase reporter assays showed that miR-454 directly binds to the 3'-untranslated region (3'-UTR) of CYLD mRNA and repressed expression at both transcriptional and translational levels. In functional assays, CYLD-silenced in miR-454-in-transfected SW480 cells have positive effect to promote cell proliferation, suggesting that direct CYLD downregulation is required for miR-454-induced CRC cell proliferation. In sum, our data provide compelling evidence that miR-454 functions as an onco-miRNA, playing a crucial role in the promoting cell proliferation in CRC, and its oncogenic effect is mediated chiefly through direct suppression of CYLD expression.

        Static Shape Adjustment and Actuator Layered Optimization for Planar Phased Array Satellite Antenna

        Shape accuracy is of great importance in space antennas, especially in large-scale planar phased array antenna. To maintain the performance of the planar phased array antenna, shape accuracy must be strictly controlled. This paper proposes an optimization method using diagonal cables as actuators to achieve the shape adjustment. As for shape control, actuator placement has a significant impact on the controlled shape accuracy. Misplaced actuators always lead to control problems, and the desired performance may not be achieved with any choice of control forces, so the actuator placement optimization is needed. The optimization problem is challenging because of the mixed discrete–continuous nature of design variables: the actuator placement corresponds to discrete variables and the control forces are continuous variables. A layered optimization method is proposed in this paper to solve the optimal actuator placement and the corresponding control forces. A genetic algorithm is applied in the outside layer to achieve the optimization of the actuator placement, and the quadratic programming method is used in the inside layer to get the corresponding optimal control forces. The proposed layered optimization method is successfully applied to the large-scale planar phased array antenna. Using this method, the influence of the number of actuators on the controlled shape accuracy is also studied.

        Deflection of battened beams with shear and discrete effects

        This paper presents a theoretical analysis for determining the transverse deflection of simply supported battened beams subjected to a uniformly distributed transverse quasi-static load. The analysis considers not only the shear effect but also the discrete effect of battens on the transverse deflection of the battened beam. The analytical solution is obtained using the principle of minimum potential energy. Numerical validation of the present analytical solution is accomplished using finite element methods. The present analytical solution shows that the shear effect on the transverse deflection of battened beams increases with the cross-section area of the main member but decreases with the cross-section area of the batten. The longer the battened beam is, or the larger the moment of inertia of the main member is, the smaller the shear effect will be.

        Loss of RTN3 phenocopies chronic kidney disease and results in activation of the IGF2-JAK2 pathway in proximal tubular epithelial cells

        Reticulon 3 (RTN3) is an endoplasmic reticulum protein that has previously been shown to play roles in neurodegenerative diseases, but little is known about its function in the kidneys. The aim of the present study was to clarify the roles of RTN3 in chronic kidney disease (CKD) and kidney fibrosis. In this study, RTN3 levels were measured in kidney tissues from healthy controls and CKD or kidney fibrosis patients. An RTN3-null mouse model was generated to explore the pathophysiological roles of RTN3 in the kidneys. The underlying mechanisms were studied in primary proximal tubular epithelial cells and HEK293 cells in vitro. The results showed that (1) a reduction in RTN3 in mice induces CKD and kidney fibrosis; (2) decreased RTN3 expression is found in patients with CKD; (3) RTN3 plays critical roles in regulating collagen biosynthesis and mitochondrial function; and (4) mechanistically, RTN3 regulates these phenotypes by interacting with GC-Rich Promoter Binding Protein 1 (GPBP1), which activates the IGF2-JAK2-STAT3 pathway. Our study indicates that RTN3 might play crucial roles in CKD and kidney fibrosis and that a reduction in RTN3 in the kidneys might be a risk factor for CKD and kidney fibrosis.

        국내 중ㆍ고등학교 보건교과서의 구강보건 내용 분석

        Objectives: This study investigated the contents and structure of chapters on oral health to determine the quality of middle and high school health textbooks. We also comparatively analyzed the learning objectives of these textbooks according to the oral health curriculum proposed by the Virginia Department of Health (VDH) and South Carolina Department of Education (SCDE). Methods: The structure and educational contents of eight health textbooks (middle school, five; high school, three) published in 2019 were analyzed methodologically. We compared the structure and contents between the middle and high school curricula. We also carried out comparisons among five publishing companies. Results: Of the eight health textbooks examined, five (middle school, four; high school, one) had chapters on oral health. In terms of structure, textbooks published by company A had the lowest percentage (0.96%) of pages on oral health. The highest percentage (1.50%) was found in a middle school textbook published by company D. We noted subject-level differences between the curriculum proposed by VDH, SCDE, and that of the published textbooks. There were more keywords pertaining to the definition and development of oral diseases than those for its prevention and management. Conclusions: It is critically important to impart oral health education to middle and high school students for self-management of oral health during adolescence, a period characterized by both rapid physical and psychological development. Therefore, we recommend that all health textbooks should include chapters on oral health. Further, in the health textbooks that already include oral health information, we propose that the revised curricula should include key learning objectives.

        Complex roles of the stroma in the intrinsic resistance to gemcitabine in pancreatic cancer: where we are and where we are going

        Pancreatic ductal adenocarcinoma (PDAC) is among the most devastating human malignancies. The poor clinical outcome in PDAC is partly attributed to a growth-permissive tumor microenvironment. In the PDAC microenvironment, the stroma is characterized by the development of extensive fibrosis, with stromal components outnumbering pancreatic cancer cells. Each of the components within the stroma has a distinct role in conferring chemoresistance to PDAC, and intrinsic chemoresistance has further worsened this pessimistic prognosis. The nucleoside analog gemcitabine (GEM) is usually the recommended first-line chemotherapeutic agent for PDAC patients and is given alone or in combination with other agents. The mechanisms of intrinsic resistance to GEM are an active area of ongoing research. This review highlights the important role the complex structure of stroma in PDAC plays in the intrinsic resistance to GEM and discusses whether antistroma therapy improves the efficacy of GEM. The addition of antistroma therapy combined with GEM is expected to be a novel therapeutic strategy with significant survival benefits for PDAC patients.

      • Unraveling the Role of Earth-Abundant Fe in the Suppression of Jahn-Teller Distortion of P′2-Type Na<sub>2/3</sub>MnO<sub>2</sub>: Experimental and Theoretical Studies

        <P>Layered Na<SUB>2/3</SUB>MnO<SUB>2</SUB> suffers from capacity loss due to Jahn-Teller (J-T) distortion by Mn<SUP>3+</SUP> ions. Herein, density functional theory calculations suggest Na<SUB>2/3</SUB>[Fe<SUB><I>x</I></SUB>Mn<SUB>1-<I>x</I></SUB>]O<SUB>2</SUB> suppresses the J-T effect. The Fe substitution results in a decreased oxygen-metal-oxygen length, leading to decreases in the <I>b</I> and <I>c</I> lattice parameters but an increase in the <I>a</I> lattice constant. As a result, the capacity retention and rate capability are enhanced with an additional redox pair associated with Fe<SUP>4+/3+</SUP>. Finally, the thermal properties are improved, with the Fe substitution delaying the exothermic reaction and reducing exothermic heat.</P> [FIG OMISSION]</BR>

        Biphasic effects of TGFβ1 on BMP9-induced osteogenic differentiation of mesenchymal stem cells

        We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]
