        HCV-Specific Interleukin-21+CD4+ T Cells Responses Associated with Viral Control through the Modulation of HCV-Specific CD8+ T Cells Function in Chronic Hepatitis C Patients

        Interleukin-21 (IL-21)+CD4+ T cells are involved in the immune response against hepatitis B virus (HBV) by secreting IL-21. However, the role of IL-21+CD4+ T cells in the immune response against chronic hepatitis C (CHC) virus infection is poorly understood. This study aimed to investigate the role of IL-21+CD4+ T cells in CHC patients and the potential mechanisms. The study subjects in-cluded nineteen CHC patients who were grouped by viral load (low, < 106 RNA copies/ml, n = 8; high, > 106 RNA copies/ml, n = 11). The peripheral frequency of HCV-specific IL-21+CD4+ T cells was higher in the low viral load group and was negatively correlated with the serum HCV RNA viral load in all CHC patients. Meanwhile, IL-21+ cells accumulated in the liver in the low viral load group. In vitro, IL-21 treatment increased the expression of proliferation markers and cytolytic molecules on HCV-specific CD8+ T cells. In summary, these findings suggest that HCV-specific IL-21+CD4+ T cells might contribute to HCV control by rescuing HCV-specific CD8+ T cells in CHC patients.

        Registry-based stroke research in Taiwan: past and future

        Stroke registries are observational databases focusing on the clinical information and outcomes of stroke patients. They play an important role in the cycle of quality improvement. Registry data are collected from real-world experiences of stroke care and are suitable for measuring quality of care. By exposing inadequacies in performance measures of stroke care, research from stroke registries has changed how we manage stroke patients in Taiwan. With the success of various quality improvement campaigns, mortality from stroke and recurrence of stroke have decreased in the past decade. After the implementation of a nationwide stroke registry, researchers have been creatively expanding how they use and collect registry data for research. Through the use of the nationwide stroke registry as a common data model, researchers from many hospitals have built their own stroke registries with extended data elements to meet the needs of research. In collaboration with information technology professionals, stroke registry systems have changed from web-based, manual submission systems to automated fill-in systems in some hospitals. Furthermore, record linkage between stroke registries and administrative claims databases or other existing databases has widened the utility of registry data in research. Using stroke registry data as the reference standard, researchers have validated several algorithms for ascertaining the diagnosis of stroke and its risk factors from claims data, and have also developed a claims-based index to estimate stroke severity. By making better use of registry data, we believe that we will provide better care to patients with stroke.

        Influence of Spatial Distribution of Pores on NMR Transverse Relaxation Time in Pebbly Sandstone

        The relationship between transverse relaxation time (T₂) and pore size distribution is the basis of NMR applications for rocks. However, the equations for T₂ are not accurate enough in rocks with complicated pore structures. Taking pebbly sandstone from the northwestern Junggar Basin in China as an example, the aim of this study is to discover the spatial distribution of pores and its influence on T₂. Porosity, permeability, micro-images and T₂ distributions were acquired from rock samples, and pore structure parameters were obtained from binarized thin section images. The results show that as the grain size increases, the proportion of dissolution pores increases and the spatial distribution of pores changes from a random to a clustered pattern. The relaxation of a hydrogen atom takes longer and T₂ is higher in dissolution pores compared with those in intergranular pores. New equations for T₂ that consider the spatial distribution of pores are proposed.

        Advanced Process Control of the Critical Dimension in Photolithography

        This paper describes two run-to-run controllers, a nonlinear multiple exponential-weight moving-average (NMEWMA) controller and a dynamic model-tuning minimum-variance (DMTMV) controller, for photolithography processes. The relationships between the input recipes (exposure dose and focus) and output variables (critical dimensions) were formed using an experimental design method, and the photolithography process model was built using a multiple regression analysis. Both the NMEWMA and DMTMV controllers could update the process model and obtain the optimal recipes for the next run. Quantified improvements were obtained from simulations and real photolithography processes.

        Preparation and Property of Wet-spun Silk Fibroin Filaments with Fibrils Regenerated by Dissolving in CaCl₂-Formic Acid

        In this work, silk fibroin (SF) filament with fibrils regenerated by dissolving in CaCl₂-formic acid (FA) was prepared by wet-spun method at room temperature. Different from traditional dissolved methods, SF solutions obtained by dissolving in CaCl₂-FA preserved fibrils, which have been recognized as the key to the high performance of native silk. The morphology of SF filament was analyzed, very dense filaments with smooth surface and circular, nanofibrils could be observed in longitudinal and cross-sections of filaments. Moreover, the breaking stress of samples was gradually increased with the increase of draw-down ratios. After 3 times drawing, the breaking stress and elongation at break of filament were 276.4±22.6 MPa and 40.8±3.1%, respectively. At the same time, the secondary structure of SF filament was typical β-sheet. In addition, SF filaments showed excellent degradation property, the mass lost of SF filament declined 42% after incubating in protease XIV solution. Above all, the human mesenchymal stem cells (hMSCs) adhered very well on the surface of the filaments, which demonstrated the good biocompatibility of SF filaments, was suitable for application in tissue engineering.

