Paenibacillus harenae sp. nov., isolated from desert sand in China

<P>A Gram-positive, endospore-forming, rod-shaped bacterium, designated strain B519T, was isolated from a desert sand sample of Gansu Province, China. Strain B519T was strictly aerobic and cells were motile by means of peritrichous flagella. The strain grew optimally at 32-35 degrees C and pH 6.5-7.0. Chemotaxonomic data supported the affiliation of the new isolate to the genus Paenibacillus, including menaquinone-7 (MK-7) as the major isoprenoid quinone, DNA G+C content of 49.9 mol%, cell-wall type A1gamma (meso-diaminopimelic acid as the diagnostic diamino acid) and anteiso-C(15 : 0), iso-C(15 : 0), C(16 : 0) and iso-C(16 : 0) as the major fatty acids. Comparative 16S rRNA gene sequence analysis showed that strain B519T was most closely related to Paenibacillus alkaliterrae KSL-134T (98.0 % similarity). DNA-DNA relatedness between strain B519T and P. alkaliterrae KSL-134T was about 12.3 %. On the basis of phenotypic characteristics and molecular properties, strain B519T is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus harenae sp. nov. is proposed. The type strain is B519T (=KCTC 3951T =DSM 16969T).</P>

<P><B><I>Introduction:</I></B> In recent years, there has been a great deal of interest in the development of vectors which are being developed based on the capacity of polymers to mediate appropriate interactions with the cellular environment, or to interface with specific cellular processes. Several such vectors have been synthesized, resulting in biomacromolecules with low cytotoxicity and higher gene delivery ability.</P><P><B><I>Areas covered:</I></B> This review briefly describes the recent success of poly(amido amine)s (PAAs) as non-viral vectors, and highlights their promising future in the development of nucleic acid-based therapy. It also provides an overview on the synthesis, characterization and application of PAAs as gene carriers, which will be useful for various biological motifs. This review helps the readers to better understand the emergence of non-viral vectors for gene therapy, especially PAAs, their properties, their advantages and disadvantages and the gene therapy based on them.</P><P><B><I>Expert opinion:</I></B> The future of gene-based therapy needs to identify approaches to develop new carriers, depending on the properties of the biological membranes they face, and their physicochemical properties, in order to successfully deliver the genes to the target sites. With the emergence of a variety of non-viral vectors, such as biodegradable polymers, it may not take long before non-viral vectors are observed that are not just safe and tissue-specific, but even more efficient than viral vectors.</P>

<P>Four new compounds, erythro-7'E-4-hydroxy-3,3'-dimethoxy-8,5'-oxyneoligna-7'-ene-7,9-dio1-9'-al (1), (7S,8S)4-hydroxy-3,1',3'-trimethoxy-4',7-epoxy-8,5'-neolign-9-ol (5), (75,85,7'E)-5-hydroxy-3,3'-dimethoxy-4',7-epoxy8,5'-neolign-7'-ene-9,9'-diol (6) and (75,85,7'E)-5-hydroxy-3,3',9'-trimethoxy-4'-7-epoxy-8,5'-neolign-7'-ene-9ol (7). Along with four known compounds (2-4, 8) were isolated from the EtOAc-soluble extract of Eleutherococcus senticosus. Their structures were elucidated on the basis of spectroscopic and physicochemical analyses. All the compounds were evaluated for in vitro inhibitory activity against PTP1B, VHR and PPl. Among them, compounds 1-4 and 6-8 were found to exhibit selective inhibitory activity on PTP1B with IC50 values ranging from 17.2 1.6 to 32.7 1.2 M.</P>

