Bovine Viral Diarrhea Virus Infection Induces Autophagy in MDBK Cells

        Qiang Fu,Huijun Shi,Yan Ren,Fei Guo,Wei Ni,Jun Qiao,Pengyan Wang,Hui Zhang,Chuangfu Chen 한국미생물학회 2014 The journal of microbiology Vol.52 No.7

Bovine viral diarrhea virus (BVDV) is an enveloped, positive-sense, single-stranded RNA virus that belongs to the genus Pestivirus (Flaviviridae). The signaling pathways and levels of signaling molecules are altered in Madin-Darby Bovine Kidney (MDBK) cells infected with BVDV. Autophagy is a conservative biological degradation pathway that mainly eliminates and degrades damaged or superfluous organelles and macromolecular complexes for intracellular recycling in eukaryotic cells. Autophagy can also be induced as an effective response to maintain cellular homeostasis in response to different stresses, such as nutrient or growth factor deprivation, hypoxia, reactive oxygen species exposure and pathogen infection. However, the effects of BVDV infection on autophagy inMDBK cells remain unclear. Therefore, we performed an analysis of autophagic activity after BVDV NADL infection using real-time PCR, electron microscopy, laser confocal microscopy, and Western blotting analysis. The results demonstrated that BVDV NADL infection increased autophagic activity and significantly elevated the expression levels of the autophagy-related genes Beclin1 and ATG14 inMDBK cells. However, the knockdown of Beclin1 and ATG14 by RNA interference (RNAi) did not affect BVDV NADL infection-related autophagic activity. These findings provided a novel perspective to elaborate the effects of viral infection on the host cells.

Efficient RNA interference for three neuronally-expressed genes in Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

        Chen Xu,Li Guo-Qing,Wan Pin-Jun,Fu Qiang 한국응용곤충학회 2017 Journal of Asia-Pacific Entomology Vol.20 No.2

In the brown planthopper Nilaparvata lugens, introduction of double-stranded RNA (dsRNA) successfully knocks down genes transcribed in several types of tissues. However, whether RNA interference (RNAi) can effectively reduce the transcripts of neuronally-expressed genes remains undetermined. In the present paper, we selected three neuronally-expressed genes respectively encoding prothoracicotropic hormone (NlPTTH), pigment-dispersing factor (NlPDF) and short neuropeptide F (NlsNPF). The transcripts of the three genes were detectable throughout all tested developmental stages, and were mainly restricted to the nervous system of N. lugens. We introduced dsRNA originated from each of the three neuronally-expressed genes at a dose of 300 ng into the nymphal body cavity by microinjection. The target mRNA was successfully knocked down. Moreover, silencing of NlPTTH downregulated the transcripts of two ecdysteroidogenesis genes (NlPHM and NlDIB), and surpressed the expression of an ecdysone-response genes (NlFTZ-F1). Furthermore, RNAi of NlPTTH caused typical 20- hydroxyecdysone deficient phenotypes: nymphal development was delayed and nymphal lethality occurred. In addition, injection of a series of dsNlsDPF solutions at the doses of 300, 60, 12, 2.4 and 0.5 ng into the fourthinstar nymphs decreased its target transcript in a dose-dependent manner: the mRNA levels reduced by 94%, 86%, 57%, 33% and 29%, respectively. Therefore, delivery of dsRNA by microinjection can effectively repress the transcripts of the neuronally-expressed genes in N. lugens.

Damage Localization of Steel Beams with Rotational Restraints

        Guo-Qiang Li,Yang Zhang,Suwen Chen 한국강구조학회 2007 International Journal of Steel Structures Vol.7 No.1

In this paper, a method is presented for damage localization of stel beams with indexes based on natural frequencies becauselocations of beams and not related to the severity of damages is presented. Based on this principle, the universal curve-bankof the natural frequency indexes for damage localization of beams with rotational restraints is established. The effectivenessand universality of the curve-bank have been demonstrated by identifying the damage location of two steel beam specimenswith only first three natural frequencies of the beams. The approach presented may be used for monitoring the damagelocalization of beam-type structures, such as bridges.

Behavior and simplified analysis of steel-concrete composite beams subjected to localized blast loading

        Guo-Qiang Li,Su-Wen Chen,Tao-Chun Yang 국제구조공학회 2009 Structural Engineering and Mechanics, An Int'l Jou Vol.32 No.2

Finite element simulations are increasingly used in structural analysis and design, especially in cases where complex structural and loading conditions are involved. Due to considerable progresses in computer technology as well as nonlinear finite-element analysis techniques in past years, it has become possible to pursue an accurate analysis of the complex blast-induced structural effects by means of numerical simulations. This paper aims to develop a better understanding of the behavior of steel-concrete composite beams (SCCB) under localized blast loading through a numerical parametric study. A finite element model is set up to simulate the blast-resistant features of SCCB using the transient dynamic analysis software LS-DYNA. It is demonstrated that there are three dominant failure modes for SCCB subjected to localized blast loading. The effect of loading position on the behavior of SCCB is also investigated. Finally, a simplified model is proposed for assessing the overall response of SCCB subjected to localized blast loading.

