        Systematic identification and characterization of miRNAs and piRNAs from porcine testes

        microRNAs (miRNAs) and PIWI-interacting RNAs (piRNAs) execute important regulatory roles in testis development and spermatogenesis, while previous studies mainly focus on the expression profiles in immature and mature porcine testes, which may cause a bottleneck for further understanding their complex physiological processes in porcine testes development and spermatogenesis. Thus, we presented the expression and characterization of miRNAs and piRNAs in DS (60-day-old), DN (90-dayold), DT (120-day-old) and DF (150-day-old) pig testes. In total, 12,834,628, 13,359,726, 12,851,249 and 12,938,601 clean reads were generated from these libraries, respectively. 293 mature and 36 novel miRNAs as well as 4923 piRNA clusters were identified from pig testes, and they showed an age-dependent manner. GO enrichment analysis of miRNA target genes and piRNA generated genes showed that they participated widely in regulating the pig spermatogenesis process. In addition, 12 differentially expressed miRNAs were randomly selected to validate using qRT-PCR. Our results provided novel comprehensive expression profiles of miRNAs and piRNAs in pig testes at different stages of sexual maturity, which will promote our knowledge of them in regulating the pig testes development and spermatogenesis process.

        Liver cancer is one of leading digestive malignancies with high morbidity and mortality. There is an urgent need for the development of novel therapies for this deadly disease. It has been proven that asparagus polysaccharide, one of the most active derivates from the traditional medicine asparagus, possesses notable antitumor properties. However, little is known about the efficacy of asparagus polysaccharide as an adjuvant for liver cancer chemotherapy. Herein, we reported that asparagus polysaccharide and its embolic agent form, asparagus gum, significantly inhibited liver tumor growth with transcatheter arterial chemoembolization (TACE) therapy in an orthotopic hepatocellular carcinoma (HCC) tumor model, while significantly inhibiting angiogenesis and promoting tumor cell apoptosis. Moreover, asparagine gelatinous possessed immunomodulatory functions and showed little toxicity to the host. These results highlight the chemotherapeutic potential of asparagus polysaccharide and warrant a future focus on development as novel chemotherapeutic agent for liver cancer TACE therapy.

        Strand-specific RNA sequencing in pig testes identifies developmentally regulated genes and circular RNAs

        Testicular development and spermatogenesis are strictly regulated with the complex interactions between several cell types in testis. However, the limited availability of genomic and molecular information limits our understanding of this complex physiological process. In this study, we characterized the transcriptome between immature (30-day-old) and mature (180-day-old) pig testes using RNA-seq technology. 24,469 known coding gene transcripts corresponding to 20,566 genes were mapped in these two developmental stages, 3,328 genes were differentially expressed, and numerous novel transcripts and alternative splicing events were also identified. Ten differentially expressed genes were validated by measuring the relative expression using real-time quantitative polymerase chain reaction (PCR). 125 Gene Ontology (GO) terms were significantly enriched, and most of them involved in GO terms related to male reproduction, testicular development, and spermatogenesis. In addition, this study also represented the prediction of 10,000 circRNAs, as well as the validation of six pig circRNAs using Find_cric algorithm. Our study substantially expanded our knowledge about the transcriptomic profile of immature and mature pig testes, and provided a useful resource to study the mechanisms of pig testis development and spermatogenesis at the molecular level.

        Effects of different working fluids on the performance of a radial turbine in an organic Rankine cycle power system

        Organic Rankine cycle (ORC) power systems are widely used for low-temperature heat recovery. The radial turbine is the most important component in the ORC system and its performance is significantly dependent on the working fluid. Therefore, the effect of different working fluid on the performance of radial turbine in ORC system for the low-temperature heat source (120~180 °C) was investigated. First, the feasibilities of different working fluids for 150 kW radial turbine using R600a as designed working fluid were investigated. The thermal aerodynamic calculation of R600a at design condition was conducted with evaporation temperature 95 °C and condensation temperature 38 °C. The operation parameters of 5 non-designed working fluids (R600, R245fa, R245ca, R123 and R601a) were selected in the same temperature variation range as R600a. Then, the performance of a radial turbine for six working fluids at design condition was analyzed, and their three-dimensional flow field performances were also obtained by CFD numerical simulation. Finally, to broaden the operation range of the radial turbine and investigate the matching relationship between operation parameters and different working fluids, the off-design performances of radial turbine using six working fluids were analyzed and the effect of rotation speed, inlet pressure and inlet temperature on the performance of radial turbine was obtained. Results show that the performance of the radial turbine using the designed working fluid R600a is the best with efficiency of 82.7 % and output power of 150.5 kW. For the non-designed working fluids, the comprehensive performance of R600 was also judged to be good owing to the highest efficiency (85.1 %) and a relatively high power output (115.8 kW) among the fluids tested. For other working fluids, the output power of the radial turbine decreased obviously. There exists an optimum rotation speed for different working fluids to ensure that the radial turbine has the highest efficiency. The inlet pressure has a larger effect on the performance of the radial turbine than that of the inlet temperature. The results can provide basic data for the optimization design and operation of radial turbines in the ORC system.

