      • Deoxycytidine kinase and deoxyguanosine kinase of Lactobacillus acidophillus R-26 are colinear products of a single gene

        Three of the four deoxynucleoside kinases required for growth of Lactobacillus acidophilus R-26 exist as heterodimeric pairs specific for deoxyadenosine(dAK) and deoxycytidine (dCK) or dAK and deoxyguanosine (dGK). However, only two tandem genes, dak/dgk, are found, and are expressed only as dAK/dGK in transformed Escherichia coli. Sequencing peptides spanning 63% of the native dCK subunit revealed a sequence identical to that deduced from dgk(beginning MTVIVL…), except that dCK lacks residues 2 and 3 (dCK is M-IVL;dGK is ·TVIVL). Also, mass spectrometry indicates that native dCK and dGK subunits are identical in mass adjusted for the first three residues. Furtheromore, the native enzymes have identical isoelectric pH values, indicating an equal number of charged residues. To enable E. coli to express peptide having the native dCK sequence, codons 2 and 3 were deleted from the dgk portion of the tandem genes, resulting in expression of protein having the specificities and regulatory properties of dAK/dCK, including heterotropic stimulation of dAK activity by deoxycytidine or dCTP (not deoxyguanosine or dGTP) and end-product inhibition of the respective activities by dATP and dCTP. Subcloning normal and mutant dgk yielded homodimeric dGK and dCK, respectively. The dCK homodimer strongly resembles human dCK with a low ?? for deoxycytidine, the ability to phosphorylate deoxyadenosine and deoxyguanosine at much higher ?? values, and end-product inhibition by dCTP. Thus two distinct and specific enzymes evidently are derived from a single Lactobacillus gene. The mechanism by which this occurs in vivo has yet to be elucidated.

      • Comparison of the implementation status of weight control and physical activities between South Korean and Japanese university students

        Purpose: Adolescent Japanese girls have a strong desire to be thin and, therefore, engage in many unhealthy weight-control behaviors such as dieting. In recent years, this drive for thinness has spread to include boys as well as girls. Meanwhile, the desire to be thin is also increasing among adolescents in South Korea. The present study aimed to investigate the correlation between weight-control (WC) behaviors and physical activity (PA) among university students in Japan and South Korea. Method: The subjects comprised 1,833 university students in Japan (male, 454; female, 456) and South Korea (male, 449; female, 474). All subjects were asked whether they engaged in WC behaviors and were queried about the intensity (mild, moderate, and vigorous), number of days per week, and duration of PA. The correlations between sex, WC behaviors, and PA were investigated by country using a chi-square test. Multiple correspondence analysis (MCA) was then applied to investigate overall correlations. Result: The rates of WC behaviors were higher among female students than male students in both countries (58.7% vs. 18.4% in Japan [χ2=151.9***] and 61.1% vs. 42.3% in South Korea [χ2=32.6***]). Regarding PA, male students tended to exercise more days per week than female students in South Korea for all PA intensities. The same tendency was observed in Japan in relation to moderate and vigorous PA, but not for mild PA which was the same for males and females. Two-dimensional (2D) plots based on the results of MCA showed a correlation between not engaging in WC behaviors and performing PA among male students and between engaging in WC behaviors and not performing PA among female students in both Japan and South Korea for mild PA (1D: α=.455, eigenvalue=1.519; 2D: α=.336, eigenvalue=1.337). This tendency became increasingly marked for moderate (1D: α=583, eigenvalue=1.776, 2D: α=.463, eigenvalue=1.531) and vigorous (1D: α=.656, eigenvalue=1.970, 2D: α=.470, eigenvalue=1.544) PA. Conclusion: Female university students in Japan characteristically engaged in WC behaviors without PA, and this tendency became more marked as exercise intensity increased. Female university students in South Korea also engaged in WC behaviors, but they also performed moderate PA; and WC behaviors were associated with PA. Male university students in South Korea engaged in WC behaviors, but these were associated with PA and the number of days per week; and durations of PA were greater than those of female university students in South Korea at all PA intensities. The rates of WC behaviors were low among male university students in Japan; however, the number of days per week and duration of PA were greater than those of male university students in South Korea at all PA intensities. The present findings show that the correlations between WC behaviors and PA observed among university students in South Korea are stronger in Japan.