The effects of different levels of natural clay enterosorbent on the growth, nutrient availability, and genital organs of post-weaning female pigs fed with an addition of zearalenone (ZEA) were investigated in the study. A total of thirty-five post-weaning gilts ($L{\times}Y{\times}D$) with an average body weight of 12.36${\pm}$1.46 kg were used in the test. The gilts were raised individually in metabolism cages and fed a corn-soybean meal-whey basal diet with an addition of 0 or 1 mg/kg of ZEA for 24 d with four levels of natural clay enterosorbent added in the feed. The treatments were: i) control; ii) control+2.5 g/kg clay; iii) control+1 mg/kg ZEA; iv) control+1 mg/kg ZEA+1.25 g/kg clay; v) control+1 mg/kg ZEA+2.5 g/kg clay; vi) control+1 mg/kg ZEA+5.0 g/kg clay; vii) control+1 mg/kg ZEA +10 g/kg clay. Pigs fed diets contaminated with additional purified ZEA had significantly reduced apparent digestibility of crude protein (CP), gross energy (GE) and apparent metabolic rate of GE (ME/GE, p<0.05) without changes of net protein utilization (NPU, p>0.05). Final body weight, average daily gain (ADG), average daily feed intake (ADFI), vulva length, vulva width, vulva area, relative weights of genital organ and proliferative changes of the ovary tissues in gilts fed ZEA-contaminated diet were increased (p<0.05) compared to the gilts fed the control diet. Addition of natural clay enterosorbent in the ZEA-contaminated diet showed a positive protection effect on ZEA feeding, and the protection was increased linearly or quadratically as clay content increased. However, in pigs fed a diet with clay alone at 2.5 g/kg level there was no significant impact (p>0.05) on all the parameters as compared to the control. It is suggested that feeding ZEA at about 1.0 mg/kg for 24 days might result in a deleterious effect in pigs, and addition of 5 or 10 g clay enterosorbent per kg diet can effectively neutralize the detrimental effects of the ZEA feeding.

This study was conducted to determine the effect of dietary soy isoflavone (ISF, 0 and 20 mg/kg) on performance, meat quality and antioxidative property in male broilers. Six replicates of 45 birds (42 d old) were used for each treatment. The birds were fed soybean meal-free diets containing 3% oxidized fish oil (acid value, 6.76 mg potassium hydroxide/g; and peroxide value, 6.18 meq/kg) for 3 wk. The results showed that average daily gain, feed intake, feed conversion and carcass traits were not affected by soy ISF. Compared with the control group, breast muscle color redness value and water holding capacity were increased (p<0.05) by ISF supplementation. The activity of plasma catalase was increased by supplementing with 20 mg ISF/kg diet. In breast muscle, Broilers fed 20 mg ISF/kg had decreased production of malondialdehyde and lactic acid. The ISF supplementation elevated total antioxidative capacity and activities of total superoxide dismutase and glutathione peroxidase. The results indicated that dietary ISF could increase redness and water holding capacity of meat, and antioxidative property of meat in male broilers fed oxidized fish oil diet.

This study was conducted to investigate the protective effect of genistein on the antioxidative defence system and membrane fluidity in chick skeletal muscle cells after supplementation with 0, 20, 40, and $80{\mu}mol/L$ genistein in $50{\mu}mol/L$ $FeSO_4/H_2O_2$ treated cells for 24 h. Genistein supplementation recovered the decreased activity of total superoxide dismutase induced by $FeSO_4/H_2O_2$, significantly increased glutathione peroxidase activity (p<0.05) and decreased malondialdehyde production (p<0.05). The treatment of 80 mol/L genistein in $FeSO_4/H_2O_2$ treated cells decreased the secretion of creatine kinase (p<0.05). Fluorescence polarization values and microviscosities observed with $FeSO_4/H_2O_2$ treated cells were significantly higher than those observed with no $FeSO_4/H_2O_2$ treated cells. The addition of $80{\mu}mol/L$ genistein improved the increased fluorescence polarization value (p<0.05) caused by $FeSO_4/H_2O_2$ treatment. The microviscosity value was significantly decreased by adding genistein (p<0.05). In conclusion, genistein protected skeletal muscle cells from oxidative damage by improving antioxidative status and membrane fluidity.

This study employed the Difference in Difference (DID) model and the bibliometrics method with aim to novel corona virus disease (COVID-19) pandemic. This study conducted a comparative analysis based on the number of publications, co-authorship, and institutional collaborations on telemedicine literature research in China before and after the outbreak of COVID-19 (October 2017 to March 2022). The quantity, classification, and research focus of Chinese telemedicine literature was also compared based on data before the outbreak (October 2017 to December 2019) and after the outbreak (January 2020 to March 2022).The results show that the number of articles published by Chinese scholars on telemedicine research have soared after the outbreak of COVID-19, with significant emergence in the trend of co-authorship and institutional collaboration. The research hotspots focused on artificial intelligence, Wise Information Technology of Med (WITMED), and public hospitals, extending from the pre-pandemic topics including teleconsultation, internet+, and hierarchical medical system.