        Three-dimensional Map Construction of Indoor Environment Based on RGB-D SLAM Scheme

        RGB-D SLAM (Simultaneous Localization and Mapping) refers to the technology of using deep camera as a visual sensor for SLAM. In view of the disadvantages of high cost and indefinite scale in the construction of maps for laser sensors and traditional single and binocular cameras, a method for creating three-dimensional map of indoor environment with deep environment data combined with RGB-D SLAM scheme is studied. The method uses a mobile robot system equipped with a consumer-grade RGB-D sensor (Kinect) to acquire depth data, and then creates indoor three-dimensional point cloud maps in real time through key technologies such as positioning point generation, closed-loop detection, and map construction. The actual field experiment results show that the average error of the point cloud map created by the algorithm is 0.0045m, which ensures the stability of the construction using deep data and can accurately create real-time three-dimensional maps of indoor unknown environment.

      • MiR-675 Promotes the Growth of Hepatocellular Carcinoma Cells Through Cdc25A Pathway

        Background: MicroRNAs (miRNAs) have fundamental roles in tumorigenesis. MiR-675 is upregulated in hepatocellular carcinoma(HCC) cells. However, the roles of miR-675 in hepatocellular carcinogenesis are still not fully elucidated. In this study, we focus on investigating the effect and mechanism of miR-675 in proliferation of HCC cells. Materials and Methods: The cell proliferation was measured by MTT assays after transfection with miR-675 inhibitor and miR-675 mimics in HCC cells. The expression level of miR-675 was detected by real-time quantitative reverse transcription polymerase chain reaction. Protein expression of Cdc25A was measured by western blotting analysis. Results: In MTT assays, overexpression of miR-675 promoted the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675mimics control group. Downexpression of miR-675 inhibited the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675inhibitor control group. In western blotting analysis, the expression level of Cdc25A was significantly increased (p<0.05) after treatment with miR-675 mimics. The expression level of Cdc25A was significantly decreased (p<0.05) after treatment with miR-675 inhibitor. Conclusions: Our results indicate that miR-675 promotes the proliferation in human hepatocellular carcinoma cells by associating with Cdc25A signaling pathway.

        Improving the Capture-range Problem in Phase-diversity Phase Retrieval for Laser-wavefront Measurement Using Geometrical-optics Initial Estimates

        To overcome the capture-range problem in phase-diversity phase retrieval (PDPR), a geometricaloptics initial-estimate method is proposed to avoid a local minimum and to improve the accuracy of laser-wavefront measurement. We calculate the low-order aberrations through the geometrical-optics model, which is based on the two spot images in the propagation path of the laser, and provide it as a starting guess for the PDPR algorithm. Simulations show that this improves the accuracy of wavefront recovery by 62.17% compared to other initial values, and the iteration time with our method is reduced by 28.96%. That is, this approach can solve the capture-range problem.

        The Relationship between HIF-2α and VEGF with Radiographic Severity in the Primary Osteoarthritic Knee

        Purpose: The aim of this study was to determine the relationship of hypoxia-inducible factor-2 (HIF-2α) and vascular endothelial growth factor (VEGF) with radiographic severity in primary osteoarthritis (OA) of the knee. Expression of these two factors in cartilagesamples from OA knee joints was examined at mRNA and protein levels. Materials and Methods: Knee joints were examined using plain radiographs, and OA severity was assessed using the Kellgren and Lawrence (KL) grading system. Specimens were collected from 29 patients (31 knees) who underwent total knee replacementbecause of severe medial OA of the knee (KL grades 3 and 4), 16 patients who underwent knee arthroscopy (KL grade 2), and 5 patients with traumatic knees (KL grade 0). HIF-2α and VEGF expression was quantified by real-time polymerase chain reactionand western blotting. Results: Cartilage degeneration correlated with the radiographic severity grade. OA severity, determined using the Mankin scale, correlated positively with the KL grade (r=0.8790, p<0.01), and HIF-2α and VEGF levels with the radiographic severity of knee OA (r=0.7001, p<0.05; r=0.6647, p<0.05). Conclusion: In OA cartilage, HIF-2α and VEGF mRNA and protein levels were significantly and positively correlated. The expressionof both factors correlated positively with the KL grade. HIF-2α and VEGF, therefore, may serve as biochemical markers as well as potential therapeutic targets in knee OA.