        Verification and Mitigation of Seismic Failure in Concrete Piers under Near-field Earthquakes

        This paper verifies the difference of the seismic behavior and seismic damage of the neighboring two reinforced concrete piers damaged by the 1995 Hyogoken Nanbu earthquake. The two piers were almost the same size, carrying slightly different dead load, and were provided with the same reinforcement arrangement except the amount of longitudinal reinforcement at the bottom portion of the piers. The pier with more reinforcement was completely collapsed due to this near field earthquake by shear failure at the longitudinal reinforcement cut-off while the other was only damaged at the bottom by flexure even though the longitudinal reinforcement cut-off was also existed at the mid height of the pier. According to the results of the pseudo dynamic test, the seismic damage was recognized to be greatly dependent on the ground motion characteristics even though the employed ground motions had the same peak acceleration. The severe damage was observed when the test employed the seismic wave that had strong influence to the longer period range compared to the initial natural period of the pier. On the other hand, based on the similar model experiment, the defect of gas-pressure welded splice of longitudinal reinforcement was revealed to save the piers against collapse due to the so-called fail-safe mechanism contrary to the intuitive opinion of some researchers. It was concluded that the primary cause of the collapse of the pier was the extremely strong intensity and peculiar characteristics of the earthquake motion according to both the site-specific and the structure-specific effects.

      • Smart System Identification of Super High-Rise Buildings using Limited Vibration Data during the 2011 Tohoku Earthquake

        A method of smart system identification of super high-rise buildings is proposed in which super high-rise buildings are modeled by a shear-bending system. The method is aimed at finding the story shear and bending stiffnesses of a specific story only from the horizontal floor accelerations. The proposed method uses a set of closed-form expressions for the story shear and bending stiffnesses in terms of the limited floor accelerations and utilizes a reduced shear-bending system with the same number of elements as the observation points. A difficulty of prediction of an unstable specific function in a low frequency range can be overcome by introducing an ARX model and discussing its relation with the Taylor series expansion coefficients of a transfer function. It is demonstrated that the shear-bending system can simulate the vibration records with a reasonable accuracy. It is also shown that the vibration records at two super high-rise buildings during the 2011 Tohoku (Japan) earthquake can be simulated with the proposed method including a technique of inserting degrees of freedom between the vibration recording points. Finally it is discussed further that the time-varying identification of fundamental natural period and stiffnesses can be conducted by setting an appropriate duration of evaluation in the batch least-squares method.

      • Comparison of Radical Cystectomy and Chemoradiotherapy in Patients with Locally Advanced Bladder Cancer

        The aim of this study was to evaluate the clinical outcomes of radical cystectomy (RC) and concurrent chemoradiotherapy (CRT) with methotrexate, vinblastine, doxorubicin, and cisplatin (MVAC) in patients with locally advanced bladder cancer (BC). From December 2000 to February 2012, 72 patients with locally advanced BC (T3-4a, N0 or N+, M0) received either RC or CRT. RC with bilateral pelvic lymph node dissection including the common iliac region as the standard procedure. Patients in the CRT group received one cycle of MVAC followed by radiotherapy with a half dose of MVAC and then two more cycles of MVAC. Standard fractionation at a daily dose of 1.8-2.0 Gy was used, with a median total dose of 50 Gy (range, 45-60 Gy). The 3-year progression-free survival (PFS) rates in the RC and CRT groups were 56.2% and 25.6%, respectively (p=-0.015) and the 3-year overall survival (OS) rates were 63.5% and 48.1% (p=0.272). Multivariate Cox proportional hazards regression analysis with application of a propensity score indicated that RC was a significant predictor of PFS (p=0.033) but not of OS (p=0.291). Among patients with locally advanced BC, PFS was significantly prolonged in the RC group compared with the CRT group. However, RC was not a significant predictor of OS. Although the sample size in this study was small, the results suggest that patient background and postoperative quality of life should be considered when choosing treatment strategy for locally advanced BC.

