P-10 : Role of Polypyrimide Tract-Binding Protein in HCV RNA Replication

Background : Translation initiation of the hepatitis C virus (HCV) is regulated by an internal ribosome entry site (IRES) which requires polypyrimidine tractbinding protein (PTB), which binds to the 5``-untranslated region (UTR) and the 3``-UTR and the end of the core coding region of HCV RNA, for its function. Aims of this Study are to determine the role of PTB in HCV replication. Methods : siRNA technology was used to specifically knockdown the PTB expression in the cell. We determined whether down-expression of PTB affects the EMCV IRES-mediated translation, and examined whether interferon (IFN) down-regulates PTB expression. Results : In this study, we discovered that the level of PTB expression was efficiently decreased by PTB-specific siRNA. PTB knockdown expression by siRNA significantly reduced the efficiency of cell colony formation induced by HCV RNA replication. PTB siRNA resulted in a significant reduction of PTB expression and therein HCV RNA replication in a subgenomic HCV replicon-bearing Huh7 cell line, suggesting that PTB is required for efficient HCV replication in vivo. Result: PTB siRNA does not significantly affect the efficiency of the EMCV IRES-mediated translation. IFN-α resulted in an inhibition of PTB expression, suggesting a potential mechanism by which IFN inhibits HCV replication. Discussion : PTB siRNA efficiently knockdowns the level of PTB expression and subsequently resulted in a significant reduction of HCV RNA replication, indicating that PTB is required for efficient HCV RNA replication in vivo. It appears that IFN-α inhibits PTB expression, suggesting that inhibition of HCV replication by IFN might be due to suppression of cellular proteins that are required for HCV RNA replication.

본 논문에서는 RFID를 이용하여 사용자를 인식한 후 사용자의 생체신호(혈압, 혈당, 체지방)를 측정하여 자가진단을 할 수 있는 지능형 헬스케어 시스템을 구현하였다. 구현한 헬스케어 자가진단 지능형 시스템은 RFID리더기, 생체신호측정기(혈압계, 혈당계, 체지방측정기), 데이터베이스 서버역할을 하는 컴퓨터, 자가진단 결과를 출력하는 프린터로 구성된 키오스크형태로 이루어졌으며 데이터베이스에서 보유한 사용자 정보 및 측정된 정보 데이터를 비교분석한 후 사용자의 건강상태를 자가진단할 수 있다. 구현된 시스템은 병원에 가지 않더라도 간단히 자가진단을 할 수 있으며, 회사나 학교 등에서 응용할 수 있다. In this paper, we implemented the intelligent healthcare system self-diagnosis that can achieve self-diagnosis by measured bio-signal(blood pressure, blood sugar, body fat monitor) after the recognize a user to access using RFID. The implemented healthcare self-diagnosis intelligent system is consist of kiosk structure that is RFID reader, bio-signal measuring instrument(hemadynamometer, glucometer, body fat monitor), computer for a part of database server and printer for print the result of self-diagnosis. It can achieve self-diagnosis of a user after compare and analyze the measured data and information of a user from database. The implemented system can make simple self-diagnosis even if not take a physical examination at hospital and apply to company, school, etc.
Vasoconstriction Potency Induced by Aminoamide Local Anesthetics Correlates with Lipid Solubility

<P>Aminoamide local anesthetics induce vasoconstriction <I>in vivo</I> and <I>in vitro</I>. The goals of this <I>in vitro</I> study were to investigate the potency of local anesthetic-induced vasoconstriction and to identify the physicochemical property (octanol/buffer partition coefficient, pKa, molecular weight, or potency) of local anesthetics that determines their potency in inducing isolated rat aortic ring contraction. Cumulative concentration-response curves to local anesthetics (levobupivacaine, ropivacaine, lidocaine, and mepivacaine) were obtained from isolated rat aorta. Regression analyses were performed to determine the relationship between the reported physicochemical properties of local anesthetics and the local anesthetic concentration that produced 50% (ED<SUB>50</SUB>) of the local anesthetic-induced maximum vasoconstriction. We determined the order of potency (ED<SUB>50</SUB>) of vasoconstriction among local anesthetics to be levobupivacaine > ropivacaine > lidocaine > mepivacaine. The relative importance of the independent variables that affect the vasoconstriction potency is octanol/buffer partition coefficient > potency > pKa > molecular weight. The ED<SUB>50</SUB> in endothelium-denuded aorta negatively correlated with the octanol/buffer partition coefficient of local anesthetics (<I>r</I><SUP>2</SUP> = 0.9563; <I>P</I> < 0.001). The potency of the vasoconstriction in the endothelium-denuded aorta induced by local anesthetics is determined primarily by lipid solubility and, in part, by other physicochemical properties including potency and pKa.</P>

