<B>Objective</B><P>Anticitrullinating autoantibodies are specific markers for rheumatoid arthritis (RA). A functional haplotype of 4 exonic single-nucleotide polymorphisms (SNPs) in a citrullinating enzyme, peptidylarginine deiminase 4 (PADI4), was shown to be associated with susceptibility to RA in a Japanese population and was shown to increase the stability of PADI4 messenger RNA. However, the association was not confirmed in 4 subsequent studies involving Caucasian RA patients living in the UK, a French Caucasian population, and a Spanish population. The aim of the current study was to investigate the association of SNPs in the PADI4 gene with RA in a Korean population.</P><B>Methods</B><P>Four exonic SNPs of the PADI4 gene (padi4_89, padi4_90, padi4_92, and padi4_104) were genotyped in 545 unrelated patients with RA and 392 controls, using the MassArray SNP genotyping system. Allelic, genotypic, and haplotypic associations of the SNPs with RA susceptibility were examined using the chi-square test and multivariate logistic regression analyses.</P><B>Results</B><P>Increased RA susceptibility was significantly associated with the minor alleles of padi4_89 (P = 2.3 × 10<SUP>−5</SUP>), padi4_90 (P = 2.3 × 10<SUP>−5</SUP>), padi4_92 (P = 2.1 × 10<SUP>−5</SUP>), and padi4_104 (P = 1.1 × 10<SUP>−3</SUP>) and the haplotype carrying the 4 minor alleles (P = 1.0 × 10<SUP>−4</SUP>). Genotypes carrying the minor alleles and HLA–DRB1 shared epitope (SE) alleles (P = 9.4 × 10<SUP>−21</SUP>) were also associated with increased RA susceptibility. The genotypic associations were sustained among individuals who did not carry any SE alleles, except in the case of padi4_104. Individuals carrying the risk SNPs and/or SE alleles were more susceptible to RA than were individuals carrying neither risk SNPs nor SE alleles.</P><B>Conclusion</B><P>The PADI4 SNPs and haplotypes are associated with RA susceptibility in Koreans. Thus, the association of PADI4 with RA may depend on genetic heterogeneity between Asians and Europeans.</P>
TP53BP2 locus is associated with gastric cancer susceptibility

<P>We investigated the association of the TP53BP2 locus with gastric cancer susceptibility in a Korean population. We assayed 9 single nucleotide polymorphisms (SNP) in an 82.5 kb region that included the TP53BP2 locus in 233 male gastric cancer patients and 390 unaffected healthy male controls. The allelic frequencies of 4 SNP within TP53BP2, g.206692C>T, g.198267A>T, g.164895G>A and g.152389A>T, differed significantly between cases and controls (p ≤ 0.0376). When compared to carriers of non-risk alleles, individuals homozygotic for each of the risk alleles had a 50% increase in risk of gastric cancer (age-adjusted odds ratio [OR] =≥ 1.48; p ≤ 0.0371). Furthermore, these 4 significantly associated SNP were in strong linkage disequilibrium (r<SUP>2</SUP> ≥ 0.51). Haplotype analysis showed that individuals with the CAGA haplotype, consisting of the risk alleles at each SNP, had a 1.55-fold higher risk for gastric cancer than individuals with the haplotype TTAT, consisting of the non-risk alleles at each SNP (OR = 1.55; 95% confidence interval [CI] = 1.13–2.14; p = 0.00705). Two other SNP were not polymorphic in the study subjects, whereas the other 3 SNP, located toward the outside of the TP53BP2 locus, were not associated with gastric cancer susceptibility. Although the location of the pathogenic variant is not yet known, our results suggest that the TP53BP2 locus is associated with susceptibility to gastric cancer in the Korean population. © 2005 Wiley-Liss, Inc.</P>

