The Two Echelon Open Vehicle Routing Problem: Optimization of Crowdshipping Based Parcel Delivery

Crowdshipping is a revolutionary concept of the sharing economy. In this study, two carriers are used to perform the following expedition: the truck starts from the depot to complete part of the deliveries, shares part of the load with the crowdshipper at the relay point, and the private driver selected as the crowdshipper continues from that point onward. This study proposes the two-echelon open vehicle routing problem with crowdshipping (2EOVRP-CS) and formulates a mathematical model to determine the crowdshipper, parcel relay location, truck route, and crowdsource route. A tangible nested genetic algorithm (NGA) is proposed, and its efficiency is demonstrated by comparison with CPLEX and genetic algorithm (GA). A real case study is investigated in Xi'an city to test the applicability of the proposed model. The results show that using crowdshipping instead of truck delivery alone can save approximately 14% of the total cost and 26% of truck vehicle miles traveled (VMT). Moreover, several sensitivity analyses are performed. The results show that crowdshipping is sensitive to the detour limit and the time value of carriers. For the detour limit, after the acceptable detour distance increases by 8%, the total cost can be reduced by up to 5.94%.
Identification and Characterization of EDT1 Conferring Drought Tolerance in Rice

Basic Leucine Zipper (bZIP) transcription factors(TFs) play important roles in many processes, especially inabiotic stress response in plants. In this study, we characterizeda new gene EHANCED DROUGHT TOLERANCE 1 (EDT1),member of group E of bZIP transcription factor family inrice. The EDT1 protein contains one bZIP domain, oneputative nuclear localization signal (NLS) and six conservedphosphorylation sites. The expression of EDT1 is suppressedby several abiotic stresses, such as cold, droutht, and salt. Corresponding with expression patterns, several stress-associatedcis-acting elements were found in the EDT1 promoter. Theresults of subcellular localization and transactivation abilityanalyses indicated that EDT1 was localized in the nucleusand functioned as a nuclear protein, with its transactivationactivity primarily located in N-terminal. Transgenic riceoverexpressing EDT1 showed drought tolerance that hasbeen significantly improved. Real-time PCR analysis revealedthat some stress-related genes, such as OsbZIP12, SNAC1,OsLEA3, OsbZIP16, OsbZIP10 and OsABI2 were up-regulatedin EDT1 overexpression lines. These results indicate thatEDT1 plays a positive role in drought tolerance and providesvaluable targets for breeding drought-tolerant rice cultivars.

<P>We report the discovery of an ultra-luminous quasar J030642.51+185315.8 (hereafter J0306+1853) at redshift 5.363, which hosts a supermassive black hole with M-BH = (1.07 +/- 0.27) x 10(10) M-circle dot. With an absolute magnitude M-1450 = -28.92 and a bolometric luminosity L-bol similar to 3.4 x 10(14) L-circle dot, J0306+1853 is one of the most luminous objects in the early universe. It is not likely to be a beamed source based on its small flux variability, low radio loudness, and normal broad emission lines. In addition, a z = 4.986 damped Ly alpha system (DLA) with [M/H] = -1.3 +/- 0.1, among the most metal-rich DLAs at z greater than or similar to 5, is detected in the absorption spectrum of this quasar. This ultra-luminous quasar puts strong constraints on the bright end of the quasar luminosity function and massive end of the black hole mass function. It will provide a unique laboratory for the study of BH growth and the co-evolution between a BH and the host galaxy with multi-wavelength follow-up observations. The future high-resolution spectra will give more insight into the DLA and other absorption systems along the line of sight of J0306+1853.</P>

The aim of this study was to screen for polypeptides binding specifically to LoVo human colorectal cancer cells using a phage-displayed peptide library as a targeting vector for colorectal cancer therapy. Human normal colorectal mucous epithelial cells were applied as absorber cells for subtraction biopanning with a c7c phage display peptide library. Positive phage clones were identified by enzyme-linked immunosorbent assay and immunofluorescence detection; amino acid sequences were deduced by DNA sequencing. After 3 rounds of screening, 5 of 20 phage clones screened positive, showing specific binding to LoVo cells and a conserved RPM motif. Specific peptides against colorectal cancer cells could be obtained from a phage display peptide library and may be used as potential vectors for targeting therapy for colorectal cancer.
Upregulation and Clinicopathological Significance of Long Non-coding NEAT1 RNA in NSCLC Tissues