        A novel M2e-multiple antigenic peptide providing heterologous protection in mice

        Swine influenza viruses (SwIVs) cause considerable morbidity and mortality in domestic pigs, resulting in a significant economic burden. Moreover, pigs have been considered to be a possible mixing vessel in which novel strains loom. Here, we developed and evaluated a novel M2e-multiple antigenic peptide (M2e-MAP) as a supplemental antigen for inactivated H3N2 vaccine to provide cross-protection against two main subtypes of SwIVs, H1N1 and H3N2. The novel tetra-branched MAP was constructed by fusing four copies of M2e to one copy of foreign T helper cell epitopes. A high-yield reassortant H3N2 virus was generated by plasmid based reverse genetics. The efficacy of the novel H3N2 inactivated vaccines with or without M2e-MAP supplementation was evaluated in a mouse model. M2e-MAP conjugated vaccine induced strong antibody responses in mice. Complete protection against the heterologous swine H1N1 virus was observed in mice vaccinated with M2e-MAP combined vaccine. Moreover, this novel peptide confers protection against lethal challenge of A/Puerto Rico/8/34 (H1N1). Taken together, our results suggest the combined immunization of reassortant inactivated H3N2 vaccine and the novel M2e-MAP provided cross-protection against swine and human viruses and may serve as a promising approach for influenza vaccine development.

      • Magnetic Resonance Image Manifestations of the Atypical Meningioma

        Through retrospective analysis of 13 cases of magnetic resonance image (MRI) manifestations of atypical meningiomas confirmed by operation and pathology in the First Affiliated Hospital of Xinxiang Medical University, the objective of this study was to evaluate the diagnostic value of MRI in order to improve the accuracy rate of preoperative diagnosis. In this retrospective analysis of MRI findings for atypical meningiomas in First Affiliated Hospital of Xinxiang Medical University from January to July in 2012, the location, morphology and tumor signals and other tumor imaging characteristics were covered. In 13 cases of atypical meningioma patients of this group, most tumors were located at typical sites (10/13), mainly the falx cerebri, parasagittal, convexity, saddle area. Only two cases were at atypical locations, 1 in the cerebellar hemisphere and 1 in a lateral ventricle. Most of the tumors showed T1 and T2 isointensity signals, and necrosis, calcification, and peritumoral edema were always featured. DWI showed isointensity in 11 cases (11/13), and hyperintensity in 2. Some 9 cases had dural tail signs, 12 had accurate positioning (12/13), and 2 were postoperative recurrences. MRI has high value in the diagnosis of atypical meningiomas, with important roles in early clinical diagnosis, treatment and prognosis evaluation.

        Synthesis of Core−Shell Au@TiO<sub>2</sub> Nanoparticles with Truncated Wedge-Shaped Morphology and Their Photocatalytic Properties

        <P>Core−shell Au@TiO<SUB>2</SUB> nanoparticles with truncated wedge-shaped TiO<SUB>2</SUB> morphology have been synthesized successfully by a simple and flexible hydrothermal route. Morphological evolution of TiO<SUB>2</SUB> shells was investigated by transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), and X-ray diffraction technique. It has been revealed that the truncated wedge-shaped TiO<SUB>2</SUB> shells experience an epitaxially segmented orientation growth. Also, the (101) crystal planes of TiO<SUB>2</SUB> crystals grow preferentially on the surface of gold nanocrystals stabilizing the heterointerfaces, then faster [001] growth results in the “budding” process occurs, producing growth sites on the initial deposition TiO<SUB>2</SUB> layers, where the TiO<SUB>2</SUB> crystals grow up into truncated wedge-shaped morphologies. It is also found that morphological evolution of TiO<SUB>2</SUB> shells is dependent on the produced F<SUP>−</SUP> ion concentration from hydrolyzed TiF<SUB>4</SUB> precursors. The produced F<SUP>−</SUP> ions not only facilitate the formation of well-defined wedge-like TiO<SUB>2</SUB> shells, but also contribute to the externally exposed truncated crystal {004} facets. As the representative photocatalyst, the catalytic activities of the resultant core−shell Au@TiO<SUB>2</SUB> nanoparticles were investigated by photoinitiated oxidation degradation of gaseous acetaldehyde. It has been indicated that the nanostructured core−shell Au@TiO<SUB>2</SUB> photocatalyst represents high photocatalytic activity when exposed to UV or visible light irradiation.       • Optimization Model of Reliable Data Storage in Cloud Environment Using Genetic Algorithm

        Massive data storage is one of the great challenges for cloud computing service, and reliable storage of sensitive data directly affects quality of storage service. In this paper, based on analysis of data storage process in cloud environment, the cost of massive data storage is considered to be comprised of data storage price, data migration and communication; and the storage reliability consists of data transmission reliability and hardware dependability. A multi-objective optimization model for reliable massive storage is proposed, in which storage cost and reliability are the objectives. Then, a genetic algorithm for solving the model is designed. Finally, experimental results indicate that the proposed model is positive and effective.