<P>Ionotropic glutamate receptors (iGluRs) are ligand-gated nonselective cation channels that mediate fast excitatory neurotransmission. Although homologues of the iGluRs have been identified in higher plants, their roles are largely unknown. In this work we isolated a full-length cDNA clone (RsGluR) encoding a putative glutamate receptor from small radish. An RsGluR: mGFP fusion protein was localized to the plasma membrane. In Arabidopsis thaliana overexpressing the full-length cDNA, glutamate treatment triggered greater Ca2+ influx in the root cells of transgenic seedlings than in those of the wild type. Transgenic plants exhibited multiple morphological changes such as necrosis at their tips and the margins of developing leaves, dwarf stature with multiple secondary inflorescences, and retarded growth, as previously observed in transgenic Arabidopsis overexpressing AtGluR3.2 [Kim et al. (2001)]. Microarray analysis showed that jasmonic acid (JA)-responsive genes including defensins and JA-biosynthetic genes were up-regulated. RsGluR overexpression also inhibited growth of a necrotic fungal pathogen Botrytis cinerea possibly due to up-regulation of the defensins. Based on these results, we suggest that RsGluR is a glutamate-gated Ca2+ channel located in the plasma membrane of higher plants and plays a direct or indirect role in defense against pathogen infection by triggering JA biosynthesis.</P>

<P><B>Abstract</B></P><P><B>BACKGROUND:</B></P><P>A primary inhibitor of plasminogen activators, SERPINE1 (serpin peptidase inhibitor 1, clade E, member 1, also known as plasminogen activator inhibitor type 1), is an important regulator in tumorigenesis and is highly expressed in many cancers.</P><P><B>METHODS:</B></P><P>Five tag single nucleotide polymorphisms (SNPs) and 1 insertion polymorphism within <I>SERPINE1</I> were genotyped in 1101 unrelated Korean individuals (a case group of 612 patients with gastric cancer and a control group of 489 healthy individuals). Associations with susceptibility to diffuse‐type gastric cancer (DGC) and intestinal‐type gastric cancer were assessed using multivariate logistic regression analyses adjusted for age and sex. Allelic differences in the contribution to gene expression were measured using luciferase assays.</P><P><B>RESULTS:</B></P><P>SNP c.1162+162C>T (rs2227692) in intron 7 was associated with susceptibility to DGC but not with susceptibility to intestinal‐type gastric cancer. The minor allele‐carrying genotypes C/T and T/T had 1.6‐fold greater odds of DGC than the C/C genotype (<I>P</I> = .00084). This SNP was linked to a repeat‐number variation c.1162+604AAAG(11_17), a deletion (del) variation c.1162+664_1162+673del, and another SNP c.1162+859T>A (rs2070683) in intron 7 based on the sequencing of 5 patients and 5 controls. The risk haplotype of the 4 variations exhibited a 30% greater gene expression level than the nonrisk haplotype in luciferase reporter assays (<I>P</I> = .025). In contrast, DGC susceptibility was not associated with the c.−1969_−1968insG polymorphism (rs1799768) in the promoter, commonly known as 4G/5G, in which the minor 5G allele is less active in transcription than the major 4G allele.</P><P><B>CONCLUSIONS:</B></P><P>An association between <I>SERPINE1</I> and DGC susceptibility was observed with 4 correlated polymorphisms in intron 7 rather than the 4G/5G polymorphism in the promoter, although all polymorphisms affected gene expression. A Feasibility Study on TETRA System Application for Train Control Systems

TETRA communication system is very versatile system which can transmit voice + data and packet data optimized. Direct mode operation permits to connect between mobiles when mobile stain is out of coverage of networks. It can be more secure communication channel for railway signaling systems. Railway signaling systems use many of wayside signal equipment, which require many maintenance efforts and budget. Many railway authorities want to reduce and replace the wayside equipment. Radio based signaling systems are one of candidate for replacing the conventional signaling systems. The radio based signaling systems can replace track circuit and wayside signal. The radio systems permit to connect between control centers and trains. The radio systems have to ensure high quality of the connectivity more or equal to the existed track circuits. We studied the application of TETRA systems for railway radio systems for bridging between train control centers and trains. We provide an operation scenario for radio based train control system to ensure the safety require to the existed trains control system and satisfied the existed operational availability. We showed the data transmission speed, maximum bit error rate, and data coding for the radio-based signal system using TETRA systems.

Building Information Modeling (BIM) is the virtual representation of the physical and functional characteristics of a facility from inception onward. As such, it serves as a shared information repository for collaboration throughout a facility’s lifecycle. BIM is a new paradigm which can perform a diverse engineering activity such as design, simulation, structure analysis, etc and support a based information model in construction industry. In these days, many construction companies tried to perform a project efficiently by constructing and using BIM and carried out a research related in PMIS(Project Management Information System), etc. But a real case of research focused on 3D modeling object and control and share of engineering information is insufficient. In this research, we suggest an efficient BIM model and information share system based PLM for collaboration based on analyzing and researching a case of constructing BIM when perform construction project.