      • LY294002 Induces G0/G1 Cell Cycle Arrest and Apoptosis of Cancer Stem-like Cells from Human Osteosarcoma Via Down-regulation of PI3K Activity

        Gong, Chen,Liao, Hui,Wang, Jiang,Lin, Yang,Qi, Jun,Qin, Liang,Tian, Lin-Qiang,Guo, Feng-Jing Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.7

Osteosarcoma, the most common primary mesenchymal malignant tumor, usually has bad prognosis in man, with cancer stem-like cells (CSCs) considered to play a critical role in tumorigenesis and drug-resistance. It is known that phosphatidylinositol 3-kinase (PI3K) is involved in regulation of tumor cell fates, such as proliferation, cell cycling, survival and apoptosis. Whether and how PI3K and inhibitors might cooperate in human osteosarcoma CSCs is still unknown. We therefore evaluated the effects of LY294002, a PI3K inhibitor, on the cell cycle and apoptosis of osteosarcoma CSCs in vitro. LY294002 prevented phosphorylation of protein kinase B (PKB/Akt) by inhibition of PI3K phosphorylation activity, thereby inducing G0/G1 cell cycle arrest and apoptosis in osteosarcoma CSCs. Further studies also demonstrated that apoptosis induction by LY294002 is accompanied by activation of caspase-9, caspase-3 and PARP, which are involved in the mitochondrial apoptosis pathway. Therefore, our results indicate PI3K inhibitors may represent a potential strategy for managing human osteosarcoma via affecting CSCs.

Immunization of BALB/c mice with Brucella abortus 2308DwbkA confers protection against wild-type infection

        Zhi-qiang Li,Dan Gui,Zhi-hua Sun,Jun-bo Zhang,Wen-zhi Zhang,Hui Zhang,Fei Guo,Chuangfu Chen 대한수의학회 2015 Journal of Veterinary Science Vol.16 No.4

Brucellosis is a zoonotic disease that causes animal and human diseases. Vaccination is a major measure for prevention of brucellosis, but it is currently not possible to distinguish vaccinated animals from those that have been naturally infected. Therefore, in this study, we constructed the Brucella (B.) abortus 2380 wbkA mutant (2308DwbkA) and evaluated its virulence. The survival of 2308DwbkA was attenuated in murine macrophage (RAW 264.7) and BALB/c mice, and it induced high protective immunity in mice. The wbkA mutant elicited an anti-Brucella-specific immunoglobulin G response and induced the secretion of gamma interferon. Antibodies to 2308DwbkA could be detected in sera from mice, implying the potential for use of this protein as a diagnostic antigen. The WbkA antigen would allow serological differentiation between infected and vaccinated animals. These results suggest that 2308DwbkA is a potential attenuated vaccine against 16M. This vaccine will be further evaluated in sheep.

Rapid Expression of Bm46 in Bombyx mori Cell Lines, Larvae and Pupae

Wang, Haiyan,Chen, Keping,Guo, Zhongjian,Yao, Qin,Wang, Qiang,Mu, Runhong Korean Society of Sericultural Science 2007 International Journal of Industrial Entomology Vol.15 No.1

In this study, ORF 46 of Bombyx mod nucleopolyhedrovirus(Bm46) fused with EGFP was expressed in Bombyx mod cell lines, larvae and pupae by BmNPV Bacmid system. Bm46 and EGFP were cloned into donor plasmid pFastBacHTb, which was transformed to competent DH10B cells containing helper and BmNPV bacmid by site-specific transposition. Recombinant bacmid was used to transfected BmN-4 cells to produce the recombinant baculovirus vBm-Bm46-EGFP. Recombination virus was injected into silkworm larvae and pupae. The expression of the fusion protein was monitored by examining green fluorescence using a fluorescent microscope. Intense fluorescence in cells and silkworm was observed at 4 days post-infection, indicating the Bm46-EGFP fusion gene was expressed successfully.

Evolution of microstructure and dielectric properties of (LiCe)-doped Na0.5Bi2.5Nb2O9 Aurivillius type ceramics

        Zhihang Peng,Qiang Chen,Dan Liu,Yadan Wang,Dinquan Xiao,Jian-guo Zhu 한국물리학회 2013 Current Applied Physics Vol.13 No.7

Aurivillius type (NaBi)0.5x(LiCe)xBi2Nb2O9 ceramics were prepared by the standard ceramics route. The single crystal structural ceramics were achieved for all compositions and lattice distortion was decreased by (LiCe) dopants. The temperature dependent dielectric properties revealed that all compositions possess a high Curie-temperature (>780 ℃). A modified CurieeWeiss relationship is used to study the diffuseness behavior of a ferroelectric phase transition indicating the degree of diffuseness of NBN-based ceramics increased with (LiCe) modifications. The degradation of resistance implied a plausible model that Ce4þ ions entered into the B-site of the pseudo-perovskite structure and acted as acceptor doping. Further investigation demonstrated that both electrical conduction and dielectric relaxation processes were associated with the oxygen vacancies produced by the substitution of Nb5+ ions by the Ce4+ ions.