        돈황사본 지론종문헌 Stein 613V 제 21장「經辨五住地煩惱義」에 나타난아비달마 교리에 대해

        The aim of my study is to clarify the position of the Dunhuang manuscript Stein 613 verso, one of the most eminent texts of so-called Dilun (地論) School. For that reason, I examined the Abhidharma doctrines found in Chapter 21 of the Stein 613 verso, “Jingbian wuzhudi fannao yi” (經辨五住地煩惱義, the doctrine of the five *sthavāsabhūmis explained in the sūtra). My findings are as follows: 1. Almost all of the following are based on the *Satyasiddhiśāstra (or *Tattvasiddhiśāstra): the Abhidharma doctrines found in the “zhang” (章, the proposition) of the “Jingbian wuzhudi fannao yi,” the definition of avidyā and of *ekadṛṣṭiṣṭhavāsabhūmi, the three *rāgasthavāsabhūmis and the avidyāvāsabhūmi. 2. It is the concept of “the equal catuḥsatya,” found in the definition of *ekadṛṣṭiṣṭhavāsabhūmi, that we should especially examine in these doctrines. This concept is based on the “theory of extinguishing kleśas by contemplating nirodhasatya,” a theory insisted on by the *Satyasiddhiśāstra. We can find the same concept in Baoliang’s (寶亮) (444-509) commentaries on the Mahāyāna Mahāparinirvāṇasūtra (MPS) which is contained in the Da banniepan jing jijie (大般涅槃經集解, Selected Commentaries on the MPS) and in Fascicle 4 of the Da niepan jing yiji (大涅槃經義記卷第四, Commentary on the MPS) copied in Datong (大統) 5 (CE 539) of the West Wei (西魏). 3. Comparing the interpretations of the concept of “the equal catuḥsatya” which are seen in some commentaries on the MPS that were written in the Southern and Northern dynasties, we can determine that the basis of the interpretations shifted from the *Satyasiddhiśāstra to the “pitan” (毘曇, Sarvāstivāda) in the following order: the Da banniepan jing jijie → Fascicle 4 of the Da banniepan jing yiji → Dunhuang manuscript Niepan jing shu (涅槃經疏, the Commentary on the MPS) (BD00260) → Huiyuan’s (慧遠) (523-592) Da banniepan jing yiji (大般涅槃經義記, the Commentary on the MPS). I thus conclude that the “Jingbian wuzhudi fannao yi” is located in the early stage of the current history of Chinese Buddhist thought. 잘 알려진 것처럼 돈황사본 중에 현존하는 지론종 관련 문헌들은 지론종연구를 위한 1차 자료 중 하나이나, 각 문헌의 정확한 성립지나 성립년대, 성립순서 등에 대해서는 아직 충분한 고찰이 이루어지지 않았다. 본고에서는 돈황사본 지론종 문헌 가운데 가장 대표적인 것 중 하나인 Stein 613V의 제21장「經辨五住地煩惱義」에 나타난 아비달마적 교리를 분석함으로써 Stein 613V의사상사적 위치를 밝히고자 한다. 본고의 고찰결과는 다음과 같다. 1. Stein 613V 「經辨五住地煩惱義」의 “章” 부분에서 無明의 정의와 見一處住 地⋅三愛住地⋅無明住地의 정의에 보이는 아비달마 교리는 거의 대부분 『成 實論』에 기초한다. 2. 이 중에서 특히 주목해야 할 것은 見一處住地의 정의 중에 보이는 “四諦 平等”이라는 개념이다. 이것은 『成實論』의 滅諦斷惑說에 기반한 것이며, 『大 般涅槃經集解』의 寶亮(444-509)釋이나 西魏 大統 5년(539) 서사된 『大涅槃經義 記』 권제4에도 동일한 용례를 찾을 수 있다. 3. “四諦平等”의 개념에 대한 남북조 시대 『涅槃經』 주석서의 주석내용을비교하면 『大般涅槃經集解』 → 大統 5년 서사 『大涅槃經義記』 권제4 → 돈황사본 『涅槃經疏』(BD00260) → 慧遠(523-592) 『大般涅槃經義記』의 순으로 『成 實論』에서 「毘曇」으로의 해석이 이행되는 형태를 확인할 수 있다. Stein 613V 「經辨五住地煩惱義」는 이러한 사상사적 흐름에서 초기 단계에 위치한다.

      • Experimental Verification of Electric Vehicle Using Electric Double Layer Capacitor

        This paper discusses to conduct experimental verification of two types of micro electric vehicles (EV) in order to realize improvement in electric mileage and shorten a charging time of the battery. First, electric double layer capacitor (EDLC) systems to use as a secondary battery are proposed. The internal resistance of EDLC is small compared with a rechargeable battery, and it is suitable for momentary charge-discharge of EV. Next, control circuits of the capacitors to increase the regenerative electric power are utilized. Then, a novel method to charge a main battery of the EV is introduced. Finally, experimental results demonstrate the validity of the proposed method.
