      • Biosynthesis of L-ergothioneine by Regulating Sulfur Assimilation in Metabolically Engineered Corynebacterium glutamicum

        L-Ergothioneine (EGT), which is known as a sulfur-containing histidine compound, has been regarded as a strong antioxidant in the cosmetic and food industries by using as an antiaging material and food preservative. It is commonly extracted from edible mushrooms, but it still has limited productivity. Here, we developed metabolically engineered Corynebacterium glutamicum producing EGT without any precursor addition. We first reinforced sulfur assimilation by exchanging a native promoter and enhanced the cysteine synthetic pathway. In this strain, EGT production was 1.48-fold more increased compared to that of control. To confirm the effects of enhanced L-histidine production, the pentose phosphate pathway was reinforced by introducing a synthetic promoter. This strain produced a 1.89-fold increased EGT compared to the strain in which only the sulfur assimilatory pathway was enhanced. Finally, the strain, which was consolidated all sulfur assimilatory, histidine, and cysteine pathways, was developed, and it synthesized a 2.95-fold increase of EGT. In this study, we suggested major-related pathways for EGT synthesis, and we first synthesized EGT by accumulating precursors in C. glutamicum.

        The purpose of this paper is for the classification of effective meditation techniques and analysis of the prerequisites concerned with consciousness level, Classification methodology based on consciousness map by the David R. However, meditation used as a tool for the relaxation and traditional religious awakening and enlightenment through the self-completion, It can not use same meditation method, apply to overcome stress and to seek mental health mindfulness(MBSR). So, demand has been increased development the new scientific meditation techniques in order to meditate more objective and efficient because of the meditation depending on what the purpose of pursuing different. Accordingly, the 17 stages of human consciousness from shame 20 to 1000 enlightenment was classified into 3 level, according to this classification, analyzed the prerequisite for the achievement of the purposes of meditation.

        다중 생체신호 측정 웨어러블 디바이스 기반 환자 모니터링 시스템 설계

        병원 현장에서 환자 모니터링 시스템을 적용하기 위해서는 환자감시장치에서 기본적으로 다루고 있는 주요 생체신호의 측정과 데이터의 분석이 가능해야 한다. 본 논문에서는 이러한 요구사항을 수용하고자 심전도와 산소포화도 측정이 가능한 웨어러블 디바이스와 환자 모니터링 시스템을 구현하였다. 구현된 시스템은 측정된 생체신호를 블루투스를 통해 간호스테이션 상의 서버로 전송되고 환자 모니터링 시스템에서 의료진이 확인할 수 있는 그래프 파형과 수치로 표현된다. 설계한 시스템을 통해 수집된 데이터와 기존 장비에서 얻어진 데이터를 비교하여 본 시스템의 유효성을 검증하였다. In order to apply the patient monitoring system to the hospital field, it is necessary to be able to measure and analysis data the major bio-signals that are basically covered by the existing patient monitoring system. We have implemented a wearable device and the patient monitoring system for measuring ECG and oxygen saturation. The implemented system transmits the measured bio-signal to the server on the nursing station via Bluetooth. It is represented by graph waveforms and numerical values that can be checked by the medical staff in the patient monitoring system. The validity of this system is verified by comparing the data collected through the designed system with the data obtained from the conventional equipment.

        Effects of Dietary Carbohydrases on Fecal Microbiome Composition of Lactating Sows and Their Piglets

        Corn-soybean meal diets are commonly used in the pork industry as a primary source of energy and protein. However, such a diet generally contains non-starch polysaccharides (NSPs) which present a challenge in finding ways to improve their availability and digestibility. Dietary multi-carbohydrases (MCs) have been proposed as an efficient approach to utilize NSPs, and can result in improved growth performance and host intestinal fitness. In this study, we evaluated the effects of MC in lactation diets on gut microbiota composition of lactating sows and their litters. The experimental design contained two dietary treatments, a diet based on corn-soybean meal (CON), and CON supplemented with 0.01% multigrain carbohydrases (MCs). Sow and piglet fecal samples were collected on days 7 and 28 after farrowing. Based on the results from 16S rRNA gene amplicon sequencing, MC led to changes in species diversity and altered the microbial compositions in lactating sows and their piglets. Specifically, the MC treatment induced an increase in the proportions of Lactobacillus in piglets. Clostridium and Spirochaetaceae showed a significantly reduced proportion in MC-treated sows at day 28. Our results support the beneficial effects of dietary carbohydrases and their link with improved production due to better host fitness outcomes and gut microbiota composition.