Background: The purpose of this study was to evaluate the association between metabolic syndrome and periodontal disease in Korean adults. Methods: This study analyzed the screening data of 12,686 adults aged ≥19 years, including demographic characteristics, lifestyle habits, and each component of metabolic syndrome, obtained from the sixth Korea National Health and Nutrition Examination Survey database. Periodontal health status was measured by the community periodontal index. Subjects with three or more risk factors were considered as having metabolic syndrome. The Rao-Scott chi-square test was performed to assess the relationships between demographic characteristics, lifestyle habits, components of metabolic syndrome, and periodontal disease. Logistic regression analysis was performed based on the complex sample to evaluate the relationship between metabolic syndrome and periodontal disease. Results: The prevalence of periodontal disease was higher among the subjects with advancing age, lower average household income and education level, those working in production, residents of eup-myeon areas, in past or current smoker, those with excessive alcohol consumption habit in a week, and reduced brushing frequency and the use of oral care products (p<0.001). Each component of metabolic syndrome was associated with higher prevalence of periodontitis in the subjects with abnormal than in those with normal levels (p<0.001). The prevalence of periodontal disease in subjects with metabolic syndrome was approximately 1.443 times higher than that in normal subjects (odds ratio,1.443; p<0.001). Conclusion: This study confirmed the association between metabolic syndrome and periodontitis in Korean adults, and further studies will be needed to determine the causal relationship between the two conditions.
Germination and Seedling Growth Affected by Seed Specific Gravity

The amount of salt to make seed sorting solution of the specific gravity of 1.13 was reconsidered and determined as 3.8 kg salt in 18 L water, which is lower amount than currently used. Five rice cultivars were examined. Percent germination and seedling emergence were not similar. Seedling emergence rate of Japonica varieties, Nampyungbyeo and Daerypbyeo-1 were 87% and 95% under specific gravity of 1.13, respectively. Seedling emergence rate of Tongil type variety, Dasanbyeo was as high as 67% in specific gravity of 1.06. Seedling emergence rate of waxy rice, Hwasunchalbyeo and Aranghangchalbyeo were examined. Seedling emergence rate was 94% in both cultivars in specific gravity of 1.04. Seedling emergence rate was same in specific gravity of 1.08 which is generally used for selecting seed currently. Early growth (plant height, leaf number, and dry weight) were not significantly different by specific gravity within species. In all cultivars except waxy rice, highest seedling emergence rate was observed in specific gravity of 1.13 which is currently used for selection and decreased as specific gravity is lowed. However, considering total amount of seeds in each group of specific gravity, amount of seed in lower specific gravity group is relatively small and total seedling emergence rate within variety dose not show big difference. However, if seeds with low speicific gravity are produced due to the bad grain filling condition and consequently total seed content of low specific gravity increases, results will be differnt. Reduction in total growth and yield could occur. It will be important to comply with the seed sorting criterion of 1.13 for Japonica, 1.06 for Tongil, and 1.04 for waxy rice variety to ensure the maximum rice growth and yield.
디메틸포름아미드 취급 사업장의 작업환경 및 보건관리 실태와 노출요인 조사

This study was aimed to assess the status of working environment, health care management status and cause of exposure in manufactories using dimethylformamide (DMF). For the purpose, airborne concentration of DMF in the workplaces and N-methylformamide (NMF) in worker`s urine were measured with job type and process. In addition, management of local exhaust ventilation system (LEV) and personal protective equipment (PPE) was evaluated at 35 work places (107 workers) located in Busan and Gimhae area. Mean DMF concentrations in work places by job type and process were of high level measured in printing and record media reproduction (5.23ppm) and flaking process (2.48ppm). Workers in adhesive job were measured a large amount of urine NMF (21.59mg/ℓ). 98.1% of DMF handling workers were provided respirators, but 67.3% of those workers used them. The main reasons for not using respirators were inconvenience and difficulty of breathing. Airborne concentrations of DMF were higher in the workplaces in which LEVs were working abnormally, but there was not statistically significant. In addition, the urine NMF levels were correlated with management of LEV within the workers who did not use the respirators (p<0.048). These results implied that LEV should be installed and maintained properly to protect the workers from the exposure to DMF. Management of PPE should be also necessary to protect the workers from chemical hazards.