Background: Recent reports have shown that nuclear enriched abundant transcript 1 (NEAT1), a long noncoding RNA (lncRNA), contributes to the precise control of gene expression and is related to several human malignancies. However, limited data are available on the expression and function of NEAT1 in lung cancer. The major objective of the current study was to profile the expression and clinicopathological significance of NEAT1 in non-small cell lung cancers (NSCLCs). Materials and Methods: NEAT1 expression in 125 NSCLC cases and paired adjacent non-cancer tissues was assessed by real-time quantitative reverse transcription-PCR (qRT-PCR). Relationships between NEAT1 and clinicopathological factors were also investigated. Results: The relative level of NEAT1 was $6.98{\pm}3.74$ in NSCLC tissues, significantly elevated as compared to that of the adjacent non-cancer lung tissues ($4.83{\pm}2.98$, p<0.001). The area under curve (AUC) of high expression of NEAT1 to diagnose NSCLC was 0.684 (95% CI: 0.619~0.750, p<0.001). NEAT1 expression was positively correlated with patient age (r=-2.007, p=0.047), lymphatic metastasis (r=-2.731, p=0.007), vascular invasion (r=-3.617, p=0.001) and clinical TNM stage (r=-4.134, p<0.001). Conclusions: This study indicates that NEAT1 might be associated with oncogenesis and progression in NSCLC, and suggests application in molecular targeted therapy.
Amelioration of radiation-induced liver damage by p-coumaric acid in mice

Radiation-induced liver damage (RILD) is a spiny problem in radiotherapy or other circumstances that exposure to radiation. The need for radioprotective agent is increasing to protect liver tissue. This study aimed to explore the hepatoprotective effect of p-coumaric acid (CA) against RILD. C57BL/6 male mice were exposed to 4 Gy irradiation and administrated with CA for 4 days starting on the same day of irradiation. Mice were sacrificed to obtain blood and liver tissues on day 3.5 or 14 post irradiation, respectively. The blood and liver tissues were collected. As compared with the only irradiated group, CA supplementation improved liver morphology, decreased serum alanine aminotransferase and aspartate aminotransferase, inhibited BCL2-associated X (BAX) protein expression, and improved the mice hematopoietic function. CA at the dose of 100 mg/kg body weight showed better effect compared to the other doses. Thus, CA might possess potential to protect against RILD.

The imbalance between the proliferation and apoptosis of B-cell precursors is an important contributor to the pathogenesis of B-cell precursor acute lymphoblastic leukemia (BCP-ALL), while its specific regulatory mechanism remains perplexing. This study aimed to expound the underlying mechanism of the proliferation and apoptosis of BCP-ALL cells from the perspective of non-coding RNA. In this study, long non-coding RNA colorectal neoplasia differentially expressed (LncRNA CRNDE) was upregulated in the bone marrow of BCP-ALL patients and BCP-ALL cell lines (NALM-6 and RS4;11). Functionally, LncRNA CRNDE knockdown restrained cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. The subsequent investigation confirmed that LncRNA CRNDE bound to miR-345-5p and negatively regulated miR-345-5p expression. The overexpression of miR-345-5p suppressed cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. Further experiments revealed that miR-345-5p downregulated cyclic AMP response element-binding protein (CREB) expression by targeting its mRNA directly. CREB overexpression reversed the effect of miR-345-5p mimic on cell proliferation and apoptosis in NALM-6 and RS4;11 cells. Finally, in vivo experiments showed that LncRNA CRNDE knockdown prolonged the survival of mice xenotransplanted with NALM-6 cells. In conclusion, LncRNA CRNDE upregulated CREB expression by suppressing miR-345-5p, thus promoting cell proliferation and reducing cell apoptosis in BCP-ALL.