        Recently, the U-Healthcare market is growing at a rapid pace by development of wireless network market and increase of people"s attention on health care. An example of this is wearable devices with healthcare function, mobile health apps, etc. Despite this development, hospital monitoring system has been still used in constraining way. In order to identify health of hospital patients have to connect electrodes and various sensors, medical devices the front on the patient"s body. In this study, in order to solve these problems designed the smart garment and the monitoring system. Through this, be able to measure for the patient"s health status in the non-restraint way. Smart garment is interconnecting to the module and sensor, battery and clothes through digital yarn, and it"s able to detachable. like this, transmit to the monitoring system through Bluetooth for measured bio-signal data of patients on non-restraint.

        현대사회의 양극화 현상과 물질문명의 한계, 지구환경의 불확정성의 증가로 인류는 어려움에 처해 있어서, 이제 서양은 동양의 명상과 깨달음에 눈을 돌리고 있는데, 동양은 아직도 앞서간 서양과 물질을 쫒고 있다. 그러나 우리가 추구하는 대상으로서 깨달음은 아직도 분명한 정의와 객관화가 부족하고 주관적 막연함 속에서 효율적이지 못한 수행법으로 시간과 에너지를 낭비하고 있다. 이에 따라 깨달음의 객관화를 시도한 선행연구를 고찰하고, 깨달음의 전제조건으로서 나없음을 분석하였으며, 자기무화를 이루기 위한 방법론으로서 정화 수련법을 제안하였다. Today humanity is in trouble. Modern society, the limits of the material civilization of the polarization and the global environment due to the increase in the uncertainty. So the West is turning to meditation of orient. However the East is still to be ahead of the West. And, we are pursuing a clear definition of the enlightenment as the realization that objectification is still insufficient, and waste of time and energy by ineffective in subjective meditation. So, attempted to objectify the previous research about enlightenment were investigated. And, proposed self-purification methodology as a prerequisite for enlightenment for achieving the self-naughting.

        성인에서 신체활동이 골절 발생에 미치는 영향: 한국 지역사회 코호트 자료에 기반

        Regular exercise and a certain level of physical activity reduce the mortality rate in the elderly. The purpose of this study was to investigate the effect of physical activity on the prevention of fracture in the middle aged or older in Korea. The basic data are based on the Ansan and Ansung community cohort studies of the Korean Genome and Epidemiology Study conducted by the Korea Centers for Disease Control and Prevention in 2001, and the fracture data from the third survey in 2005 to the sixth survey in 2011. The physical activity of the aged in the 40s was mostly distributed in the World Health Organization (WHO) recommended range of 7.5 to 30.0 metabolic equivalent·hr/wk, and the activity was gradually divided into the low and high groups in the 50s and 60s. In the 60s, the risk of fracture was reduced to 0.63 times compared to that of the 50s when physical activity was the recommended level (odds ratio, 0.63; p<0.001). For Korean adults, there was no significant difference in fracture incidence according to the amount of physical activity in the middle-aged people. However, for the elderly aged 60 and over, the risk of fracture decreased when the WHO recommended level of activity was performed, and the risk increased when less or more activities were performed.

        <P>Embryonic stem (ES) cells are used in cell therapy and tissue engineering due to their ability to produce different cells types. However, studies of ES cells that are derived from fertilized embryos have raised concerns about the limitations imposed by ethical and political considerations. Therefore, many studies of stem cells use the stem cells that are derived from unfertilized oocytes and adult tissue. Although parthenogenetic embryonic stem (ESP) cells also avoid ethical and political dilemmas and can be used in cell-based therapy, the ESP cells exhibit growth retardation problems. Therefore, to investigate the potential for muscle growth from genetically modified ESP cells, we established four ES cell types, including normal embryonic stem (ESN) cells, ESP cells, ESP cells that overexpress the insulin-like growth factor?2 (Igf2) gene (ESI) and ESP cells with down-regulated H19 gene expression (ESH). Using these cells, we examined the expression profiles of genes that were related to imprinting and muscle using microarrays. The gene expression patterns of ESI and ESH cells were similar and were more closely related to the ESN pattern than that of the ESP cells. Differentiated ESH cells exhibited increased expression of bone morphologic protein?4 (BMP4), which is a mesoderm marker, compared with the differentiated ESI cells. We showed that Igf2 expression was induced by H19 silencing in the ESP cells via hypermethylation of the H19 imprinting control region?1 (ICR1). Moreover, the proportion of ESH-derived chimera was slightly higher than those produced from the ESP cells. In addition, we detected increased cell proliferation in the MEF cells following H19 knock-down. These results indicate that the ESH cells may be a source of cell-based therapy for conditions such as muscular atrophy.</P>
